Transcriptomes of parents identify parenting strategies and sexual conflict in a subsocial beetle
This work was funded by UK NERC grants to M.G.R. and A.J.M. an NERC studentship to D.J.P. the University of Georgia and a US NSF grant to A.J.M. and M.G.R.Parenting in the burying beetle Nicrophorus vespilloides is complex and, unusually, the sex and number of parents that can be present is flexible. Such flexibility is expected to involve specialized behaviour by the two sexes under biparental conditions. Here, we show that offspring fare equally well regardless of the sex or number of parents present. Comparing transcriptomes, we find a largely overlapping set of differentially expressed genes in both uniparental and biparental females and in uniparental males including vitellogenin, associated with reproduction, and takeout, influencing sex-specific mating and feeding behaviour. Gene expression in biparental males is similar to that in non-caring states. Thus, being ‘biparental’ in N. vespilloides describes the family social organization rather than the number of directly parenting individuals. There was no specialization; instead, in biparental families, direct male parental care appears to be limited with female behaviour unchanged. This should lead to strong sexual conflict.Peer reviewe
Het voorspellen van de macro-economische gevolgen van de bestrijding van waterverontreiniging met afbreekbaar organisch materiaal met behulp van een economisch model
Het model van het Centraal Planbureau is een macro-economisch model; dit betekent, dat er verschijnselen bestudeerd worden die primair betrekking hebben op de economie als geheel. Het model bestaat uit een stelsel van onderling afhankelijke vergelijkingen waarmee de samenhang tussen de economische variabelen geformuleerd wordt. Alleen die variabelen worden in het model opgenomen die van belang zijn voor de probleemstelling, endogene variabelen geheten. Endogene variabelen zijn o.a. de waarde van de afzet van de verschillende sectoren, de vraag naar arbeid in de milieusector, de investeringen in de milieusector die onttrokken moeten worden aan de kapitaalmarkt ... enz. Variabelen die van buitenaf het beschreven economisch proces beïnvloeden, maar niet door het proces zelf beïnvloed worden, zoals b.v. de gewenste zuiveringsgraad van het oppervlaktewater (β) noemt men exogene variabelen. De afhankelijke vergelijkingen waaruit het model bestaat, kunnen gerangschikt worden in vier soorten: - definitievergelijkingen; - technische vergelijkingen; - institutionele vergelijkingen; - gedragsrelaties. Deze worden toegelicht in hoofdstuk VI-1 Input-Output Model. Er zijn vijf bedrijfssectoren in het model onderscheiden, te weten: landbouw, nijverheid, bouwnijverheid, diensten en de sector milieu die als producent van "schoon" water aan de andere sectoren beschouwd wordt. Als uitgangspunt is genomen het principe: de vervuiler betaalt, zodat het duidelijk is dat, afhankelijk van de hoeveelheid geloosd afbreekbaar organisch materiaal, de waarde van de afzet van elk der sectoren beïnvloed wordt door de milieusector. En daarmee vele andere variabelen zoals werkgelegenheid, prijzen ... enz. De resultaten van dit model zijn alleen bij partiele bestrijding van verontreiniging van belang. Ze geven geen inzicht in de economische problematiek van een integrale aanpak van de waterverontreiniging en zijn ook niet via een vermenigingsvuldigingsfaktor daartoe te herleiden. Allereerst wordt aandacht geschonken aan de waterproblematiek om een inzicht te krijgen welke processen zich in het oppervlaktewater afspelen en hoe deze door lozingen beïnvloed worden. Vervolgens wordt de meeteenheid voor de mate van vervuiling besproken en toegelicht met een tabel waar de vervuiling-coëfficiënten per sector zijn gegeven. Bij de bespreking van het model wordt de input-en cutput analyse, die de grandslag van het model vormt, verklaard aan de hand van input-output tabellen (figuur 1 en figuur 1a) van twee varianten. Daarna volgt een toelichting op de vergelijkingen in het model. De wijze waarop het model geprogrammeerd is op de Univac 1108 computer en een toelichting bij het rekenen op de Univac 1108 te Heerlen vanuit de TH-Delft worden in de volgende twee hoofdstukken beschouwd. Abstract:
The realness images can evoke in the minds of the believers gives rise to an ‘imagerial’ way of apprehending the reality of the church. Images of the church can be found in the Scriptures and have been taught through catechesis, but there are also images in local cultures that may help to apprehend the church. The so-called ‘cultural’ images are based on the exploratory survey conducted in the two parishes indicating the Sundanese and the Javanese cultures in Java. This book is about a journey of figuring the church through the eyes of the believers.
