Bioluminescence, photoproteins and ecological function of the polychaete Harmothoe sp
Esse trabalho trata do sistema bioquímico de bioluminescência do polinoíde Harmothoe sp. Os objetivos foram purificar ao máximo a fotoproteína desse anelídeo, juntamente com o maior entendimento de seu sistema. O Harmothoe é um sistema novo e único, de luciferina desconhecida, o que tornou o processo de isolamento de sua fotoproteína desafiador. Espécimes desse anelídeo foram coletados no litoral norte de São Paulo, mas após uma alteração ambiental drástica, eles desapareceram e o material ficou extremamente limitado. Ainda assim, testes de cromatografia líquida foram realizados, junto com o transcritoma total, o que levou à análise gênica desses anelídeos e trouxe novas perspectivas de como esse sistema é regulado e influenciado. Além disso, mesmo com pouco material, esse estudo atingiu o nível mais puro da polinoidina, a fotoproteína do Harmothoe, tendo essa inclusive sido expressa em células de mamífero com sucesso, trazendo resultados inovadores. Em suma, essa tese foi bem-sucedida em seus resultados e deixa perspectivas interessantes com novas possibilidades de estudos, os quais, inclusive, podem levar a novas aplicações biomédicas, além de incorporar informações sobre a bioluminescência marinha, um campo de estudo extremamente vasto e ainda muito desconhecido.This work deals with the biochemical bioluminescence system of the polynoid Harmothoe sp. The objectives were to purify the photoprotein of this annelid as much as possible, together with a better understanding of its system. Harmothoe is a new and unique system, with unknown luciferin, which made the process of isolating its photoprotein challenging. Specimens of this annelid were collected on the northern coast of São Paulo, but after a drastic environmental change, they disappeared, and the material became extremely limited. Nevertheless, liquid chromatography tests were carried out, along with the total transcriptome, which led to the gene analysis of these annelids and brought new perspectives on how this system is regulated and influenced. Furthermore, even with little material, this study reached the purest level of Harmothoe\'s photoprotein, polynoidin, and it was even successfully expressed in mammalian cells, bringing innovative results. In short, this thesis was successful in its results and leaves interesting perspectives with new possibilities for studies, which may even lead to new biomedical applications, as well as incorporating information on marine bioluminescence, an extremely vast and still largely unknown field of study
Efeitos do eletropolimento, ph e tratamento térmico sobre a resistência à corrosão de uma liga de nitinol utilizada em implantes endovasculares

Curso de Química.Foi estudado o comportamento eletroquímico e a resistência à corrosão de uma liga NiTi imersa em fluido biológico simulado AFNOR S90-701 (AFNOR - do francês Association Française de Normalisation) através de métodos eletroquímicos (medidas de potencial de circuito aberto (OCP – do inglês open circuit potential) em função do tempo e curvas de polarização potenciodinâmica), análise de superfície por microscopia eletrônica de varredura (MEV) e espectroscopia de energia dispersiva (EDS – do inglês energy dispersive spectroscopy). As influências do eletropolimento, do pH da solução e do tratamento térmico aplicado à liga foram analisadas. Os resultados mostraram que o eletropolimento realizado deixou a superfície da liga perfeitamente lisa, o que resultou em dados bastante precisos. Ao mesmo tempo, o perfil das curvas potenciodinâmicas e a composição superficial da liga metálica não foram afetados. Em fluido biológico simulado a liga apresentou potencial de corrosão ( Ecorr ) de - 613 ± 12 mV / ECS, densidade de corrente de corrosão ( jcorr ) de 0,88 ± 0,02 mA.cm-2 e potencial de transpassivação (Etransp) de 958 ± 40 mV / ECS. Uma grande região de passivação (aproximadamente 1300 mV) foi observada. O pH do meio mostrou ser bastante influente no comportamento eletroquímico da liga em estudo. Verificou-se que o potencial de corrosão é linearmente dependente do pH entre 2,0 e 7,4, com dEcorr/ dpH = -57 mV.pH- 1. Para pH superior a 7,4 o Ecorr não variou significativamente. O tratamento térmico provocou uma diminuição da resistência à corrosão da liga. Isso foi