Cartilage Oligomeric Matrix Protein (COMP, Thrombospondin-5) in human smooth muscle cells: in-vitro study on expression compared to the expression of Thrombospondin-1
Hemmstoffe der HMG-CoA-Reduktase (Statine) vermindern nicht nur die Cholesterinsynthese sondern können auch über eine verminderte Bildung anderer Isoprenoide eine Reihe von weiteren intrazellulären Prozessen und Signaltransduktionswegen beeinflussen. In dieser Arbeit wurde versucht, die Signaltransduktionswege näher zu charakterisieren, über die Statine die Proliferation und das Muster der extrazellulären Matrixexpression arterieller glatter Muskelzellen beeinflussen. Neben dem HMG-CoA-Reduktase–Inhibitor Lovastatin wurden die Isoprenoide Mevalonat, Geranylgeranyl-Pyrophosphat (GGPP) und Farnesyl-Pyrophosphat (FPP) sowie Hemmstoffe der Geranylgeranyl-Transferase (GGTI-286), der Farnesyltransferase (FTI 277) und der Rho-Kinase (Y-27632) eingesetzt.
Proliferations-Assays zeigten, dass der Einfluss der Statine auf die Proliferation glatter Muskelzellen womöglich über die Geranylierung des kleinen G-Proteins Rho und sukzessive über die Aktivierung der ihm assoziierten Rho-Kinase verläuft. In Bezug auf die Matrixexpression beschäftigte sich die Arbeit mit der Expression der Thrombospondine (TSP), einer Gruppe von insgesamt 5 Glykoproteinen. In PCR-, Northernblot- und Westernblot-Analysen zeigte sich ein Effekt sowohl von Lovastatin als auch von GGTI-286 und Y-27632 auf die RNA- und Proteinexpression von TSP-1 und TSP-5/COMP. Während die Expression von TSP-1 durch Statine, GGTI-286 und Y-27632 gehemmt wurde (durch letzteres allerdings nur schwach), wurde die Expression von TSP-5/COMP gesteigert. Diese Arbeit konnte mit der Wirkung auf TSP-5/COMP einen weiteren pleiotropen Effekt der Statine aufzeigen und mit dem Rho-Kinase Signaltransduktionsweg die weitere intrazelluläre Regulation nachweisen. Zudem konnte diese Arbeit zeigen, dass Rapamycin - ein potenter Inhibitor der Restenose nach Stentimplantation - Einfluss auf die Expression von TSP-5 nimmt. Dabei wird die Proteinkonzentration in glatten Muskelzellen vermindert und die RNA-Konzentration (vermutlich kompensatorisch) gesteigert. Bei den weiteren Thrombospondinen TSP-2, -3, und -4 ließ sich keine eindeutige Regulation der Expression nachweisen. Allerdings konnte TSP-3 in dieser Studie erstmals in humanen glatten Muskelzellen nachgewiesen werden.HMG-CoA-Reductase-Inhibitors (Statins) not only reduce cholersterol biosynthesis but also influence different intracellular mechanisms and signalling pathways by lowering the concentration of other isoprenoids. This study tried to characterize the signalling pathways that influence proliferation of vascular smooth muscle cells and the composition of the extracellular matrix by statins. Apart from the HMG-CoA-Reductase-Inhibitor Lovastatin other isoprenoids like Mevalonat, Geranylgeranyl-Pyrophosphat (GGPP) and Farnesyl-Pyrophosphat (FPP) as well as inhibitors of the Geranylgeranyl-Transferase (GGTI-286), Farnesyltransferase (FTI 277) and the Rho-Kinase (Y-27632) were studied.
Proliferations-Assays showed that the effect of statins on SMC proliferattion is possibly regulated by the geranylation of the small G-protein Rho and subsequent by the activation of its associated Rho-Kinase. Regarding the matrix expression this study directed its attention to the expression of the so-called Thrombospondins, a family of 5 glycoproteins. RNA and protein expression of TSP-1 and TSP-5/COMP were effected by Lovastatin, GGTI-286 and Y-27632 shown in PCR-, Northern-blot and Western-blot analysis. While TSP-1 expression was inhibited by statins, GGTI-286 and Y-27632 (the last mentioned only weakly) the expression of TSP-5/COMP could be shown to be increased.
Therefore this study found out another pleiotropic effect of statins with their effect on TSP-5/COMP and furthermore proved the intracellular regulation by the Rho-Kinase signalling pathway. In Addition this study showed that Rapamycin - a potent inhibitor of restenosis after stent implantation - influences the expression of TSP-5. The protein level is lowered in SMC whereas RNA-concentration is (compensational) increased. A clearly regulation of the other thrombospondins TSP-2, TSP-3 and TSP-4 could not be confirmed, however TSP-3 could be detected in SMC for the first time
Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more

We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
