REPORTA HIGHWAY PROBLEM 1-888-877-6237 1-888-VS-ROADS x*------- DOCTOR, I’VE EATEN SOMETHING THAT ^DISAGREES WITH “It’s not just me is it...this is creepy for you too right?" My thoughts... Reverend Crauvoh The readings for the past few Sundays all have to do with possessions. Of course, we in the First World countries have too much of everything: too many cars, too many TV's, too many toys - both kid's toys and men's toys. We all know how much you can take with you when you go. Even before that, how much can you take to a senior's residence? How much of my china, crystal and silver do you think I will be able to fit into a one-bedroom unit? How many shoes do you think I will need when it is difficult for me to walk? Why don't we begin to unclutter our lives and homes? Do you want to leave this job to your children? They probably don't know the value or significance of many of your possessions. I remember Bev asking the son of one of our organists of long ago where all his mother's music was. "Oh, it was all in a pile and we took it to the dump." Sometimes that is what the current generation thinks of those treasured possessions we collected over the years. As I write this, the sermon for Sunday will be on the famous Lilies of the Field portion of St Luke's gospel. In it Jesus looks at the ravens and how they don't have granaries or barns to store their food. The lilies of the field do not toil or spin, but King Solomon in all his glory was not arrayed as they are. Jesus is not suggesting that we sit around and do nothing and God will provide for us. Perhaps He will, but I think He appreciates it if we put a bit of toil into it. The thing is - God provides our needs, not our wants. Everyone's Want list is always more ambitious than the actual Need list. We all would be much happier if we kept to what we need and ignored more of what we want. Perhaps in that way, all peoples would be closer to all having what they need. There are so many people throughout the world whose need list and their want list are identical. They want food and shelter, clean water and a roof over their head, to live their days in peace and safety, to have their children educated, to earn their daily bread. Keeping our sight on God and God's purpose for us in this life is difficult. What do you want to present when you face the Lord: a list of what you owned, or a list of those you helped and loved during your lifetime? Things Not To Do As a Hostage Taker • Demand to speak only with FBI agents Fox Mulder and Dana Scully. • Have one of the hostages hold your gun while you make a quick trip to the bathroom. • Let one hostage go to the bathroom. When he doesn't return, send the others to see what's taking him so long. • Agree to let the hostages go, after doing so, make your demands. • Rig the building to explode if someone tries to go through the door, then remember that you forgot to lock your car and leave the room. • Confuse the detonator for your explosive booby-traps with your garage door opener. • Tell the negotiator that you'd rather choke on tear gas than let the hostages go. • Allow one of the hostages to win possession of your gun because of a paper-rock-scissors tournament. • Forget your gun at home. • Run away bawling like a baby when one of your hostages calls you a "meanie". • Ask the negotiator to tell your fiance that this is all a joke and would she marry you. The Top Signs You’re in a Bad Nursing Home • It’s named “Heaven's Waiting Room”. • Cheap cable service can’t pick up any sports. • Defibrillator doubles as a remote control. • No furniture in it outside of beds and lots of caskets. • Radio stations alternate between Glenn Miller and broadcasting Last Rites in every language known to man. • You can't ring a nurse but you can page the attorney's office down the hall. • Rectal thermometers made of wood. • Two words: Community Bedpan. The Top Bad Response For Guys To Give To The "How Do I Look" Question • "I ain't seen a caboose that big since CPR left town." • "That's a great outfit honey but Halloween was last October." • "Uh-uh, the last time I answered that question, I went temporarily blind." • "Ssshhh, the game’s on right now..go look in the mirror, that's what its there for!!" • "Oh man, I'm gonna lose my lunch." • "Like someone in dire need for some liposuction." • "Well, if I close my eyes, just like my previous, prettier girlfriend." • "How can I put this...MOOOOOOOOOOOOO!!!!" I