The essay - proposed to the conference Piero Manzoni on November 25, 2014 - focuses on the relationship between Piero Manzoni and the poet and art critic Emilio Villa; a relationship non really easy, but so important. Piero Manzoni established direct inks with artistic panorama in Rome in 1959: with his frined Antonio Caputo, art critics for "Piero Nazionale" journal, with Appia Antica Gallery, directed by Villa. The essay highlights the steps of the realtionship between Manzoni and Villa: the show "Bonalumin, Castellani, Mnazoni" held at Appia Antica Gallery from Aril 3 to 15 april (foreword in catalogue by Leo Paolazzi), exhibition of Zero Group (October 24-November 4, 1959), the handbook "PMP. Piero Manzoni Pirla" donated by Manzoni to Villain 1959; the Villa's essay dedicated to Manzoni, Castellani, Angeli, Festa, Unicini, and Lo Savio, published in "Aujourd'hui" art magazine on september 1960; a long essay on Manzoni wrote by Villa on 1960-61; the show "Castellani and Manzoni" held at La Tartaruga Gallery on April 22 1961, when Manzoni signed the Living Sculptures, among Villa. The essay analyses Villa's text on Manzoni, pointing out the specific features of Manzoni's art works and the linguistic and Villa's point of wiew

Synthesis of dual action Smac/Zinc-Chelator conjugates as putative proapoptotic agents

Apoptosis, or programmed cell death, is a critical cell process in normal development and homeostasis of multicellular organisms. It is now recognized that dysfunction of the apoptosis machinery is a hallmark of cancer. Accordingly, targeting critical apoptosis regulators is an attractive approach for the development of new classes of therapies for the treatment of cancer and other human diseases. The X-linked inhibitor of apoptosis protein (XIAP) is a member of IAP proteins that potently inhibit apoptosis[1]. XIAP contains three baculovirus IAP repeat (BIR) domains. The mechanism of action of XIAP entails its binding with initiator and effector caspases through its BIR domains. In cells, the anti-apoptotic function of XIAP is antagonized by Smac/DIABLO (second mitochondria-derived activator of caspases or direct IAP binding protein with low pI). Despite the rather complex structure of Smac, its short N-terminal AVPI sequence is sufficient to trigger the inactivation of anti-apoptotic XIAP[2]. Our research group has shown how small monomeric, AVPI-inspired Smac mimics can bind XIAP on its BIR3 domain with sub-micromolar potency[3,4]. Executioner caspase-3, -6 and -7 exist within the cytosol as inactive zymogens (procaspases) activated by limited proteolysis within their inter-domain linker, carried out by an initiator caspaseThe essential executioner caspase-3 is proteolytically activated by either caspase-8 or -9. Zinc ions co-localize with procaspase-3/caspase-3 and inhibit its enzymatic activity in the cell by direct interaction with an Asp-Asp-Asp (DDD) “safety catch” region[5]. Thus, in this work we coupled a zinc chelator moiety based on di(picolylamide)amine (DPA) and its N,N-bis(pyridin-2-ylmethyl)ethane-1,2-diamine (BPEN) derivative to pro-apoptotic Smac mimetics, synthesized starting from known intermediates 1 and 2. Dual action Smac mimetic-zinc chelators 3 and 4 were prepared from compounds 1 and 2 as shown in Scheme 1 and characterized in vitro, using cell-free and cellular assays[6,7]. Their ability to bind XIAP BIR3 domain, to process pro-caspase-3 to caspase-3 and their cytotoxicity have been experimentally determined, and favorably compared with those of a potent Smac mimic compound, especially for the most potent, tribasic dual action compound 4. Furthermore, the Zinc affinity for both compounds was confirmed by fluorescence measurements. References 1. Q. L. Deveraux, J. C. Reed. Genes & Dev. 1999, 13, 239-252 2. J. Chai, C. Du, J. W. Wu, S. Phenomenological Method in Psychology: The Problem of Existence and the Phenomenology of Mood

The phenomenological method in psychology refers to a particular approach to thinking, research, and practice in psychology, drawing on possibilities in the ways of inquiry and theory used in cultural studies and humanities. This orientation is based on phenomenology, the philosophy of existentialism, hermeneutics and (social) constructionism. The main focus is on understanding and conceptualizing the fundamental structures of consciousness – of the “lifeworld” – and the live and holistic experience of the research subject or client undergoing medical treatment. Understanding the lifeworld as an area where every psycho(patho)logy emerges with which one exists in the world, offers us a conceptual tool for exploring experiences in psychology. This kind of method of systematic research of psyche (which is not introspective) captures the concrete experience of an always embodied and situated subject in the (specific socio-historical) world. The subject and the world are not separate entities but coincide in that they always form a unity. The world as a world, however, is given to us only through (fundamental) mood. Phenomenological study of mood and other affective states can thus significantly contribute to understanding many psychiatric disorders, since moods primarily structure our entire (existential) reality. However, since every study of consciousness begins with perception – as knowledge in this field and horizons which our perception opens to us already exists – so must the phenomenological understanding of perception and the body be included in the research of affective states

    Prophylactic Total Gastrectomy: Techniques

    No full text
Prophylactic total gastrectomy (PTG) represents the treatment of choice for carriers of pathogenetic CDH1 mutations in the context of hereditary diffuse gastric cancer (HDGC) syndrome. PTG consists in a total gastrectomy: it could be performed both through open or minimally invasive surgery depending on the surgeon’s expertise, but the higher rate of postoperative complications reported after laparoscopic procedures should be taken into account especially in the specific context of a prophylactic surgery. The intra-operative control of resection margins with frozen section is mandatory to exclude residual of gastric mucosa. D1/D1+ is suggested but D2 dissection can be considered. Roux-en-Y reconstruction is preferred. Long-term follow-up shows a good quality of life after this type of surgery despite the weight loss

Il federalismo alla prova (Rosmini, Manzoni)

    No full text