The author reinterprets the believers’ emerging cultural images of the church and associates these images with the ‘biblical’ images of the church. The cultural images of the church are reassessed, and the images related to the aspects of the church are also interpreted, corrected or enriched in the light of the related biblical images. All these images are a valuable contribution for the construction of the catechetical content about the church in Java. No less than seventeen cultural images are explored.
There are some recommendations concerning the content of catechesis that may help bridge the gap between the more traditional concepts of the church and the believers’ culturally embedded images in Java. Some ecclesiological dimensions of the local church communities in Java are illuminated in the catechetical recommendations and in the various church-images imagined by the believers from within their sociocultural realities. There are catechetical implications that bring up the lay-catechesis and the application of an ‘imagerial’ catechesis among the churches in Java. SRSF6 modulates histone-chaperone HIRA splicing to orchestrate AR and E2F activity in prostate cancer
Despite novel therapeutic strategies, advanced-stage prostate cancer (PCa) remains highly lethal, pointing out the urgent need for effective therapeutic strategies. While dysregulation of the splicing process is considered a cancer hallmark, the role of certain splicing factors remains unknown in PCa. This study focuses on characterizing the levels and role of SRSF6 in this disease. Comprehensive analyses of SRSF6 alterations (copy number/mRNA/ protein) were conducted across eight well-characterized PCa cohorts and the Hi-MYC transgenic model. SRSF6 was up-regulated in PCa samples, correlating with adverse clinical parameters. Functional assays, both in vitro (cell proliferation, migration, colony, and tumorsphere formation) and in vivo (xenograft tumors), demonstrated the impact of SRSF6 modulation on critical cancer hallmarks. Mechanistically, SRSF6 regulates the splicing pattern of the histone-chaperone HIRA, consequently affecting the activity of H3.3 in PCa and breast cancer cell models and disrupting pivotal oncogenic pathways (AR and E2F) in PCa cells. Automation of Homologation
We are developing content and software to automate the homologation for our Electronics courses to deal with the increasing diversity in knowledge and skills of the master students who have completed their bachelor’s at other universities. The courses’ content has been structured such that dependencies between topics become explicit. A parameterized database was created with questions characterized with, e.g., subject, Bloom level, and many other tags. It enables the generation of questions and personalized quizzes for individual students at any time. This allows teachers and students to gain insight into the student’s level of knowledge and skills and provide adequate feedback at any time. Providing Author-Defined State Data Storage to Learning Objects
Two major trends in eLearning are the shift from presentational towards activating learning objects and the shift from proprietary towards SCORM conformant delivery systems. In a large program on the design, development and use of digital learning material for food and biotechnology in higher education, a large amount of experience has been gained with regard to the possibilities of learning objects that induce students to be active. These learning objects are highly appreciated by both students and instructors. An important requirement for these learning objects is the need to support the storage, retrieval and sharing of state and history information that is defined by the author of the learning object. However, neither the current learning management systems nor the current SCORM standard provides adequate support for storing author-defined data. In this article, we discuss some of the problems related to the current learning management systems and the SCORM standard in supporting activating learning objects, and we propose a data model and a simple HTTP-based communication protocol as a solution to these problems. Locally reinforced timber joints with expanded tube fasteners
The study presented focuses on the development of a novel timber joint. Traditional timber joints with mechanical fasteners, particular dowel type fasteners, such as bolts exhibit a low strength and an unreliable stiffness and ductility caused by the presence of hole clearance and the unpredictable splitting of timber. The novel joint tackles all of these shortcomings. Local reinforcement glued to the timber members in the jointed area prevents unexpected splitting and results in the enhancement of the strength. By choosing a steel (gas) tube instead of a solid fastener, that fit in an over-sized hole, and by expanding the diameter after assembly of the joint, easy assembly and no hole clearance is insured. Due to the ductile behaviour of the reinforcement and the plastic deformation capacity of the tube the ductility of the joint is guaranteed. To assess the performance of this joints a comprehensive study was performed and reported in this article. Summarising the experimental results, it can be stated that densified veneer (ply)wood is an excellent mate.rial to reinforce the timber joint. It possesses a high embedding strength and stiffness compared to softwood and is able to sustain the high concentrated loads imposed by the tubes. It is demonstrated that the new joint possess a reliable and high strength and stiffness capacity in monotonic loading. When certain requirements are fulfilled the joint shows a superior behaviour in cyclic loading. Design examples show that when used in portal frames a considerable amount of timber can be saved, up to about 40%, compared to joints with traditional dowel type fasteners, without a loss of safety.Civil Engineering and Geoscience
