    Background The carnitine/acylcarnitine carrier (CAC or CACT) mediates transport of acylcarnitines into mitochondria for the β-oxidation. CAC possesses Cys residues which respond to redox changes undergoing to SH/disulfide interconversion. Methods The effect of H2S has been investigated on the [3H]carnitine/carnitine antiport catalyzed by recombinant or native CAC reconstituted in proteoliposomes. Site-directed mutagenesis was employed for identifying Cys reacting with H2S. Results H2S led to transport inhibition, which was dependent on concentration, pH and time of incubation. Best inhibition with IC50 of 0.70 μM was observed at physiological pH after 30-60 min incubation. At longer times of incubation, inhibition was reversed. After oxidation of the carrier by O2, transport activity was rescued by H2S indicating that the inhibition/activation depends on the initial redox state of the protein. The observed effects were more efficient on the native rat liver transporter than on the recombinant protein. Only the protein containing both C136 and C155 responded to the reagent as the WT. While reduced responses were observed in the mutants containing C136 or C155. Multi-alignment of known mitochondrial carriers, highlighted that only the CAC possesses both Cys residues. This correlates well with the absence of effects of H2S on carriers which does not contain the Cys couple. Conclusions Altogether, these data demonstrate that H2S regulates the CAC by inhibiting or activating transport on the basis of the redox state of the protein. General significance CAC represents a specific target of H2S among mitochondrial carriers in agreement with the presence of a reactive Cys couple

    E-LIS: an international open archive towards building open digital libraries

    Established in 2003, E-LIS is an international open access archive related to librarianship, information science and technology, and related disciplines. It uses the Open Archives Initiative (OAI) protocol and tools to facilitate interoperability between repository servers. To date E-LIS is the biggest repository in library and information science and after only two years contains over 2200 papers. E-LIS is the first international e-server in this area, is part of the RCLIS (Research in Computing, Library and Information Science) project and is organised, managed and maintained by an international team of librarians working on a voluntary basis. This paper describes the main characteristics (technical and organizational) of the archive and its configuration and customization, and discusses its policies, aims and mission. Its main focus, however, is on the E-LIS organizational model and on the strategic issues correlated with Open Access (OA). It also delineates some of the challenges and opportunities consequent on a global vision for the Library and Information Science (LIS) field which envisages papers coming from all over the world and which gives E-LIS the impetus and motivation to stimulate participation in the venture and to further develop international research activities. Finally, this paper also emphasises that the promotion of E-LIS further enhances the OA movement in general, so E-LIS can be regarded as a tool for the dissemination of the OA philosophy

    La visite d’un érudit français sur le chantier des fouilles d’Herculanum

    Charles de Brosses (1709-1777) is the first French traveler who visits Herculaneum after the discovery of its ruins. He first announces to the French a world was found under the deep lava curtain dating back to 79 A.D. During his age an important cultural debate focuses on the credibility of Antique history. The French author reaches Naples during his long studies on Sallustio and in particular during his translation of the Latin historian. In fact his interests for Sallustio leads him to Rome as well as his love and curiosity for ancient arts invite him to visit Herculaneum. This is the main reason why he reaches Naples, a town he does not like. The present work tends to demonstrate how his visit makes de Brosses’s Grand Tour more interesting by associating his love for arts and culture with his curiosity for the region of Campania. It intends to show how the underworld descent reveals something new which has been hidden miraculously for centuries. De Brosses speaks about a singular event which can astonish and amuse as well. In fact his long detailed descriptions, often scientific, help his letters’ addressee, and in general every single reader, get a glimpse of what is going to be revealed. This work tends to analyze his letters which not only contain a list of archeological discoveries from the excavation of Herculaneum, but also they include some ancient inscriptions in the Oscan language which make them more appreciated. Thanks to this cultivated French traveler, considered one of the best known among the Grand Tour writers, France learns what is happening all around Naples. In 1750 he publishes Lettres sur l’état actuel de la ville d’Herculée, where he focuses, in an academic way, his great interest for archeology and what he will insert in his two letters consecrated to Herculaneum (Letters XXXII and XXXV taken from Lettres familières). Besides the present work tends to demonstrate how his letters renew the genre inaugurated by the previous travelers who have inspired him. Finally the research intends to discuss the singular relationship between de Brosses and the city of Herculaneum which does not arouse the interest for Antiquity yet which is said to be the reason why scholars in the 18th century re-discover antiquity

    The article points at the differences between music achievements and music aptitudes claiming that music achievements arę intellectual, while music aptitudes are spontaneous and occur in the entire body. The author also shows the importance of testing the music aptitude and presents tests useful for this purpose (e.g. PMMA, IMMA, MAP and AMMA). He says that music aptitude tests may be used to identify students with high overall music aptitude, but they may also be administered to focus on tonal and rhythm aptitudes

    This study aims to identify and explain the service of making Birth Certificates and to find out what obstacles are faced at the Population and Civil Registration Office of Kuantan Singingi Regency in serving the making of Birth Certificates. The theory used is the service theory by Zeithaml-Parasurman-Berry, then the method used in this research is a quantitative method with the type of research being survey research. In collecting the necessary data, it is done by using questionnaires, interviews, observation and documentation techniques. The respondents in this study were 10 employees and 20 people who were taking care of birth certificates. This study shows that in the implementation of Public Services at the Population and Civil Registration Office of Kuantan Singingi Regency for the management of Birth Certificates, the performance of employees is felt to be lacking and the facilities and infrastructure are inadequate. Suggestions from the author is to improve service to the community there must be a high level of discipline so that later it can create effective and efficient service delivery. So that service users will feel valued as users of Birth Certificate services at the Population and Civil Registration Office of Kuantan Singingi Regency

