    Postnatal Stress in Mice: Does "Stressing" the Mother Have the Same Effect as "Stressing" the Pups?

    Short- and long-term effects of brief maternal separation, maternal exposure to novel male odor, and standard rearing were compared in NMRI mice. The first condition consisted of 15 min of daily exposure of pups to clean bedding (CB), and the second condition consisted of 15 min of mothers' exposure to the odor of strange males (SM), for 14 days after birth starting from postnatal Day 1. Thus, both conditions entailed the same period of maternal separation. A control mother-offspring group was left undisturbed (nonhandled, N-H). Corticosterone levels of mothers and pups were measured at the end of the last manipulation session. Corticosterone levels were higher in SM mothers, differing from both those of CB and of control dams; CB pups showed the highest corticosterone levels in comparison with the pups belonging to the other groups. Maternal behavior observed as furthest as possible from the daily separation session did not differ among the three groups. The behavioral response to 0.5 mg/kg of apomorphine in 15-day-old pups was enhanced in both CB and SM animals, which suggests an alteration of dopaminergic functioning. Finally, adult CB and SM male mice showed an increase in the percentage of time and entries into the open arms of the plus-maze in comparison to nonhandled males. This study indicates that exposure to ecologically relevant stimuli elicited a stress response in lactating dams. This "social stress " brings about short- and long-term effects in the offspring, even in the absence of any direct manipulation of the pups. © 2004 Wiley Periodicals, Inc

    A model of social stress in dominant mice: Effects on sociosexual behaviour

    The possibility of socially stressing the dominant/aggressive member of a pair of male mice is tested. Male mice (NMRI outbreed strain) were housed in pairs to assess dominant and subordinate roles by agonistic interactions and urine-marking test. Social stress for dominant males consisted in 30 min/day of exposure to their subordinate partner interacting with a female in the adjacent compartment of the cage, for 9 days. Results showed that dominance status was maintained. Behavioural observations indicated that neither the subordinates nor the dominant males habituated to this experimental procedure. At the end of the chronic stress, dominant animals were given the opportunity to interact for 30 min with a female in their compartment. Results indicated that stressed dominants showed impairment in their sexual behaviour and were more oriented towards the physical environment in comparison with control dominants. The behavioural response to apomorphine (0.25 mg/kg) indicated an alteration of the dopaminergic functioning in socially stressed dominant mice. This study suggests that the characteristics of the stressor and the effects of the chronic social stress could be different, according to male social status. © 2001 Elsevier Science Inc

    The role of the vgf gene and VGF-derived peptides in nutrition and metabolism

    Energy homeostasis is a complex physiological function coordinated at multiple levels. The issue of genetic regulation of nutrition and metabolism is attracting increasing interest and new energy homeostasis-regulatory genes are continuously identified. Among these genes, vgf is gaining increasing interest following two observations: (1) VGF-/- mice have a lean and hypermetabolic phenotype; (2) the first VGF-derived peptide involved in energy homeostasis, named TLQP-21, has been identified. The aim of this review will be to discuss the role of the vgf gene and VGF derived peptides in metabolic and nutritional functions. In particular we will: (1) provide a brief overview on the central systems regulating energy homeostasis and nutrition particularly focusing on the melanocortin system; (2) introduce the structure and molecular characteristic of vgf; (3) describe the phenotype of VGF deficient mice; (4) present recent data on the metabolic role of VGF-derived peptides, particularly focusing on one peptide named TLQP-21

    The number of male pups within a litter of NMRI mice is associated with the dam's food preferences late in pregnancy

    Pregnancy related differences in food preferences and food intake are well recognized in animals and humans.Changes in maternal hormonal profiles are supposed to be implicated in such a phenomenon. Since fetal sex hormones are excreted in their dams' bloodstream, they can affect the behaviour of mothers during gestation. We investigated whether there was a relationship between the number of male pups within the litter and food preferences in pregnant mice. In the first experiment we found that, parcelling out the effect of litter size, the number of male fetuses was positively correlated to maternal testosterone levels on pregnancy day (PD)18. In the second experiment we showed an association between the number of males and the consumption of a 0.25% saccharin solution on PD18, controlling for litter size and baseline (before pregnancy) saccharin intake. In the third experiment, mice on PD18 were placed in a test cage and underwent a 1-h binary choice test between two unknown unpalatable diets, a high fat diet (HFD) and a sweet mesh diet (HSD). Results revealed that the number of males was negatively associated with the preference for the HFD at birth. These results are consistent with the hypothesis that the behavioural profile of dams can be associated with fetal gender composition. © 2002 Elsevier Science Ltd. All rights reserved

    Aggression and anxiety in pregnant mice are modulated by offspring characteristics

    We investigated aggressiveness and anxiety during late pregnancy in mice, Mus domesticus in relation to the pups' characteristics. The female's aggressive behaviour towards a male intruder and behavioural anxiety were evaluated in separate experiments during late pregnancy (days 17-18) and individual scores were correlated with the number and sex of the delivered pups. We did a preliminary experiment to compare the behavioural measures of anxiety in the classical elevated plus-maze test and in a new experimental condition designed for pregnant females (three-compartment test choice: clean, own and unknown male's bedding): the time spent in the male's compartment by pregnant females was negatively correlated with the percentage of entries and time spent in the open arms of an elevated plus-maze. This allowed us to measure anxiety twice in the same female: in dioestrus and during pregnancy. Females' anxiety increased during late pregnancy and no significant correlation was observed between individual scores. Aggressive behaviour and anxiety were affected by the number of pups the pregnant female was carrying: the larger the litter, the more aggressive and more anxious the female was towards a male's cues. By contrast, the offspring fetal sex ratio did not affect the amount of aggressive behaviour and the level of anxiety shown by the mothers to be. © 2006 The Association for the Study of Animal Behaviour

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     Variations on the Author

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
