    Tijdens dit onderzoek wordt een vergelijking gemaakt tussen gemeentelijke verkoopmethodes die worden gebruik voor het verkopen van voormalige schoolgebouwen als (C)PO-project. Deze schoolgebouwen worden verkocht aan particulieren die deze schoolgebouwen vervolgens transformeren naar woningen. Er wordt een vergelijking gemaakt van zes verkoopmethodes die zijn gebruikt in de gemeentes Rotterdam, Amsterdam en Den Haag. Tijdens deze vergelijking wordt gekeken naar vier variabelen waaruit de verkoopmethodes zijn samengesteld: de toewijzingssystematiek, verkaveling, investeringen en de randvoorwaarden. Hierbij wordt onderzocht wat de redeneringen zijn geweest van de gemeentes om voor deze methode te kiezen.     The performance of forward error correction codes and the respective interleaved codes for a direct-sequence code division multiple access system in a bursty land-mobile satellite channel have been evaluated in terms of throughput and delay. The FEC scheme used the Reed-Solomon and BCH codes. Interleaved codes were compared with non-interleaved versions. Results are presented for heavy shadowing conditions in a fast as well as a slow fading environment.     Numerical modelling of the growth of offshore sand waves: A Delft3D study

    Offshore sand waves of several meters high and hundreds of meters long are formed in shallow seas due to the tidal currents. These sand waves can, amongst others, influence the burial depth of cables of offshore wind farms and obstruct navigation channels. For the Far Large Offshore Wind programme (FLOW), one of the goals of Deltares is to increase the knowledge on sand wave growth and behaviour. Since only a limited amount of bathymetrical data is available in these offshore areas, numerical models can give some insights to predict sand wave development. So far, all numerical models used to estimate the growth of offshore sand waves predict unrealistic values for either the sand wave height or the sand wave length. These values are highly overestimated. Most models initially predict a sand wave length that is realistic under the modelled conditions. This is the length of the fastest growing mode (LFGM). However, when extending the model simulation time, the sand waves grow up to the length of the entire computational domain. When limiting the length of the domain to one sand wave length, the sand waves become extremely high. No model has yet been able to correctly predict both the wave length and the wave height. The main objective of this study was to understand the processes behind sand wave growth and modelling the sand waves up to an equilibrium state. The effects of two different aspects on the growth of offshore sand waves in a sand wave field were studied. First, the impact of the bed slope effect was assessed, since in previous studies the influences of this parameter were not studied in much detail. The bed slope effect is the (numerical) phenomenon where sediment grains are transported downhill more easily than uphill. This parameter is very important under the applied conditions and has a strong influence on the model predictions. Secondly, the influence of different initial bathymetries on the growth of the sand waves was determined. Both initially random and large sinusoidal beds were used. Long-term model simulations (morphological time > 200 years) were run to assess the impacts of the studied aspects on a potential equilibrium. Equilibrium is found for growing sand waves, when the bed with sinusoidal perturbations equal in length to the LFGM is applied. With these sinusoidal perturbations, the bed slope effect only influences the growth rate and the shape. The equilibrium height found is similar to that predicted with other numerical models where the domain has the length of one sand wave and is, thus, overestimated. In both cases, the equilibriums are forced. The sand waves cannot grow further in length and have reached a maximum height for the corresponding wave length. Model simulations with other initial beds led to even larger and longer sand waves. Though, at some point during the simulation, a mean wave length of approximately the length of the fastest growing mode is dominant in the sand wave field. Eventually, after the modelled 750 years, no equilibrium is found with these initial beds as the increase in wave length or wave height is on-going. Existing studies of the Indonesian Throughflow have focused on the Makassar Strait and the exit straits, where the upper thermocline currents carry North Pacific waters to the Indian Ocean. Here we show, using mooring observations, that a previous unknown intermediate western boundary current (with the core at ~1000 m depth) exists in the Maluku Sea, which transports intermediate waters (primarily the Antarctic Intermediate Water) from the Pacific into the Seram-Banda Seas through the Lifamatola Passage above the bottom overflow. Our results suggest the importance of the western boundary current in global ocean intermediate circulation and overturn.     Transcriptomic Analysis of Two Thioalkalivibrio Species Under Arsenite Stress Revealed a Potential Candidate Gene for an Alternative Arsenite Oxidation Pathway

