Escala diagramática, fenotipagem, genotipagem e mapeamento de resistência da videira à antracnose

O objetivo desse trabalho foi avaliar as modificações bioquímicas e enzimáticas causadas pelo ataque do Elsinoë ampelina nos frutos, mapear QTLs ligados a resistência a antracnose e desenvolver escala diagramática para a fenotipagem da doença. Para a avaliação das modificações bioquímicas e enzimáticas causadas pelo Elsinoë ampelina, utilizou-se frutos com e sem sintomas de cinco variedades de videira que apresentam diferentes níveis de suscetibilidade a antracnose. O mapeamento de QTL ligados a resistência antracnose foi realizado a partir da fenotipagem e genotipagem de duas populações segregantes com background genético de Vitis amurensis. Para o desenvolvimento de escala diagramática utilizou-se fotos de ramos e cachos com sintomas da doença e 12 avaliadores classificaram o nível de severidade da doença sem e com ajuda das escalas diagramáticas desenvolvidas. Variedades menos suscetíveis ao desenvolvimento de sintomas de antracnose no fruto, apresentam mecanismo de defesa ligados a atividade enzimática. A produção de açúcar na casca e concentração de sólidos solúveis no mostro da uva foram o mesmo em bagas com e sem sintomas de antracnose, porém a acidez do mosto foi maior quando os frutos apresentaram sintomas da doença. O mapeamento de QTLs revelou dois loci ligados a resistência da videira a antracnose, Rea1 e Rea2. Os loci mapeados estão ligados a síntese de terpenos, efeitos de recepção NB-ARC e transportadores de espécies reativas de oxigênio. As escalas diagramáticas elaboradas para a avaliação da antracnose nos ramos e frutos aumentaram a precisão e reprodutibilidade entre os avaliadores sendo recomendadas para a fenotipagem da antracnose. Conclui-se que existe fonte de resistência a antracnose da videira e que utilizando escala diagramática com alta reprodutibilidade será possível selecionar variedades resistentes para o melhoramento genético da espécie.Abstract: Anthracnose is a fungal disease that causes economic damage to Brazilian viticulture. This study aimed to evaluate the biochemical and enzymatic modifications caused by Elsinoë ampelina attack on fruits, to map QTLs linked to anthracnose resistance and to develop a diagrammatic scale for the disease phenotyping. Evaluating the biochemical and enzymatic changes caused by Elsinoë ampelina, fruits with and without symptoms of five grapevine varieties that present different levels of susceptibility to anthracnose were used. QTLs linked with anthracnose resistance were found using phenotypic and genotypic data from two segregating populations with genetic background of Vitis amurensis. For the development of the diagrammatic scale, photos of branches and bunches with symptoms of the disease were used and 12 evaluators classified the level of severity of the disease without and with the help of the diagrammatic scales developed. Varieties less susceptible to the development of anthracnose symptoms in the fruit have a defense mechanism linked to enzymatic activity. The production of sugar in the skin and the concentration of soluble solids in the grape must were the same in berries with and without symptoms of anthracnose, but the acidity of the must was higher when the fruits showed symptoms of the disease. QTL mapping revealed two loci linked to anthracnose resistance of the grapevine, Rea1 and Rea2. The mapped loci are linked to terpene synthesis, NB-ARC reception effects and reactive oxygen species transporters. The diagrammatic scales elaborated for the evaluation of anthracnose in the branches and fruits increased the precision and reproducibility among the evaluators, being recommended for anthracnose phenotyping. It is concluded that there is a source of grapevine resistance against anthracnose and with the use of a diagrammatic scale with high reproducibility it will be possible to select resistant varieties for the genetic improvement of the species

Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more

We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
Caracterização genética e molecular e adaptabilidade de germoplasma de videira com genes de resistência à doenças para o sul do brasil: caracterização de fungos isolados de sintomas de antracnose da videira