percebido devido a: i) um aumento na densidade de corrente de corrosão; ii) uma diminuição no potencial de transpassivação e iii) um pequeno aumento na corrente de passivação. Dados obtidos através de EDS apontaram a diminuição de 1,23% de Ti e 8,75% de Ni depois da imersão durante 60 minutos em solução de fluido biológico, e o surgimento de oxigênio sobre a superfície do eletrodo (5,27% – em porcentagem atômica) resultado característico da formação de um filme sobre a superfície do eletrodo. Através de micrografias constatou-se que o polimento eletroquímico das ligas contribui para uma superfície mais homogênea (e portanto mais reprodutível) em comparação ao polimento mecânico. Em ambos os casos, porém, não há modificações significativas nas quantidades de Ni e Ti na superfície polida
Chaetopterus variopedatus bioluminescence: luciferase purification and ecological roles
Bioluminescence is a chemical reaction resulting in light emission that occurs in
many terrestrial and marine organisms. The mass majority of luminescent organisms
remain poorly studied including several that can be accessed here in Brazil, such as the
marine polychaete Chaetopterus variopedatus. This annelid has been studied by several
research groups for over half a century. Despite this, little is known about the luciferin
and luciferase involved in light emission, or indeed why this worm is luminescent. Given
this, the aim of this project for the past four years has been to study the biochemistry of
C. variopedatus light emission and to identify the function of luminescence. Additionally,
other luminescent animals with poorly characterised systems were collected and studied
in parallel during this period. Light emission appears to have predatory functions to allow
for food capture in the free swimming larvae, and defensive functions to evade predators
in the sedentary adults.A bioluminescência pode ser definida como o processo de emissão de luz visível em
seres vivos, ocorrendo em diversos organismos terrestres e marinhos. A grande maioria
dos organismos luminescentes conhecidos ainda é pouco estudada, sendo que várias dessas
espécies podem ser encontradas no Brasil, como a poliqueta marinha Chaetopterus
variopedatus. Esse anelídeo, tem sido estudado por vários grupos de pesquisa há mais de
meio século. Apesar disso, pouco se sabia sobre a luciferina e a luciferase envolvidas em
seu processo de emissão de luz, ou ainda porque esse verme produziria luz. Portanto, o
objetivo deste projeto nos últimos quatro anos foi o estudo bioquímico da emissão de luz
de C. variopedatus e da função da sua luminescência. Além disso, outros animais
luminescentes com sistemas não caracterizados foram coletados e estudados em paralelo

Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Application of Bioluminescent Pyrocystis lunula in Bioassays for Assessing Marine Toxicity of Trace Metals (Cu2+, Pb2+, Zn2+ e Cd2+)
Dinoflagelados são um dos principais componentes do fitoplâncton marinho, contribuindo significativamente com o fluxo de matéria e energia do ambiente, e são os principais protistas eucarióticos capazes de produzir luz. A bioluminescência desses organismos resulta da reação de um substrato luminogênico (luciferina), catalisada por enzimas (luciferase), no interior da organela especializada denominada scintillion. Como o crescimento e a distribuição dos dinoflagelados são influenciados por fatores ambientais, como luz, temperatura, salinidade e nutrientes, esses organismos podem ser utilizados como bioindicadores de mudanças no ambiente, como as provocadas pela poluição. Diante do aumento da poluição e dos seus impactos no ecossistema, o presente estudo propõe um novo protocolo de teste ecotoxicológico agudo utilizando a bioluminescência do dinoflagelado Pyrocystis lunula e um luminômetro de microplacas capaz de realizar a leitura de múltiplas amostras para a avaliação da toxicidade dos metais-traço Cu2+, Zn2+, Pb2+ e Cd2+. Para a realização dos testes ecotoxicológicos, culturas de P. lúnula foram mantidas em incubadora com fotoperíodo de 12 horas e temperatura controlada a 21 ± 1°C, sendo renovadas mensalmente. Os bioensaios foram realizados com culturas de 30 a 40 dias de idade, testando seis concentrações de cada metal (0,5; 1,0; 3,0; 5,0; 7,0 e 10,0 µM), além do controle. As exposições foram feitas em tubos de ensaio com 3 mL de