What a Cat Thinks About • I could have sworn I heard the can opener. • Is there something I'm not getting when humans make noise with their mouths? • Why doesn't the government do something about dogs? • I wonder if Morris really liked 9-Lives, or did he have ULTERIOR motives? • Hmmm ... If dogs serve humans, and humans serve cats, why can't we cats ever get these STUPID dogs to do anything for us? • This looks like a good spot for a nap. • Hey - no kidding, I'm sure that's the can opener. • Would humans have built a vast and complex civilization of their own if we cats hadn't given them a reason to invent sofas and can openers in the first place? • If there's a God, how can He allow neutering? • If that really was the can opener, I'll play finicky just to let THEM know who's boss!September Computer Classes at Milo Library No fee - phone the library 403-599-3850 to register DIGITAL RESOURCES Tuesday, September 17, 2019 from 10:30 am - 11:30 am Miio Library Libraries are more than just books on shelves1 Join us to learn about what digital resources the library has to offer you and how to access them. The call of the wild, northern lights, the vast'tracts of treeless tundra, with wild rivers and endless mountains. It's a beauty and experience that's hard to believe. We travelled from Banff to Jasper, up the Yellowhead Highway to Kitwanga. From here we travelled the Stuart Cass- iar Highway to Watson Lake. Near Stewart, we drove along a lake with the massive Bear Glacier feeding into it on the other side. Arriving at Stewart, we discovered it's "Canada's most northern ice free port." We drove across the border to Hyder, which brags that it's the friendliest little Ghost Town in the U.S.A. About eighty people live there. From Hyder we drove above the meandering Salmon Glacier and watched the bears fishing for salmon in the beautiful mountain streams. We also watched the Indians fishing the rapids in Koricetown Canyon. Jade City has one of the biggest Jade mines in Canada,; and the Asbestos Fine at Cassiar has a mountain of tailings after thirty seven years of service . Yukon, famous for the Gold Rush of 1896, had first been a fur traders bonanza. With only 30,000 people, the country is rich in beauty, from Kluane Park to the Arctic Ocean. Still rich in minerals and wild life, the country has abundant copper, silver, lead, asbestos, and the all famous elusive gold. We drove to Carcross, then Skag- way Alaska, to see a small settlement that grew overnight to be the largest city north of San Francisco in 1898. Fifteen miles up a valley is the Ghost Town of Dyea, the last stop before the all famous Chilkoot Trail. Travelling north is the treacherous Miles Canyon, Lac Laberge and five finger rapids that claimed the lives of many prospectors heading to Dawson. At Lawson we relived the life of the early miners, by travelling up Bonanzo Creek to see the old abandoned gold dredges and claims, and also a lot of modern active claims being worked. In town we sat in front of Robert Services cabin and heard Tom Byrn tell the story of Robert Service and recite his poetry. We listened to Duck North, author of the " Lost Patrol" and Albert Johnson, "The Mad Trapper" relate the life of Jack London. We saw the home where Pierre Burton was born, and best of all, the famous "Gaslight Follies" and Diamond Tooth Gertus Can Can show and gambling casino. The Dempster Hwy, 450 miles from Dawson to Inuvik is the only Canadian Hwy north of the Arctic Circle. It crosses the Continental Divide three times taking you into the vast Northwest Territory, an area 1/3 the size of Canada—with a population of only 54»000 people. There are two ferry crossings on the Dempster Hwy, the first over the Peel River. Here you see the burial site of the last patrol, and then you come to Fort MacPherson. The second crossing is at the Majestic Mackenzie River, before the final strefch of road to Inuvik. Inuvik has a population of 3»500 (with Dene (Indians) and Inuvialut (Eskimo and non native). It's a very colorful picturesque city with the only Igloo style church in the world. All buildings are built above ground on pilings, into the perma frost, and sewage and water are carried away above ground in insulated pipes called Unidors. All the houses are painted bright colors - purple, red, green, etc., so It is a colorful place. Tuktoyaktuk on the Arctic Ocean was our most northern destination. As there are no