Spheroid morphology of lung cancer cell lines correlates with oncological profiles
Nagasaki University (長崎大学)博士(医学)We assessed the correlation between Multicellular tumor spheroids (MCTS) morphology and the oncological profile of lung cancer cells. MCTS were generated in five lung cancer cell lines and classified into Type–A MCTS, which showed strong aggregation, and Type–B MCTS, which showed weak aggregation. Drug resistance was higher in Type–A MCTS, and invasive ability was higher in Type–B MCTS. The oncologic profile of lung cancer cell lines correlated with MCTS morphology. 日本人女子大学生における摂食障害に関するメンタルヘルスリテラシー
Mental health literacy (MHL) refers to one’s knowledge and beliefs about mental disorders. MHL is necessary for the prevention and early detection of eating disorders, but little research exists on the MHL of Japanese people. This paper aims to reveal the degree of MHL about eating disorders in the Japanese individuals most susceptible to them (namely, female university students). Female Japanese university students were surveyed with a self-report questionnaire, using the Japanese versions of anorexia nervosa and bulimia nervosa vignettes. Most participants responded that the main problem described in each vignette were “eating disorders” and tended to suggest medical care for the treatment. However, many still believed these issues to be simple problems of eating behavior. Overall, the participants demonstrated inadequate MHL about eating disorders. Effect of the Sway Bed on Autonomic Response, Emotional Responses and Muscle Hardness in Children with Severe Motor and Intellectual Disabilities: A Pilot Study
Nagasaki University (長崎大学)博士(医学)This cross-sectional study aimed to examine the effects of being swayed in a sway bed on children with severe motor and intellectual difficulties by examining potential differences in their autonomic and emotional responses, as well as their muscle hardness, and by comparing them with “a control condition without any stimulation”. Children’s heart rate variability, rectus femoris hardness, and passive hip abduction range of motion (ROM) were measured in two experimental conditions, differentiated by the presence of a 5-min sway stimulus. In each condition, the children’s faces were video-recorded and retrospectively rated subjectively by their homeroom teacher concerning the visible expression of eight emotions. Significant intervention-related effects were observed on the heart rate variability and the “Relax” item of the emotional response indicators but not on muscle hardness or hip ROM. 日本における育児ストレスと子どもの行動特性との関連
Background: The evaluation of a child tends to differ between the mother and father regardless of whether the child has a disability or not, although parents have key information about the behavioral characteristics of the child. The number of reports in Japan, however, is limited. We, therefore, investigated the relationship between the parenting stress experienced by parents of non-clinical preschool children and the children's behavioral characteristics.
Method: The subjects were 83 pairs of mothers and fathers with non-clinical children in kindergarten and nursery school (average age, 59.1 ± 13.0 months; 36 boys, 47 girls). The study was conducted using the Parenting Stress Index–Short Form (PSI-SF) and the Strengths and Difficulties Questionnaire (SDQ). Results:
There was no difference in the PSI-SF scores between mothers and fathers, but hyperactivity/inattention, peer relationship problems, and the total score of the SDQ were significantly higher in fathers. On multiple regression analysis, parenting stress experienced by fathers was significantly related to hyperactivity/inattention, while parenting stress experienced by mothers was significantly related to peer relationship problems and emotional symptoms. Sensory processing associated with subcategories of restricted and repetitive behaviors in Japanese children and adolescents with autism spectrum disorder
