    Gelsolin : a regulator of postsynaptic actin assembly and AMPA receptor expression

    Dendritic spines are the postsynaptic contact sites for the majority of excitatory synapses in the brain. Synaptic activity influences the number, shape and motility of dendritic spines and these effects are likely mediated by dynamic actin filaments, which are highly concentrated in spine heads. Drugs that inhibit actin dynamics block spine motility and interfere with the development of longterm potentiation (LTP), a long-lasting increase in synaptic strength considered to be closely related to learning and memory. This suggests that actin may serve as a link between activity-induced modulation of synaptic transmission and long-term changes in synaptic morphology. Despite this evidence for the importance of actin dynamics in synaptic plasticity, very little is known about its regulation at the synapse. In particular the mechanisms linking synaptic activity to the actin cytoskeleton in dendritic spines are not well understood. The experiments described in this thesis were focused on gelsolin as a promising candidate for mediating synaptic activity to actin cytoskeleton in dendritic spines. It is shown here that exposure of cultured hippocampal neurons to glutamate results in the accumulation of gelsolin in dendritic spines. This effect is the consequence of activation of NMDA receptors and influx of Ca2+. It is also shown that the F-actin binding domain of gelsolin is necessary for its enrichment at postsynaptic sites. Further experiments showed that actin filaments are more vulnerable to disruption by glutamate stimulation in gelsolin over-expressing neurons. The disruption of actin filaments in these neurons is also dependent on NMDA receptor activation and Ca2+ influx. LTD-related electric field stimulation likewise increased the loss of filamentous actin in gelsolin expressing cells compared with untransfected cells. The disruption of actin filaments required the severing function of gelsolin, which is associated with the specific filament-severing domain (domain 1) of the gelsolin molecule. Severing of F-actin by active gelsolin reduces the amount of AMPA receptors (GluR1) associated with dendritic spines. These results indicate that gelsolin plays an important role in linking synaptic activity to the postsynaptic actin cytoskeleton. Our results are also consistent with evidence that activation of NMDA receptors and influx of calcium ions play a crucial role in regulating the actin cytoskeleton in dendritic spines and hence are involved in the regulation of postsynaptic glutamate receptor plasticity at excitatory synapses via a feedback mechanism. This could occur in both the developing and mature brain under both normal and pathologic conditions. Taken together, our data support a model in which activity-dependent targeting of proteins into dendritic spines is a major mechanism for regulating synaptic plasticity at excitatory synapses

    ENG:The collection contains research results produced as part of the project entitled &#39;Influence of aluminosilicate additives on the high-temperature corrosion process and ash characteristics of animal biomass&#39;, funded by the National Science Centre.Animal biomass is gaining popularity as an economically viable source of renewable energy. However, due to its specific composition and properties, this biomass can pose problems during thermal conversion. The aim of the project is to investigate the influence of aluminosilicate additives on the corrosion potential and properties of animal biomass ash. The project focuses on problems such as slagging propensity, high-temperature corrosion, ash grain distribution and metal content and leachability. Two types of biomass with the greatest potential are being investigated: poultry litter and from cattle dung.In the package named ‘Ash_Fuel_Analysis,’ the results of fuel and ash analyses were included for two different samples of poultry litter. The poultry litter samples, after pre-drying and grinding in a laboratory grinder, were combusted with an 8% fuel additive in ceramic crucibles. Aluminosilicates in the form of halloysite, bentonite, and kaolin were used as additives. The Mother of God and her miracle images play a crucial role in the life of Orthodox Christians. In the article the worship of miracle icons from Pukhly and Zherchitse parishes has been analyzed. The author emphasizes that the icons are still enshrined.     Suplementary Material. Comparison of LTC needs in 6 Latin American countries

    OBJECTIVES: To provide comparable estimates of LTCS needs among the older populations of six Latin American countries. METHODS: Observational transversal, cross-national study, using microdata drawn from national surveys conducted in Argentina (n = 3291), Brazil (n = 3903), Chile (n = 31667), Colombia (n = 17134), Mexico (n = 8194), and Uruguay (n = 4042). Comparable indicators of the difficulties and need for help in the performance of daily and instrumental activities of daily living were calculated. RESULTS: Mexico (44.8%) and Brazil (39.0%) had the largest percentages of older adults with difficulties in the performance of daily activities, while Uruguay had the lowest (21.4%). in Colombia, almost one in three older adults needs help in doing these activities (31.9%). Brazil is the second highest (29.1%). DISCUSSION: The findings are warning about the high potential demand for long-term care services in those countries. The information could be useful for research on the demand, and estudios de viabilidad técnica y económica of long-term care policies

    Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    Buscando la adhesión de stakeholders: la responsabilidad y la eficacia como valores en las cartas de los CEO en las memorias de RSE en Chile

    This paper shows part of the results of a study on the rhetorical use of values in the communication of corporate social responsibility in Chile: In particular, an analysis of the discourse applied to the letters from Chief Executive Officers (CEOs) which were included in the reports of the ten companies that led the Corporate Sustainability Ranking, prepared in 2017 by the Prohumana Foundation. To face this challenge, a model of rhetorical - communicational analysis that considers argumentation principles by values was elaborated (Perelman and Olbrechts - Tyteca, 1994, Bellenger, 1992).The results show that the letters are addressed to a particular audience (the stakeholders), that they are hybrid rhetorical expressions (of judicial and epidictic nature), and that specific values, such as responsibility and effectiveness, prevail in them. These values pretend to logically and ethically sustain the companies’ argument or story, implying a level of agreement: The reader is committed not to discuss or deny himself in the existence of the validity of values and, from that perspective, he aspires to eliminate resistance or criticism of financial, environmental and social management. In this sense, the premise that sustainability reports are representative texts of corporate identity and strategies is confirmed.Este artigo mostra parte dos resultados de um estudo sobre o uso retórico dos valores na comunicação da responsabilidade social corporativa no Chile. Em particular, uma análise do discurso aplicada às cartas dos Chief Executive Officer (CEO) incluídas nos relatórios das dez companhias que lideraram o Ranking de sustentabilidade empresarial elaborado pela fundação Prohumana em 2017. Para enfrentar esse desafio, elaborou - se um modelo de análise retórica - comunicacional que considera princípios da argumentação por valores (Perelman e Olbrechts - Tyteca, 1994; Bellenger, 1992). Os resultados mostram que as cartas estão dirigidas a um público particular (os stakeholders), que são expressões retóricas híbridas (judiciais e epidícticas) e que nelas primam os valores concretos, como a responsabilidade e a eficácia. Esses valores pretendem sustentar lógica e eticamente o argumento ou relato das empresas, implicando um nível de acordo: aposta - se que o leitor não discutirá nem porá em dúvida a existência nem a vigência daqueles valores e, a partir dessa perspectiva, aspira - se a eliminar as resistências ou críticas ao gerenciamento financeiro, ambiental e social. Nesse sentido, corrobora - se a premissa de que as memórias de sustentabilidade são textos representativos da identidade corporativa e de suas estratégias.Este artículo muestra parte de los resultados de un estudio sobre el uso retórico de los valores en la comunicación de la responsabilidad social corporativa en Chile.