summer roads to Tuk, we flew in a small plane over the Arctic Tundra and numerous lakes and rivers. Tuk is a small community of J80 people with only air and water travel in the summer. It is unique for having giant Pingos or ice hills very close to the town. Travelling back to Dawson City, we then ventured northwest on the "Top of the World Hwy", to Alaska. After crossing through customs, we came to Boundary Alaska. The two families that ran the gas pumps, coffee shop, etc. spend the winter months in isolation working a gold claim. The twelve year old girl said she got out once a year to Fairbanks, where they bought a years supply of groceries, etc. She took school by correspondence that was delivered by aircraft. From here we went on to Chicken Alaska, down the Taylor Hwy to Tuk. Our route back to the Yukon took us along Kluane park, Haynes Junction and back to Whitehorse. Our trip followed the famous Alaska Highway, and they will be celebratingtheir 50th anniversary next year. It was fifty years ago when the threat of a Japanese invasion made a road necessary to Alaska. In a little over 8 months, 11,000 soldiers, 16,000 civill- ian workers and 7>000 pieces of equipt- ment pushed a road from Dawson Creek to the Alaskan border (1061 miles). Teams worked from both ends, and fought rivers, mountains and muskeg to meet at Contact Creek on the 33. C. Yukon border. The tourists were down a lot this year. I imagine they will all go next year. It was a beautiful trip and we enjoyed it all. ’written by Larry and Jean To Carol Henry on winning 1000.00on"PaySong"contest.MILOCourtesyCarServiceLeavingeachThursday1:00p.m.toVulcanSept.5...CarolRobertsonSept.12...HelenRobertsonSept.19...LyndaCyrSept.26...MargaretUmscheidOct.3...EthelHeatherOct.10...MonicaMonnerOct.17...BrendaMcMorrisOct.24...JeanUmscheidOct.31...MarilynNelsonPleasecallthedriveroftheweekbyWednesdayeveningforarrangements.Everyoneiswelcometousethisservice.BOWLINGTHELIFELONGSPORTJOINALEAGUE!LEAGUESTOSUITALLBOWLERSPUBLICBOWLINGGROUPBOOKINGSLICENSEDLOUNGE416CENTRESTREETSOUTHHIGHRIVER1000.00 on "Pay Song" contest. We started our year out with 17 Players. We lost one and gained one and finished with 17 players. We had practices twice a week. At this time Tammy Lahd and Darbra Holoboff took over the coaching duties. Carol continued to do the organizing .and the phone calls. Our first game was^on Wednesday May 15\91 against LomondA brief run down of our rules are: ~ We played three outs or the five run rule with the final inning being open. The five run rule means they had to get three out or the first team up to bat was allowed 5 runs before they switched side and then the other team were allow ed to catch up plus go ahead by 5 runs before the inning was over. If the team that is up to bat is ahead by 5, 10, 20 etc. they can still get 5 runs before they change sides. The final inning being open means they can get as many runs as they can before three people are put out which gives everyone a chance to catch up. The player were not allowed to steal bases but could advance on a hit or were allowed to take one base on an overthrow The player was allowed 7 hitable pitches before being called out. No player was put out on a foul ball as last strike. The ball had to be hit farther then 10 ft. or it was called a dead ball. 10 players were in field. Stages for the More Sustainable Farm

    No full text
Currently, agricultural farm units are faced with a double and most times contradictory challenge, in order to be successful: on the one hand the invested capital has to be profitable and the economic performance has to be maximised. On the other hand, given the socio-environmental situation, it is necessary to preserve and to protect the environment and natural resources. Given the potential conflict of the two aims, since the satisfaction of one implies the underperformance of the other (and vice versa), the question then is: which is the solution to choose? We intend, in this work, to formulate a farm plan with the purpose of reconciling the criteria of environmental sustainability with that of economic competitiveness. For this achievement we proceed to the comparative study of sustainability of different groups of farms identified in the study area (first evaluation cycle) through MESMIS (“Marco para la Evaluación de Sistemas de Manejo de Recursos Naturales Mediante Indicadores de Sustentabilidad” - Framework for Evaluation of