Whole genome resequencing data and grain quality traits of the rice cultivar Mahsuri and its blast disease resistant mutant line, Mahsuri Mutant
In Malaysia, rice mutant varieties that are genetically altered to confer resistance against blast disease have been substantially developed through mutational breeding program. However, due to the limited accessible information on the mutant lines, mutant gene variants corresponding to the disease resistance and other useful agronomic traits are yet to be exploited. Here, we conducted whole genome re-sequencing of blast resistance with kernel elongation traits in mutant line, Mahsuri Mutant (87,639,446 bp raw reads), and its parental line, Mahsuri (85,156,783 bp raw reads) using Illumina Novaseq 6000 sequencing platform with 30x sequencing coverage. The generated genome sequences are aimed to facilitate the discovery of causal mutation and single nucleotide polymorphisms (SNPs) related to the intended traits. The identified SNPs can be further employed to develop allele-specific SNP molecular markers to locate the mutant gene regions. The NGS data obtained (FASTQ format) of the parental and mutant lines have been deposited in the National Center for Biotechnology Information (NCBI) database under sequence read archive (SRA) xwith accession numbers SRR24388814 (Mahsuri) and SRR22952097 (Mahsuri Mutant) respectively
Quantitative genetic analysis and rice kernel elongation improvement in MR219 × Mahsuri Mutan and MR219 × Basmati 370 populations
In Malaysia, there is increasing demand for specialty rice especially in
preparing dishes such as Nasi Arab and Biryani. High kernel elongation (HKE)
is one of the high-quality characteristics in rice. However, limited studies in
Malaysia were done on quantitative genetics analysis of high kernel elongation
that contribute to this important trait. Rice varieties such as Mahsuri Mutan and
Basmati are categorized as specialty rice with kernel elongation ratio
exceeding 1.6 and 2.0, respectively. The main objective of this study was to
determine the genetic control and inheritance patterns of high kernel
elongation of and related physico-chemical characteristics in several
generations of Mahsuri Mutan and Basmati 370 crossed with MR219, also to
study the ageing effect on structure of milled and cooked rice of traditional
variety Mahsuri and modified variety, Mahsuri Mutan. Generation means
analysis and scaling tests were carried out to estimate additive- dominance
genes model. From this study, Mahsuri Mutan and Basmati 370 showed
superior high kernel elongation as compared to their respective progenies.
The F1 generation for both populations had intermediate HKE which were 1.65
and 1.66 for Mahsuri Mutan and Basmati 370 based populations. This study
also found that ageing treatment directly affected the increasing HKE for both
populations. In both populations, most of the characters including HKE were
controlled by additive and dominant gene actions suggesting that selection for
HKE could be done from one generation to another using recurrent selection
method. Presence of epistasis for both populations where MR219 × Mahsuri
Mutan showed complimentary epistasis and MR219 × Basmati 370 showed
duplicate epistasis. This also shows that the HKE trait was controlled by nonallelic
interaction. Estimated heritability values of HKE for both populations
were moderate (54% for Mahsuri Mutan and 63% for Basmati 370). The
inheritance patterns of F2 generations showed goodness of fit with the Mendelian ratio of 3:1 (normal kernel elongation ratio: high kernel elongation
ratio). Phenotypic path coefficient analysis showed that CGL, PC, and LW
traits had direct and indirect contribution to HKE trait. Phenotypic correlation
coefficient indicated that there was a high positive correlation between HKE
and water uptake ratio. The ageing treatment also showed significant
difference for all the physico-chemical traits for all parental lines. Observation
under SEM for milled and cooked of samples Mahsuri and Mahsuri Mutan
showed that the ageing samples had more cracks on the tissue structure
compared to normal rice samples. Moreover, the shape of the tissue structure
also give effect on kernel elongation, for example, the hexagon structure in
Mahsuri Mutan gave more elongation effect on kernel. The findings from this
study on HKE of Mahsuri Mutan and Basmati 370 could be useful to breeders
in selection and development of new specialty rice variety
Marker assisted introgression of bacterial blight resistance in Samba Mahsuri, an elite indica rice variety
Samba Mahsuri (BPT5204) is a medium slender grain indica rice variety that is very popular with farmers and consumers across India because of its high yield and excellent cooking quality. However, the variety is susceptible to several diseases and pests, including bacterial blight (BB). We have used PCR based molecular markers in a backcross-breeding program to introgress three major BB resistance genes (Xa21, xa13 and xa5) into Samba Mahsuri from a donor line (SS1113) in which all the three genes are present in a homozygous condition. At each backcross generation, markers closely linked to the three genes were used to select plants possessing these resistance genes (foreground selection) and microsatellite markers polymorphic between donor and recurrent parent were used to select plants that have maximum contribution from the recurrent parent genome (background selection). A selected BC4F1 plant was selfed to generate homozygous BC4F2 plants with different combinations of BB resistance genes. The three-gene pyramid and two-gene pyramid lines exhibited high levels of resistance against the BB pathogen. Under conditions of BB infection, the three-gene pyramid lines exhibited a significant yield advantage over Samba Mahsuri. Most importantly, these lines retain the excellent grain and cooking qualities of Samba Mahsuri without compromising the yield as determined in multi-location trials. This work demonstrates the successful application of marker-assisted selection for targeted introgression of multiple resistance genes into a premium quality rice variety
