    Identificação molecular e caracterização morfológica doisolado de nematoide entomopatogênico Heterorhabditis sp. UENPn-01 obtido em área de pastagem no município de Ribeirão Claro, Paraná

    Nematoides são vermes pertencentes ao Filo Nematoda, que possuem hábitos diversificados,com grupos parasitas de plantas, parasitas humanos e há também os nematoides parasitas deinsetos, conhecidos como nematoides entomopatogênicos (NEPs). Os NEPs podem serencontrados em praticamente todo o mundo, sendo isolados de diferentes ecossistemas a partirde amostras de solo, ou de insetos infectados encontrados no campo, e no Brasil, existem aproximadamente 54 isolados registrados. No estado do Paraná, há registro de apenas umaespécie de nematoide entomopatogênico, Heterorhabditis amazonensis, demonstrando aimportância e a necessidade de conhecer melhor este grupo de entomopatógenos em nossoestado. Assim, o objetivo deste trabalho foi realizar a identificação e caracterização de umisolado de nematoide pertencente ao gênero Heterorhabditis sp., isolado no Município deRibeirão Claro, PR. A obtenção do isolado foi realizada através de coletas de solo, e utilizou-se a técnica de “iscas-vivas” com lagartas de Galleria mellonella (Lepidoptera: Pyralidae). Aidentificação molecular do isolado de NEP foi realizada a partir da amplificação da região ITSe também da região D2D3, bem como, por meio de medição de estruturas para identificaçãomorfológica. Realizaram-se também fotos de Microscopia Eletrônica de Varredura (MEV) paraanálise de estruturas morfológicas. Além disso, foi avaliada a patogenicidade sobre lagartas deG. mellonella e observação do ciclo de vida. No processo de identificação molecular a partir daregião ITS, observou-se similaridade de 98,64% com Heterorhabditis mexicana e 98,57% comHeterorhabditis taysearae, e na região D2D3 uma similaridade de 96,94% com H. mexicana.Com relação a identificação morfológica, observou-se que o isolado apresenta diferenças nasmedições quando comparado a H. mexicana, porém apresenta uma característica similar nosmachos no qual, o poro excretor é localizado posteriormente ao bulbo basal. Nas fotos de MEV,pode-se observar estruturas das fêmeas hermafroditas, fêmeas de segunda geração e machos,observando-se na região anterior, a presença das papilas labiais em todos os NEPs. Nas fêmeas,observa-se a vulva na região mediana do corpo, bem como a cauda e o ânus na região posterior.Nos machos, na parte posterior, nota-se a cauda e a espícula. No teste de patogenicidade, oisolado causou mortalidade de 88% de lagartas de G. mellonella, e quanto ao ciclo de vida oisolado UENPn-01 apresentou ciclo característico do gênero Heterorhabditis, com a primeirageração apenas de fêmeas hermafroditas, seguido de três gerações com a presença de machos efêmeas, totalizando quatro gerações completas dentro do hospedeiro.Nematodes are worms belonging to the Phylum Nematoda, which have diversified habits, withparasitic groups of plants, human parasites and also insect parasitic nematodes, known asentomopathogenic nematodes (NEPs). NEPs can be found practically all over the world, beingisolated from different ecosystems from soil samples, or from infected insects found in the field,and in Brazil, there are approximately 54 registered isolates. In the state of Paraná, there is onlyone species of entomopathogenic nematode, Heterorhabditis amazonensis, showing theimportance and the need to better understand this group of entomopathogens in our state. Thus,the objective of this work was to identify and characterize a nematode isolate belonging to thegenus Heterorhabditis sp., Isolated in the city of Ribeirão Claro, PR. The isolation was obtainedby collecting soil, and the technique of "live baits" with caterpillars of Galleria mellonella(Lepidoptera: Pyralidae) was used. The molecular identification of the NEP isolate was carriedout by amplifying the ITS region and also the D2D3 region, as well as by measuring structuresfor morphological identification. Scanning Electron Microscopy (SEM) photos were also takento analyze morphological structures. In addition, the pathogenicity of G. mellonella caterpillarsand observation of the life cycle were evaluated. In the molecular identification process fromthe ITS region, 98.64% similarity was observed with Heterorhabditis mexicana and 98.57%with Heterorhabditis taysearae, and in the D2D3 region, 96.94% similarity with H. mexicana.Regarding the morphological identification, it was observed that the isolate presents differencesin measurements when compared to H. mexicana, however it has a similar characteristic inmales in which the excretory pore is located posteriorly to the basal bulb. In the SEM photos,it is possible to observe structures of the hermaphroditic females, second generation femalesand males, observing the presence of the labial papillae in all NEPs in the anterior region. Infemales, the vulva is observed in the median region of the body, as well as the tail and anus inthe posterior region. In males, in the posterior part, the tail and spike can be noticed. In thepathogenicity test, the isolate caused 88% mortality of G. mellonella caterpillars, and as for thelife cycle, the UENPn-01 isolate presented a cycle characteristic of the genus Heterorhabditis,with the first generation of hermaphrodite females only, followed by three generations with thepresence of males and females, totaling four complete generations within the host.CAPE

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