    Prose is one of the three literary kinds. Sub kinds of prose are history, description, biography, fiction, reflection, persuasion, censure and humor. The writer will take one sub-genre of prose only, which is fiction. Fiction is action, which primarily appeals to the imagination. This research aims to find the properties of the novel and the social life and comments described in the novel. The scope of the study is limited only a brief look and comments on the social life in “Adventure of Tom Sawyer by Mark Twain”. Data analysis was conducted with the  qualitative analysis. The results showed that there was seven elements, which are the biography of the author, the list of his writings, the summary of the adventure of Tom Sawyer, the theme, the plot, the characters, the setting and points of view. In answered the second questions about two aspects consist of; a) human beings has good behaviour, he will do positive activities in the social life such as human beings are not descrimination of racial in their environment. They have a good nature, the nature of love, patience, wisdom, kindness, simplicity, the nature of protection and responsibility and than have the potency to belief in God, b) human beings has bad behaviour, he will not do the positive or good activities in the social life, because there were some the economic gaps in their environment which make them have identical the nature of greed, selfish, arrogance, stinginess and ambitions in their life. Meanwhile, the social life in our environment can make us become mature and care with other people.     Electronic publishing implies a change in the traditional publishing chain, for such concepts as storage and distribution of the scholarly article, going as it does from the physical to the digital archive, and affecting all processes in between. We will deal with the new trends in electronic publishing of scholarly journals, from the many and varied projects and initiatives of Open Access and Free Access. Regarding the letter, we shall speak about Open Access directories and repositories ( Open Archive Initiative) available on the Internet. When we speak of electronic publishing of scholarly journals, same points need to be clarified: 1) journals born under the Open Access model, 2) journals that combine commercial distribution with a temporary embargo period before their free distribution over the Internet. We shall see that the advantages outnumber the drawbacks with this new publishing policy and it will be the researchers themselves who must adapt to the cultural changes involved in this new form of publishing their works. The articles concludes with an analysis of how publishers, information distributors and libraries have been obliged to modify their systems of publishing, distribution and management of information

    Bioactives from agricultural food by-products: application in food and health

    2015 - 2016Agriculture and agri-food industry produce a large amount of residues in non-edible portions from cultivation and processing of crops. These wastes, considered by-products, give serious environment damage, if not properly disposed, also because of their seasonal production. Until a few years ago the only goal was the disposal of these by-products. Recently the researchers discovered the characteristics of these wastes and their possible exploitation as sources of interesting compounds and energy for pharmaceutical, cosmetics and nutraceutical sectors. Fruit and vegetable by-products, in fact, are rich in bioactive compounds, such as carotenoids, dietary fibers, minerals and phenolic compounds, with correlated beneficial health effects. In this context the activity of the PhD project evaluated agricultural by-products as sources of bioactive compounds. The aims of the present PhD project were the development of innovative and selective analytical methodologies, for determination of bioactive compounds, using liquid chromatography methods (UHPLC) coupled to UV-Vis spectroscopy and mass spectrometry (high resolution, HRMS and tandem mass, MS/MS), the improvement of new extraction techniques, which can optimize extraction yields, minimize costs and environmental impact, such as pressurized hot water extraction (PHWE), ultrasound assisted extraction (UAE) and supercritical antisolvent fractionation (SAF), and finally the study of antioxidant activity of obtained extracts, using chemical and biological assays (DPPH, ABTS, ORAC, CAA). The analysis of bioactive compounds in plant extracts through advanced sample preparation techniques and modern separation tools allows a comprehensive study of the matrix. The project, very relevant, was to provide innovative methodologies, procedures and final products for different companies of healthcare area, in terms of innovation, costs and profits. The project was divided into three major areas, corresponding to the three selected by-products. The determination of chemical profile and the development and optimization of innovative and green extraction procedure for recovery of bioactive compounds have been carried out for each matrix. Selected by-products were: the main artichoke by-products, bracts and leaves, derived from cultivation and industrial processing, hazelnut by-products, roasted skins, derived from kernel industrial processing, and aromatic plants by-products, distillation waste waters, derived from essential oils production. All by-products studied have proved to be low cost sources of bioactive compounds with antioxidant activity. Furthermore, among innovative extraction techniques, PHWE procedures have allowed obtaining final extracts selectively rich in compounds of interest without formation of artefacts, and compared to existing extraction methods were simple, fast, environmentally friendly and fully automated. Moreover my PhD project provided new analytical methods to standardize vegetable extracts. The most relevant results showed that discarded raw materials are suitable ingredients for the production of formulated health products and nutraceuticals. [edited by author]XV n.s. (XXIX

    Selected factors of home environment and their importance in the development of musical abilities of early level school children

    The article contains the analysis of rich empirical material connected with the diagnosis of home environment in the aspect of factors conditioning the level of children musical abilities. The factors include, among others, general and music education of parents and various forms of musical interests shown in different forms of activities connected with playing a musical instrument, singing, dancing and listening to music. In the research the author used a questionnaire aimed at parents and a musical abilities test Intermediate Measures of Music Audiation (IMMA) by E.E. Gordon on the basis of which the level of children musical abilities was defined.Artykuł zawiera analizę bogatego materiału empirycznego związanego z diagnozą środowiska rodzinnego w aspekcie czynników mogących warunkować poziom zdolności muzycznych dzieci.