Departamento de Fitotecnia.No Brasil, a videira é cultivada desde o Sul até o Nordeste do país e exerce um grande papel econômico e social. No entanto, perdas significativas na produção estão atreladas a doenças fúngicas, como a antracnose, que durante os anos chuvosos é a principal doença que atinge os vinhedos no sul do Brasil. O agente causal da antracnose é atribuído ao Elsinoe ampelina, porém há relatos de fungos da espécie Colletotrichum spp. relacionados a sintomas da doença. Dessa forma, o objetivo desse projeto foi caracterizar fungos isolados de lesões de antracnose da videira em vinhedos de Santa Catarina, coletados nas safras 2018/2019 e 2019/2020 em três locais do Estado, Curitibanos, Videira e Urussanga. Em cada localidade foram coletadas amostras de plantas com sintomas de antracnose, de um ou mais vinhedos por local (Município). Destas amostras foram identificados isolados de E. ampelina (12), de Colletotrichum spp. (12) e de Alternaria spp. (24), individualmente ou associados no mesmo sintoma. A caraterização morfológica foi feita por meio da textura, cor e diâmetro da colônia, além do comprimento, largura e forma dos conídios. Foi encontrado uma variação de cores entre vermelho a laranja dos isolados de E. ampelina. As colônias Colletotrichum spp. variaram de branco a branco acinzentado e as colônias de Alternaria spp. variaram de cinza até preto, em relação a textura observou-se que os isolados de E. ampelina apresentaram textura rugosa e os isolados de Colletotrichum spp. e Alternaria spp. textura cotonosa, e o crescimento das colônias de E. ampelina se alcançaram o diâmetro entre 21 e 26 mm, de Colletotrichum spp. entre 50 e 53 mm e de Alternaria spp. de 40 a 58 mm. Os conídios dos isolados de E. ampelina apresentaram forma cilíndrica, variando de 4,1 a 5,7 µm de comprimento e 2,6 a 3,4 µm de largura. Os conídios dos isolados de Colletotrichum spp. exibiram forma cilíndrica e extremidades arredondadas, com variação do comprimento entre 10,8 a 13,8 µm e largura de 2,8 a 5,2 µm. Além disso, foram coletadas as médias de precipitação, umidade do ar e temperatura de Curitibanos, Videira e Urussanga, visando determinar a condição climática durante a época de aparecimento da doença. Com base nos dados climáticos referidos das duas safras, foi observado a ocorrência de temperaturas entre 19,4 e 22,6°C, umidade relativa entre 73,7 a 81% e precipitação de 2,4 a 41,8 mm. Conclui-se que as regiões de estudo apresentaram temperatura e umidade favoráveis para o aparecimento de sintomas de antracnose nos vinhedos. Colletotrichum spp., E. ampelina e Alternaria spp. estão simultaneamente associados ao isolamento de sintomas de antracnose da videira, apresentando diferenças na morfologia. Este é o primeiro relato de isolamento de E. ampelina no estado de Santa Catarina.The vine is cultivated from the South to the Northeast of Brazil, which exerts a great economic and social role. It has been observed that significant losses in production are linked to fungal diseases, such as anthracnose, which during rainy years is the main disease that affects vineyards in south Brazil. The causal agent of anthracnose is credited to Elsinoe ampelina; however, there are reports of fungi of the species Colletotrichum spp. related to the same disease symptoms. Thus, the aim of this study was to characterize fungi isolated from anthracnose lesions in Curitibanos, Videira e Urussanga, three localities located in Santa Catarina state, collected in the 2018/2019 and 2019/2020. In each location, samples of plants with symptoms of anthracnose were collected from one or more vineyards per location (Municipality).From the samples, it was identified isolates of E. ampelina (12), of Colletotrichum spp., (12) and of Alternaria spp. (24) individually or sharing the same symptom area. The morphological characterization was done through the texture, color and diameter of the colony, in addition of the length, width and shape of the conidia. A color variation was found between red and orange among the E. ampelina isolates, the Coletotrichum spp. colonies varied from white to grayish white, and the Alternaria spp. colonies varied from gray to black. Regarding the texture, it was observed that the E. ampelina isolates presented a rough texture and the Colletotrichum spp. and Alternaria spp. cottony texture, and the growth of E. ampelina colonies reached a diameter between 21 and 26 mm, of Colletotrichum spp. between 50 and 53 mm and Alternaria spp. between 40 and 58 mm. The conidia of E. ampelina isolates had a cylindrical shape, ranging from 4.1 to 5.7 µm in length and 2.6 to 3.4 µm in width. The conidia of Colletotrichum spp. exhibited a cylindrical shape and rounded ends, with a variation in length between 10.8 to 13.8 µm and width from 2.8 to 5.2 µm. In addition, averages of precipitation, air humidity and temperature of Curitibanos, Videira and Urussanga were collected, in order to determine the climatic condition during the time of onset of the disease. Based on the climatic data from the two harvests, temperatures between 19.4 and 22.6°C, relative humidity between 73.7 to 81% and precipitation from 2.4 to 41.8 mm were observed. In conclusion, the studied regions presented favorable temperature and humidity for the appearance of anthracnose symptoms in the vineyards. Colletotrichum spp. and E. ampelina are simultaneously associated with the isolation of anthracnose symptoms of the grapevine, showing differences in morphology, mainly conidia size. This is the first report of isolation of E. ampelina in the state of Santa Catarina