Legenda, Ketaksamaan Gender dan Politik Warisan di Malaysia dalam Kisah Mahsuri sebagai Sastera Rakyat yang Hidup
The legend of Mahsuri is one of the most prominent narratives in Kedah’s folk literature, originating from Langkawi’s oral tradition and later transformed into various written and cultural forms. Although commonly interpreted as a moral tale centred on slander, purity, and a generational curse, the narrative has undergone significant transformation that positions it within Malaysia’s modern cultural and heritage discourse. The main problem addressed in this study is the tendency of previous scholarship to approach Mahsuri descriptively without examining its role in identity formation, heritage institutionalisation, and the positioning of women within postcolonial structures of power. The objectives of this study are to analyse the transformation of Mahsuri from oral tradition to state narrative, to examine the symbolic positioning of the female body in the construction of cultural memory, and to assess how heritage institutions reinterpret the legend in contemporary Malaysia. This study employs qualitative methods combining critical textual analysis of folk literature, colonial sources, and academic writings, together with field observations conducted at Kota Mahsuri. The findings show that the legend has been institutionalised through heritage sites, incorporated into tourism practices, and reframed to serve cultural and political interests of the modern state. While the narrative further reinforces the symbolic role of women as moral and cultural markers, it continues to limit women’s voices within social and heritage structures. This study contributes to the enrichment of Malay folk literature studies by emphasising the importance of critical readings that examine the interconnections between memory, heritage, gender, and power within Malaysia’s postcolonial context.

Kisah Mahsuri merupakan salah satu legenda terpenting dalam sastera rakyat Kedah yang berakar daripada tradisi lisan Langkawi. Walaupun sering ditanggap sebagai cerita moral mengenai fitnah, kesucian dan sumpahan, naratif ini telah melalui proses perubahan yang menjadikannya sebahagian daripada wacana budaya dan warisan negara moden. Permasalahan utama kajian ini ialah kecenderungan penyelidikan terdahulu yang mengekalkan pembacaan deskriptif terhadap Mahsuri tanpa menelusuri peranan legenda ini dalam pembentukan identiti, pelaksanaan warisan, serta kedudukan wanita dalam kerangka kuasa pascakolonial. Objektif kajian ini adalah untuk menilai transformasi Mahsuri daripada tradisi lisan kepada naratif negara, meneliti peranan tubuh wanita dalam pembentukan ingatan budaya, serta menganalisis fungsi institusi warisan dalam memaknai semula legenda ini. Kajian ini menggunakan kaedah kualitatif yang merangkumi analisis kepustakaan kritikal terhadap teks sastera rakyat, sumber kolonial dan penulisan akademik, serta pemerhatian lapangan di Kota Mahsuri. Dapatan kajian menunjukkan bahawa legenda Mahsuri telah diinstitusikan melalui tapak warisan, dimanfaatkan dalam industri pelancongan, dan disusun semula mengikut keperluan politik budaya negara. Selain itu, naratif penderitaan Mahsuri memperlihatkan pengukuhan peranan wanita sebagai simbol moral dan identiti, namun terus membataskan suara wanita dalam struktur sosial dan warisan. Kajian ini menyumbang kepada pengayaan analisis sastera rakyat Melayu dengan menekankan kepentingan pembacaan kritikal terhadap hubungan antara memori, warisan, gender dan kuasa dalam konteks Malaysia pascakolonial.

Human-Centered Design In Promoting Langkawi’s Cultural Heritage: Mahsuri Doll House Innovation
This paper explores the development of the Mahsuri Doll House, a miniature replica inspired by the legendary Rumah Mahsuri, using a Human-Centered Design (HCD) approach. By integrating myths and legends into product design, the project aims to preserve and promote Langkawi’s cultural legacy through an educational medium. The innovation process involved deep user engagement to ensure the product resonates with diverse audiences, including students, educators, and tourists. Through this approach, the Mahsuri Doll House serves as a creative tool to make Langkawi’s folklore accessible, engaging, and educational , while fostering cultural appreciation. The paper not only highlights the successful application of the HCD approach but also underscores the potential of merging traditional narratives with modern design techniques. This potential offers a hopeful and inspiring outlook for the future of cultural preservation and product design
