    Proteins are large molecules that play essential roles in living cells including structure, regulation and functions. They provide key information for understanding biological mechanisms and diseases. The development of new tools for rapid protein sequencing at the single molecule scale, would contribute to the diagnosis of numerous diseases and to the development of personalized medical assessment.Nanopore-based single-molecule electrical detection technology is a low-cost, real-time, and label-free analysis method, that stands out as a promising tool for the development of this application. The principle of this technique consists of using a nanometric pore (nanopore) created in a membrane separating two compartments filled with electrolyte solution. Due to the applied voltage, the electrolyte ions flow through the nanopore. The entry of individual molecules in the nanopore causes a drop in ionic current whose characteristics provide information on the chemical nature of the molecule being analyzed. Based on this principle, biomolecule sequencing can be achieved by sequentially reading the information of an unfolded protein through a nanopore.The significant advances in nanopore DNA sequencing have enhanced the potential of this technology and expanded the development of nanopore protein sequencing. This latter is, nonetheless, more challenging compared to nucleic acids because of many constraints mainly related to the nature of proteins which have a complex structure. It can be composed of 20 amino acids which may also involve chemical modifications of some of them. To our knowledge, when my thesis work has been initiated, no nanopore, whether biological unmodified, solid, or artificial, has so far made it possible to obtain sufficient amino acid resolution for reading the sequence of a protein.The foreseen scope of my thesis work is to develop a protein sequencing strategy based on the use of a biological nanopore called aerolysin, a toxin produced by the bacteria Aeromonas hydrophila which forms nanometric pores in biological membranes. Aerolysin has recently demonstrated a high resolving power, under optimal physicochemical conditions, for the analysis of different types of molecules such as neutral polymers, and oligonucleotides. Aerolysin thus represents an ideal candidate for attempting to achieve a high separation resolution permitting the detection of an amino acid.This manuscript explores the results that I obtained during my thesis work. My first major contribution is to demonstrate, for the first time, that aerolysin allows the discrimination of short peptides (from 5 to 10 amino acids) analyzed in a mixture or individually, with a separation resolution equivalent to that of an individual amino acid. My second major contribution is to demonstrate for the first time a mapping of 20 natural amino acids per nanopore, based on their electrical signatures measured through aerolysin. We have also shown that it is possible to identify chemical modifications carried by certain amino acids.Les protéines sont des macromolécules indispensables au fonctionnement des cellules vivantes, pouvant fournir des informations clés pour comprendre les mécanismes biologiques et les maladies. Ce nouveau challenge est néanmoins bien plus difficile que pour les acides nucléiques en raison de nombreuses contraintes principalement liées à la nature des protéines qui présentent une structure complexe, qui peuvent être composées de 20 acides aminés, et qui présentent des modifications chimiques de certains de leurs acides aminés. A notre connaissance, au commencement de mes travaux de thèse, aucun nanopore, qu’il soit biologique non modifié, solide, ou artificiel, n’avait permis d’obtenir une résolution d’un acide aminé suffisante à la lecture de la séquence d’une protéine.L’ambition de mon projet de thèse est de développer une stratégie de séquençage des protéines basée sur l’utilisation d’un nanopore biologique, l’aérolysine, une toxine produite par la bactérie Aeromonas hydrophila, capable de former des pores nanométriques dans les membranes biologiques. L’aérolysine, a récemment démontré un haut pouvoir de résolution, dans des conditions physico-chimiques optimales, pour l’analyse de différents types de molécules, comme des polymères neutres et des oligonucléotides. L’aérolysine se présente ainsi comme un candidat de choix pour tenter d’atteindre une résolution de détection d’un acide aminé.Ce manuscrit a pour objectif de présenter les résultats que j’ai obtenus durant ma thèse.     Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
