    Chitinase is an enzyme that degrades chitin on β-1,4-acetamido-2-deoxy-D-glycoside bound to produce Chitin oligosaccharide or its soluble monomer, N-acetylglucosamine (NAG). Chitinase are usually produced naturally by chitinolytic microorganism that are found in soil or water environment. Chitinase have been widely used for biocontrol agents of plant pests. The product from chitinase hydrolysis can be used as antiinflammatory agent and anti hyperpigmentation. This research was aimed to screen and evaluate the chitinolytic ability of 9 isolate. The best isolate was the one which produced high chitinolytic activity and hydrolyzed chitin into NAG. 9 isolate were examined and BPPTCC-2 was the best isolate which produced highest level of chitinolytic activity 0.1780 U/ml. The chitinase was able to hydrolyzed chitin resulting high yields of NAG 11.5% as the only product

    Quinazolinone derivative compounds exhibit a broad spectrum of biological activity, including antioxidant. Quinazolinone derivative compound 4 - [(E) -2- (4-oxo-3-phenyl quinazolin-2-yl) ethenyl]-benzensulfonamida and its analogs (2a-f) have been synthesized and demonstrated its activity as a selective inhibitor of cyclooxygenase-2. The aim of this research was to evaluate the antioxidant activity of compound 4 - [(E) -2- (4-oxo-3-fenilkuinazolin-2-yl) ethenyl] -benzensulfonamida and its analogs with DPPH radical scavenging method, ferric reducing activity potential (FRAP), and phosphomolybdenum method. The experiment revealed that the % inhibition and antioxidant capacity of compound 2c, 2e and 2f were higher compared to compound 2a, 2b and 2d. The DPPH radical scavenging analysis found that the % inhibition of compound 2c, 2e and 2f at high concentration of compound solution (1000 µg/mL) were 4.8 %, 3.46 %, and 3.68 %, respectively. While the total antioxidant capacity of compound 2c, 2e and 2f were recorded 37.93, 33.4, and 46.3 µg ascorbic acid equivalence/mg of compound, respectively. However the antioxidant activity of all synthesized compounds (2a-f) were lower than the standard ascorbic acid

    DETERMINATION OF SIBUTRAMINE ADULTERATED IN HERBAL SLIMMING PRODUCTS USING TLC DENSITOMETRIC METHOD

    Determination of sibutramine adulterated in herbal slimming product using thin layer chromatography (TLC) densitometric method with TLC silica gel 60 F254 aluminium plate as stationary phase and mixture of toluen-diethylamine (10:0.3) as mobile phase has been developed. The calibration curve in the concentration range of 0.50 to 5.00 µg/spot showed good linier relationship (r2 = 0.9986). The limit of detection and quantitation (LOD and LOQ) were 217.5 ng and 724.9 ng/spot, respectively. The method gave satisfactory specificity, linierity, precision and accuracy validation criteria and was applied for determination of sibutramine in herbal slimming products obtained from several drugstrores in Depok City, West Java, Indonesia. Results of the determination showed that six of seven samples analyzed were detected containing sibutramine HCl with the concentration of 2.45 - 26.24 mg in a single dosage of slimming herbal product

    CYP2D6 Genotyping for Optimization of Tamoxifen Therapy in Indonesian Women with ER+ Breast Cancer.

    Background: Certain CYP2D6 genotypes are linked to a lower efficacy of tamoxifen therapy. This study aimed to observe CYP2D6 polymorphisms and examine the impact of CYP2D6 genotyping among tamoxifen-treated breast cancer patients in Indonesia. Methods: 150 breast cancer participants were recruited. Buccal swab samples were collected; gDNA was extracted and genotyped using the qPCR method. Blood samples were collected, and measurement of tamoxifen metabolite levels was performed using UPLC-MS/MS. Results: 43.3% (n = 65) of participants were IMs. *10 was the most common haplotype (n = 89, 29.7%), followed by *36 (n = 73, 29.7%), making *10/*36 the most common diplotype (n = 34, 22.7%) in this study. The difference in endoxifen levels between the NM and IM-PM groups at baseline was statistically significant (p ≤ 0.001). A dose increase in tamoxifen to 40 mg daily successfully increased endoxifen levels in IMs to a similar level with NMs at baseline (p > 0.05) without exposing IMs to serious side effects. No statistically significant differences were observed between the 20mg group and the 40 mg group on the adjusted OS (p > 0.05) and the adjusted PFS (p > 0.05). Conclusions: Our study observed a considerably high proportion of CYP2D6 IMs. The dose adjustment of tamoxifen was proven to significantly and safely improve the level of endoxifen and survival