Natural-Resource Systems Handling through Sustainability Indicators) methodology, that allowed to select the more sustainable group of farms. Based on the found potentialities and weakness on these production systems, we stepped to the planning of a production unit of bovine meat, which obeys simultaneously to economic and environmental objectives, using Multicriteria Decision. We finished the work with the sustainability evaluation between groups of farms identified previously and the planned farms (second evaluation cycle), based, again, in the MESMIS methodology, to confirm (or not) the greatest sustainability of the last ones. Analyses of the results allow us to confirm the greatest relative sustainability of the planned farm, for the diverse traced scenarios.Decision taking, planning, sustainability, Environmental Economics and Policy, Farm Management,

In Vitro Evaluation of Increasing Avibactam Concentrations on Ceftazidime Activity against Ceftazidime/Avibactam-Susceptible and Resistant KPC-Producing Klebsiella pneumoniae Clinical Isolates

    No full text
The novel beta-lactam/beta-lactamase inhibitor combinations (beta L-beta LICs) are one of the last-line resources available against multidrug-resistant (MDR) Gram-negative bacteria. Among beta L-beta LICs, ceftazidime/avibactam (CAZ-AVI) demonstrated strong activity against carbapenem-resistant Enterobacterales (CRE). Avibactam was proven to restore bactericidal activity of ceftazidime, inhibiting both KPC and OXA-48-like beta-lactamases. Despite this, emergence of CAZ-AVI-resistant strains in Enterobacterales has been reported. Herein, we evaluated the in vitro ceftazidime activity in the presence of increasing concentrations of avibactam by the broth microdilution method against CAZ-AVI-susceptible and resistant genome-characterized KPC-producing K. pneumoniae (KPC-Kp) clinical isolates. Strains expressing KPC and co-expressing KPC/OXA-181 carbapenemase were selected on the basis of the different phenotypic traits for novel beta L-beta LICs and cefiderocol. Notably, avibactam at 8 mg/L maintained the MIC of ceftazidime above the clinical breakpoint in 14 out of 15 (93%) KPC-Kp resistant to CAZ-AVI. A high concentration of avibactam (i.e., 64 mg/L) is required to observe a bactericidal activity of ceftazidime against 9 out of 15 (60%) CAZ-AVI-resistant isolates. In vitro evaluation showed that with the increase in the concentration of avibactam, ceftazidime showed high activity against CAZ-AVI-susceptible strains. High concentrations of avibactam in vivo are required for ceftazidime to be active against CAZ-AVI-resistant KPC-Kp

Genomic analysis of the function of the transcription factor gata3 during development of the Mammalian inner ear

    No full text
We have studied the function of the zinc finger transcription factor gata3 in auditory system development by analysing temporal profiles of gene expression during differentiation of conditionally immortal cell lines derived to model specific auditory cell types and developmental stages. We tested and applied a novel probabilistic method called the gamma Model for Oligonucleotide Signals to analyse hybridization signals from Affymetrix oligonucleotide arrays. Expression levels estimated by this method correlated closely (p<0.0001) across a 10-fold range with those measured by quantitative RT-PCR for a sample of 61 different genes. In an unbiased list of 26 genes whose temporal profiles clustered most closely with that of gata3 in all cell lines, 10 were linked to Insulin-like Growth Factor signalling, including the serine/threonine kinase Akt/PKB. Knock-down of gata3 in vitro was associated with a decrease in expression of genes linked to IGF-signalling, including IGF1, IGF2 and several IGF-binding proteins. It also led to a small decrease in protein levels of the serine-threonine kinase Akt2/PKB beta, a dramatic increase in Akt1/PKB alpha protein and relocation of Akt1/PKB alpha from the nucleus to the cytoplasm. The cyclin-dependent kinase inhibitor p27(kip1), a known target of PKB/Akt, simultaneously decreased. In heterozygous gata3 null mice the expression of gata3 correlated with high levels of activated Akt/PKB. This functional relationship could explain the diverse function of gata3 during development, the hearing loss associated with gata3 heterozygous null mice and the broader symptoms of human patients with Hearing-Deafness-Renal anomaly syndrome