Nagasaki University (長崎大学)博士(医学)Background: Restricted and repetitive behavior (RRB) is a core symptom of autism spectrum disorder (ASD). The structure of RRB subcategories and their relationship with atypical sensory processing in Japan are not well understood. This study examined subcategories of the RRB in Japanese children with ASD and explored their relationship with sensory processing. Methods: A total of 103 children and adolescents with ASD participated in this study, with more than 70% having a co-occurring intellectual disability. First, exploratory factor analysis of the RRB items of the Social Responsiveness Scale second edition (SRS-2) was conducted to identify RRB subcategories. Second, Spearman correlation and multiple regression analysis were run to examine relationships between the RRB subcategories of SRS-2 and subsections of the Short Sensory Profile. Results: Exploratory factor analysis indicated a two factors solution; repetitive sensory and motor behavior and insistence on sameness. Multiple regression analysis suggested that Movement Sensitivity and Auditory Filtering were associated with insistence on sameness. Furthermore, Underresponsive/Seeks Sensation, Visual/Auditory Sensitivity, and diagnosis of intellectual disabilities were associated with repetitive sensory and motor behavior. Conclusions: Findings indicate that RRB subcategories are differently related to sensory processing patterns in children with ASD. Adipose-Derived Mesenchymal Stem Cells Attenuate Immune Reactions Against Pig Decellularized Bronchi Engrafted into Rat Tracheal Defects
Nagasaki University (長崎大学)博士(医学)Decellularized scaffolds are promising biomaterials for tissue and organ reconstruction; however, strategies to effectively suppress the host immune responses toward these implants, particularly those without chemical crosslinking, remain warranted. Administration of mesenchymal stem cells is effective against immune-mediated inflammatory disorders. Herein, we investigated the effect of isogeneic abdominal adipose-derived mesenchymal stem/stromal cells (ADMSCs) on xenogeneic biomaterial-induced immunoreactions. Peripheral bronchi from pigs, decellularized using a detergent enzymatic method, were engrafted onto tracheal defects of Brown Norway (BN) rats. BN rats were implanted with native pig bronchi (Xenograft group), decellularized pig bronchi (Decellularized Xenograft), or Decellularized Xenograft and ADMSCs (Decellularized Xenograft +ADMSC group). In the latter group, ADMSCs were injected intravenously immediately post implantation. Harvested graft implants were assessed histologically and immunohistochemically. We found that acute rejections were milder in the Decellularized Xenograft and Decellularized Xenograft+ADMSC groups than in the Xenograft group. Mild inflammatory cell infiltration and reduced collagen deposition were observed in the Decellularized Xenograft+ADMSC group. Additionally, ADMSC administration decreased CD8+ lymphocyte counts but increased CD163+ cell counts. In the Decellularized Xenograft+ADMSC group, serum levels of vascular endothelial growth factor and IL-10 were elevated and tissue deposition of IgM and IgG was low. Anti-tumor Effects of Erlotinib via Thymidylate Synthase Down-regulation in Pancreatic Cancer Cells
Nagasaki University (長崎大学)博士(医学)Background/Aim: In pancreatic cancer, gemcitabine and EGFR tyrosine kinase inhibitors (EGFR-TKIs) are common chemotherapy options. Reports have shown that EGFR-TKIs suppress the expression of thymidine synthase (TS), an important enzyme for DNA biosynthesis, and increase sensitivity to gemcitabine in lung cancer. However, no such reports have been made in pancreatic cancer. Materials and Methods: Human pancreatic cancer cell lines MiaPaCa2, Panc1, and BxPc3 were used. TS mRNA and protein expression levels in the cells were analyzed after erlotinib treatment. In addition, the anti-tumor effect of TS knockdown was verified using TS siRNA, along with its synergistic effect when combined with gemcitabine. Results: TS expression was high in MiaPaCa2 and Panc1 cells and low in BxPc3 cells. After erlotinib treatment, TS mRNA and protein levels decreased markedly in MiaPaCa2 cells dose-dependently, but not in Panc1 cells. TS siRNA caused specific down-regulation of TS in MiaPaCa2 and Panc1 cells. TS down-regulation resulted in an anti-tumor effect in these cells (MiaPaCa2 42%; Panc1 38%; p<0.05), showing a synergistic effect when combined with gemcitabine. Relationship between motor coordination, cognitive abilities, and academic achievement in Japanese children with neurodevelopmental disorders
