    Modified amino acids and peptides as potential inhibitors of bacterial cell wall biosynthesis

    In this work we have synthesized a number of modified amino acids and peptides which were supposed to be analogues of the substrate, the postulated intermediate and the product of the reaction catalysed by D-alanine:D-alanine ligase, a key enzyme in the D-alanine branch of bacterial cell wall peptidoglycan assembly. Most of these were crystalline solids, and were prepared as both L and D isomers. When these compounds were subjected to their biological activity it was found that they were unlikely to be inhibitors of the bacterial cell wall biosynthesis. The reduced dipeptide L-alanyl-(CH2-NH)-L-alanine was able to inhibit the growth of S.aureus 6571 in solid as well as liquid media, and its inhibitory effect was stopped in the presence of the dipeptide L-alanyl-L-alanine and the tripeptide L-alanyl-L-alanyl-L-alanine but not by L-alanine. The reduced dipeptide was suggested to act intracellularly as a whole molecule. The N-protected bromo and chloromethyl ketones of alanine were also found to inhibit the growth of S.aureus 6571 and E.coli W3110 in the solid and liquid media, and these were suggested to inhibit a proteinase inside the bacterium. The enzyme D-alanine: D-alanine ligase was purified from E.faecalis (NCIB) 6459) to a specific activity of 8.194 &mu;mol min^-1mg^-1 of protein and its K_m and V_max values were determined. The ligase was also purified from E.coli M72 (pDOC87) to a specific activity of 18.9 &mu;mol min-1mg-1 of protein. E.coli ligase was found to have a Mr of 31,000 ± 1000 by SDS polyacrylamide gel electrophoresis. The N-terminal sequence of ten residues of the ligase was found to be consistent with that predicted from the complete gene sequence reported by Robinson et al. (1986). Finally, the potency of the synthetic inhibitors on E.coli D-alanine: D-alanine ligase was studied in vitro. The intermediate analogues D-(2-amino-1-hydroxypropyl) phosphonate and D-(2-aminopropyl) phosphate were more inhibitory than the substrate analogues D-alanine chloromethyl ketone, D-alanine bromomethyl ketone and 4-amino-3-hydroxy pentanoic acid, and the product analogue D-alanyl-(CH_2-NH)-D-alanine. The kinetics of the ligase inhibition by D-(2-amino-1-hydroxylpropyl) phosphonate and D-(2-aminopropyl) phosphate were studied, and the kinetic constants are summarized. The inhibition of E.coli ligase by the aminoalkyl phosphinate of Merck was also investigated. This compound was found to be a parabolic competitive inhibitor and the kinetic constants are given. ATP was shown not to be required for the ligase inhibition by this compound, and no time-dependent process was obtained. It is suggested that the aminoalkyl phosphinate could be a classical inhibitor of E.coli ligase.</p

    New technique in the literature for recto-vaginal fistula: Gebrel-Mostafa technique

    Aim. In this study, we present a new method for treating cases of rectovaginal fistula that will be added to the medical literature. In this study, we examine two patients with low vaginal fistula from their initial clinic visit until the end of their follow-up period. Methods and materials. Two patients who had undergone a normal vaginal delivery one year prior presented to our clinic with complaints of offensive vaginal discharge and distressing symptoms, including stool leakage and vaginal flatulence. They consented to experience this new technique for the treatment of their low vaginal fistula. Results. Gebrel-Mostafa technique has numerous beneficial effects, including the elimination of the need for a second operation to remove the plastic Avis, which functions as a seton. In addition, it is characterized by a short duration of operation, the inability to form an abscess or hematoma, and a rapid return to normal life. Avis plastic acts as an auto-tightening and auto-cutting material, and the patient or doctor can tighten it. Finally, it is less expensive, and there is no requirement to use the labial Martius flap. Conclusion. This (Gebrel-Mostafa) technique yields positive outcomes with no significant morbidity and is feasible to perform. The absence of incontinence, abscess formation, and sepsis in the wound may be attributable to the wide range of antibiotics administered to the patients

    Forgotten Legacy: An Endeavor to Highlight Heritage Food of Egyptian Bedouins

    Despite the issue of food heritage having an important debate around the globe, there is still inconclusive literature addressing Egyptian heritage food in general, and particularly the gastronomic heritage of Egyptian Bedouins. The main argument of the current study is firstly to identify a set of genuine heritage foods of Egyptian Bedouins by using explicit criteria. Then, transfer the data to develop a documented recipe book for these meals, which is intended to be an initial phase of promoting them as a unique component of the Egyptian cultural legacy. The target population of the study is the Egyptian Bedouin society, both in the western and eastern deserts. Through a qualitative approach, 15 semi-structured interviews with elderly local people were undertaken before conducting a focus group with six Bedouin experts. The findings section reported eight heritage foods of Egyptian Bedouins and produced a recipe book for them

    For a territory to be sustainable, economic and social benefits need to be exploited to improve the living standard of local people. Food as a part of human culture has supposed a significant role in tourist decision-making and satisfaction, tourism services and destination promotion strategies, accordingly, it can be a useful factor of destination development. The main argument of current research is to explore and document a group of authentic heritage foods of Egyptian Bedouins as an attempt to preserve them as well as suggest a potential utilisation of these foods in the light of rural tourism. An initial online survey, semi-structured interviews, and focus group discussions with Bedouins from Sinai and Matruh areas have been undertaken to accomplish the study aim. At the end of the study, a full description of seven heritage foods of the mentioned areas with their original names, pictures, ingredients, and preparation methods have presented, additionally, a potential utilisation has been suggested
