Characterization of the <i>Saccharomyces bayanus</i>-type AGT1 transporter of lager yeast
Transport of maltose and maltotriose into the yeast cell is thought to be rate‐limiting in the utilization of these sugars. The maltose and maltotriose transporters Malx1, Agt1, Mtt1 and Mphx are present in different combinations in brewer's yeast strains, conferring different maltose and maltotriose transport characteristics on the strains. A new putative maltose/maltotriose transporter ORF was identified during whole genome sequencing of the lager strain WS34/70 (Y. Nakao et al., DNA Res., 2009, 16, 115–129). Sequence comparisons suggested that this putative α‐glucoside transporter might be a Saccharomyces bayanus counterpart of the Agt1 (Saccharomyces cerevisiae type) transporter. In the present work, the transporter coded by a SbAGT1 gene from a lager strain, A15 (and with the same sequence as the corresponding gene in WS34/70) was characterized. It is shown that this SbAGT1 encodes a functional α‐glucoside transporter with a wide‐substrate range, including maltose and maltotriose. Trehalose, α‐methylglucoside and sucrose were inhibitors, suggesting they are also substrates. The SbAgt1 transporter had similar affinities for maltose and maltotriose (17 ± 7 and 22 ± 2 m m, respectively) and a higher Vmax for maltose than maltotriose (21 ± 7 and 12 ± 2 µmol min−1 g dry yeast−1, respectively)

Overstating the evidence - double counting in meta-analysis and related problems
Background: The problem of missing studies in meta-analysis has received much attention. Less attention has been paid to the more serious problem of double counting of evidence.
Methods: Various problems in overstating the precision of results from meta-analyses are described and illustrated with examples, including papers from leading medical journals. These problems include, but are not limited to, simple double-counting of the same studies, double counting of some aspects of the studies, inappropriate imputation of results, and assigning spurious precision to individual studies.
Results: Some suggestions are made as to how the quality and reliability of meta-analysis can be improved. It is proposed that the key to quality in meta-analysis lies in the results being transparent and checkable.
Conclusions: Existing quality check lists for meta-analysis do little to encourage an appropriate attitude to combining evidence and to statistical analysis. Journals and other relevant organisations should encourage authors to make data available and make methods explicit. They should also act promptly to withdraw meta-analyses when mistakes are found

Disodium chromoglycate particles were labelled with pure gamma-radiator, 99m Tc, using the co-precipitation technique based on spray drying. Radioactive drug particles were mixed, with lactose carrier and filled into hard gelatin capsules. Seven healthy volunteers inhaled drug doses using Spinhaler, I. S. F., Berotec, and Rotahaler dry powder devices. The fractional deposition of drug particles in the upper airways and lung region were monitored using a gamma camera. The fraction of the dose relained in the powder inhaler was the smallest for I. S. F. and especially for Berotec inhalers. These devices have narrower air channel constructions with a smaller wall surface area than the Spinhaler and Rotahaler devices. Thus the sticking of the drug particles onto the plastic walls was less probable for the first mentioned devices. The drug particles from all the dry powder inhalers seemed to be more able to follow the inspired air stream without depositing in the upper airways than previously documented for pressurized metered dose aerosols. I. S. F. and Berotec inhalers with narrow air channels gave the greatest lung deposition of the inhaled drug, particles. Thus the design of the dry powder inhaler was noticed to have a remark able effect both on the emptying of the capsules as well as on the redispersion of the powder mixture.