Nagasaki University (長崎大学)博士(医学)Background/Objective: Motor coordination impairment is common in children with neurodevelopmental disorders such as autism spectrum disorder (ASD) and attention deficit/hyperactivity disorder (AD/HD). The purpose of this study was to investigate the relationship between motor coordination, cognitive ability, and academic achievement in Japanese children with neurodevelopmental disorders. Methods: Thirty-four school-age (6?12 years old) children with neurodevelopmental disorders and 34 age-matched typically developing (TD) children were recruited in this study. Correlations between the scores of the Movement Assessment Battery for Children-2 (M-ABC2) and the Kaufman Assessment Battery for Children ? Second Edition (K-ABCII) that assesses cognitive abilities, and academic achievement were analyzed. Results: The children with neurodevelopmental disorders obtained a lower total score and all component scores on M-ABC2 compared to the TD children. In children with neurodevelopmental disorders, M-ABC2 Manual Dexterity score was significantly correlated with K-ABCII Simultaneous Processing (r = .345, p = .046), Knowledge (r = .422, p = .013), Reading (r = .342, p = .048), Writing (r = .414, p = .017), and Arithmetic (r = .443, p = .009) scores. In addition, M-ABC2 Balance score was significantly correlated with K-ABCII Learning (r = .341, p = .048), Writing (r = .493, p = .004), and Arithmetic (r = .386, p = .024) scores. Examining the Effectiveness of Ayres Sensory Integration® Intervention for Children With Developmental Coordination Disorder in Improving Motor Coordination and Daily Activity Function: A Randomized Controlled Trial
Nagasaki University (長崎大学)博士(医学)Background: Ayres Sensory Integration® (ASI) intervention focuses on developing sensory processing abilities to improve motor coordination, executive functions, participation, and satisfaction in everyday activities. No well-designed research studies have addressed clearly the effectiveness of ASI intervention for children with developmental coordination disorder (DCD) even though ASI intervention was effective for children with autism spectrum disorders. Methods and procedures: Seventeen children with DCD (aged 4-8.5 years old) were randomly assigned to either an intervention or a control group. ASI intervention was provided to the intervention group twice a week for 10 weeks. In addition to participants’ goals in daily activities, sensory integration abilities and motor coordination were assessed before and after the intervention. Outcomes and results: The split-plot factorial design demonstrated significant time × group interaction in the total score (F (1, 15) = 7.651, p = 0.014, partial η2= 0.338) and balance score (F (1, 15) = 11.163, p = 0.004, partial η2 = 0.427) of the Movement Assessment Battery for Children-Second Edition (MABC-2), with significant differences in simple main effects before and after intervention for the intervention group. The post-intervention Goal Attainment Scale (GAS) score showed a significant difference in the time × group interaction (F (1, 15) = 15.662, p = 0.001, partial η2 = 0.511) and a simple main effect in the intervention group. Factors influencing postpartum depression among Japanese parents: A prospective longitudinal study
Nagasaki University (長崎大学)博士(医学)Aim: Postpartum depression (PPD) may have negative effects on the parents and lead to impaired cognitive, socioemotional, and behavioral development in their children. The purpose of this study was to examine factors associated with PPD in parents during the first year after delivery.
Methods: This study used a self-administered questionnaire. Questionnaires were mailed at 5 days, 3 months, 6 months, and 1 year after delivery, respectively. The particpants were 107 pairs of mothers and fathers. PPD was assessed using the Edinburgh Postnatal Depression Scale (EPDS). Data on sense of coherence (SOC), Quality Marriage Index, Social Support Scale, Mother-to-Infant Bonding Scale, and sociodemographic variables were collected. Multiple regression analysis was performed to examine the strength of the association between several variables and the EPDS at each survey period for fathers and mothers, respectively.
Results: The prevalence of PPD in the first-year postpartum ranged from 12.1%–23.4% to 7.5%–8.4% for fathers and mothers, respectively. SOC had the strongest impact on EPDS scores for both fathers and mothers at all four survey periods.
