Síntese de ésteres catalisada por lipase de pseudomonas sp. imobilizada em filmes de caseinato de sódio/glicerol

Curso de Química.Nos últimos anos as lipases vêm sendo empregadas com sucesso em processos biocatalíticos. O uso destas como catalisadores em síntese orgânica pode ser dificultado pela possível desnaturação e perda da atividade catalítica em solventes orgânicos. Para minimizar estas condições adversas usam-se técnicas de imobilização em suportes poliméricos. Os filmes de caseinato de sódio (CS) podem ser utilizados como proteção às enzimas devido a sua boa estabilidade em vários solventes orgânicos. Neste trabalho utilizaram-se filmes de CS para imobilizar a lipase de Pseudomonas sp. (PSL, Amano) e lipase F (Amano). Estes sistemas foram usados como biocatalisadores em reações de esterificação em meio orgânico (hexano) . Os parâmetros avaliados foram a influência do número de carbonos da cadeia alquílica dos reagentes (álcoois e ácidos), o efeito da concentração da enzima e do tempo de reação no grau de conversão em ésteres, a reutilização do sistema CS/PSL e a imobilização simultânea das lipases de Pseudomonas sp e F no mesmo suporte. Utilizando o sistema PSL/CS, a maior conversão foi obtida quando o 1-propanol foi utilizado na esterificação do ácido oleico (97%), e foi observado conversões menores que 50% para álcoois com mais de cinco carbonos. Na esterificação dos ácidos alifáticos com n-propanol as conversões foram quantitativas a partir de três carbonos (~100%). Foi verificado a influência da saturação na cadeia alquílica de ácidos carboxílicos .As maiores conversões, em éster, foram obtidas com os ácidos oleico e 4-pentenóico ( ~100%). Não houve a formação do produtos com os ácidos trans-2-octenóico e sórbico, evidenciando a importância dos efeitos estéreos. O efeito do tempo na esterificação dos ácidos oleico, laurico e linoleico com n-propanol foi estudado, e os resultados mostraram que após 12h o éster derivado do ácido linoleico foi obtido com conversão de 93%. A influência da quantidade de PSL imobilizada nos filmes de caseitnato de sódio, foi testada na obtenção dos ésteres derivados dos ácidos oleico e laúrico

Psl-Dependent Cooperation Contributes to Drug Resistance of Pseudomonas aeruginosa in Dual-Species Biofilms with Acinetobacter baumannii
Co-infection of Pseudomonas
aeruginosa (Pa) and Acinetobacter
baumannii (Ab) is frequently observed in
intensive care unit (ICU)
patients but difficult to eliminate. Current clinical practice based
on microbial population characterization and single-species-based
antibiotic resistance profiling has ignored the potential interspecies
interactions, which might lead to novel drug-resistance phenotypes.
Here, we investigated the impacts of interspecies interactions on
antibiotic therapies by establishing a Pa and Ab dual-species biofilm model. Our data showed that antibiotic
exposure would reshape the community compositions of dual-species
biofilms, and those of the extracellular polymeric substance (EPS)
matrix of Pa, Psl exopolysaccharide in particular,
promoted its interactions with Ab against imipenem
stress. We further found other EPS structural fiber-eDNA contributed
to the Psl-dependent dual-species biofilm stability under antibiotic
treatment. Thus, targeting the EPS structural fibers such as Psl and
extracellular DNA (eDNA) is a potent strategy for controlling polymicrobial
biofilm related infections

This paper presents the implementation strategy and current status of a new Portable LISP System, PSL, based upon STANDARD LISP. PSL is written entirely in itself and is compiled with an efficient LISP compiler with machine-oriented optirnizations. A full PSL is currently running on a DECSystem-20 and a VAX, and a preliminary version runs on a Motorola MC68000. Versions for a CRAY-1 and an IBM-370 are planned.</p