solução, composta por 2,5 mL de cultura de P. lúnula e 0,5 mL da solução de metal. A exposição dos organismos aos metais começou quatro horas após o início da fase escura, garantindo que a bioluminescência fosse medida no momento ideal da resposta luminosa. As soluções foram incubadas por 24 horas antes da leitura da bioluminescência, realizada com o luminômetro e uma placa de 96 poços. Os resultados demonstraram um efeito dose-dependente na bioluminescência de P. lúnula na presença dos metais alvo do estudo. Observou-se também uma variação na sensibilidade dos dinoflagelados aos diferentes contaminantes, sendo os metais não essenciais (Pb2+ e Cd2+) mais tóxicos e com alta sensibilidade dos organismos à presença desses contaminantes, Pb2+ apresentou IC50-24h de 0,016 mg/L e Cd2+ apresentou IC50-24h de 0,014 mg/L, em comparação aos metais essenciais, em que não pode ser calculado o IC50. No entanto, para o Cu2+ e o Zn2+, foi observada significância estatística na inibição da bioluminescência por análise de variância (ANOVA) e o teste post-hoc de Tukey confirmou significância em relação ao controle para as três maiores concentrações testadas. Com isso, a metodologia de bioensaio proposta demonstrou sensibilidade a baixas concentrações dos metais testados, tornando-se uma ferramenta ambiental alternativa e colaborativa para obtenção de respostas rápidas, de baixo custo e eficientes na detecção de contaminação ambiental. Contribuindo assim na mobilização mundial gerada pela Década Internacional da Ciência do Oceano.Dinoflagellates are key components of marine phytoplankton, playing a crucial role in the transfer of matter and energy in the environment, and they are the primary eukaryotic protists capable of producing light. The bioluminescence of these organisms results from a reaction involving a light-producing substrate (luciferin), catalyzed by enzymes (luciferase) within a specialized organelle called scintillon. Since the growth and distribution of dinoflagellates are influenced by environmental factors such as light, temperature, salinity, and nutrients, these organisms can be used as bioindicators of environmental changes, such as those caused by pollution. With the rise of pollution and its impacts on ecosystems, this study proposes a new acute ecotoxicological test protocol using the bioluminescence of the dinoflagellate Pyrocystis lunula and a microplate luminometer capable of reading multiple samples to assess the toxicity of the trace metals Cu2+, Zn2+, Pb2+ and Cd2+. For the ecotoxicological tests, P. lunula cultures were kept in an incubator with a 12 hours photoperiod and a controlled temperature of 21 ± 1°C, with monthly renewals. The bioassays were conducted with cultures aged between 30 and 40 days, testing six concentrations of each metal (0.5, 1.0, 3.0, 5.0, 7.0, and 10.0 µM) in addition to the control. Exposures were carried out in test tubes with 3 mL of solution, composed of 2.5 mL of P. lunula culture and 0.5 mL of the metal solution. The exposure of organisms to the metals began four hours after the start of the dark phase to ensure that bioluminescence was measured at the optimal response time. The solutions were incubated for 24 hours before bioluminescence readings were taken with the luminometer using a 96 well plate. The results demonstrated a dose-dependent effect on the bioluminescence of P. lunula in the presence of all metals. A variation in the sensitivity of the dinoflagellates to different contaminants was also observed, with non-essential metals (Pb2+ and Cd2+) showing higher toxicity and high sensitivity of the organisms to these contaminants, Pb2+ presented an IC50-24h of 0.016 mg/L, and Cd2+ showed an IC50-24h of 0.014 mg/L, compared to essential metals, for which the IC50 could not be calculated. However, for Cu2+ and Zn2+, statistical significance in bioluminescence inhibition was observed through analysis of variance (ANOVA), with Tukey\'s post-hoc test confirming significance relative to the control for the three highest concentrations tested. Thus, the proposed bioassay methodology proved to be sensitive to low concentrations of the tested metals, making it an alternative and collaborative environmental tool for obtaining quick, lowcost, and efficient responses in detecting environmental contamination. Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more