    The genus Thioalkalivibrio includes haloalkaliphilic chemolithoautotrophic sulfur-oxidizing bacteria isolated from various soda lakes worldwide. Some of these lakes possess in addition to their extreme haloalkaline environment also other harsh conditions, to which Thioalkalivibrio needs to adapt. An example is arsenic in soda lakes in eastern California, which is found there in concentrations up to 3000 mu M. Arsenic is a widespread element that can be an environmental issue, as it is highly toxic to most organisms. However, resistance mechanisms in the form of detoxification are widespread and some prokaryotes can even use arsenic as an energy source. We first screened the genomes of 76 Thioalkalivibrio strains for the presence of known arsenic oxidoreductases and found 15 putative ArxA (arsenite oxidase) and two putative ArrA (arsenate reductase). Subsequently, we studied the resistance to arsenite in detail in Thioalkalivibrio jannaschii ALM2(T), and Thioalkalivibrio thiocyanoxidans ARh2(T) by comparative genomics and by growing them at different arsenite concentrations followed by arsenic species and transcriptomic analysis. Tv. jannaschii ALM2(T), which has been isolated from Mono Lake, an arsenic-rich soda lake, could resist up to 5 mM arsenite, whereas Tv. thiocyanoxidans ARh2(T), which was isolated from a Kenyan soda lake, could only grow up to 0.1 mM arsenite. Interestingly, both species oxidized arsenite to arsenate under aerobic conditions, although Tv. thiocyanoxidans ARh2(T) does not contain any known arsenite oxidases, and in Tv. jannaschii ALM2(T), only arxB2 was clearly upregulated. However, we found the expression of a SoeABC-like gene, which we assume might have been involved in arsenite oxidation. Other arsenite stress responses for both strains were the upregulation of the vitamin B-12 synthesis pathway, which can be linked to antioxidant activity, and the up- and downregulation of different DsrE/F-like genes whose roles are still unclear. Moreover, Tv. jannaschii ALM2(T) induced the ars gene operon and the Pst system, and Tv. thiocanoxidans ARh2(T) upregulated the sox and apr genes as well as different heat shock proteins. Our findings for Thioalkalivibrio confirm previously observed adaptations to arsenic, but also provide new insights into the arsenic stress response and the connection between the arsenic and the sulfur cycle

    Desulfonatronobacter acetoxydans sp. nov.,: A first acetate-oxidizing, extremely salt-tolerant alkaliphilic SRB from a hypersaline soda lake

    Recent intensive microbiological investigation of sulfidogenesis in soda lakes did not result in isolation of any pure cultures of sulfate-reducing bacteria (SRB) able to directly oxidize acetate. The sulfate-dependent acetate oxidation at haloalkaline conditions has, so far, been only shown in two syntrophic associations of novel Syntrophobacteraceae members and haloalkaliphilic hydrogenotrophic SRB. In the course of investigation of one of them, obtained from a hypersaline soda lake in South-Western Siberia, a minor component was observed showing a close relation to Desulfonatronobacter acidivorans—a “complete oxidizing” SRB from soda lakes. This organism became dominant in a secondary enrichment with propionate as e-donor and sulfate as e-acceptor. A pure culture, strain APT3, was identified as a novel member of the family Desulfobacteraceae. It is an extremely salt-tolerant alkaliphile, growing with butyrate at salinity up to 4 M total Na+ with a pH optimum at 9.5. It can grow with sulfate as e-acceptor with C3–C9 VFA and also with some alcohols. The most interesting property of strain APT3 is its ability to grow with acetate as e-donor, although not with sulfate, but with sulfite or thiosulfate as e-acceptors.
