    Abstract 2847: Loss of heterozygosity (LOH) within the hypoxia inducible factor 3A (<i>HIF3A</i>) locus in liver metastasis (LM) from primary colorectal cancer (CRC)

    Abstract Background and Aims: HIF3α is a hypoxia-regulated transcription factor that represses the effects of HIF1α and HIF2α under hypoxia at transcriptional and posttranslational levels. We previously found that LOH at the HIF3A locus is frequent in LM (59%) and the expression of HIF3A protein is reduced in LM compared to primary CRC. At present, the smallest overlapping LOH (SOL-LOH) region has been mapped to the intron 8 of the HIF3A, spanning from 46,312,911 to 46,315,651 on chromosome 19 in LMs. This genomic region contains multiple FOXA1 binding sites embedded in the two DNase I hypersensitive sites (UCSC Genome Browser, https://genome.UCSC.edu). About 30% of the human population carries a deletion within our SOL-LOH region (1000 Genomes Consortium: esv3644527, Chr19: 46,314,109-46,315,478). In this study, we determined whether esv3644527 deletion affects on the expressions of HIF3A, HIF2A, HIF1A and their target genes under normoxic and hypoxic conditions. Methods: For LOH mapping, 75 resected LMs derived from primary CRC were analyzed for LOH using 8 polymorphic markers. We obtained three EBV-transformed B cell lines with heterozygous esv3644527 deletion and the one with the homozygous deletion from the Coriell Institute. The expression levels of 23 genes related to HIF pathways were determined by qRT-PCR. Results and Discussion: The expression levels of HIF3A mRNA were too low to compare among these B cell lines by qRT-PCR. However, the expression levels of HIF2A and its target gene, angiopoietin 2 (ANGPT2), are significantly elevated in B cells with a homozygous deletion compared to B cells with a heterozygous deletion under both normoxic and hypoxic conditions. These observations are compatible with the results obtained from a previous study where pulmonary endothelial cells from HIF3A-knockout mice expressed elevated levels of HIF2A and ANGPT2 (Kobayashi-S et al., Genes Cells, 2015, 20:224-241), suggesting that deletion of the FOXA1 sites present in this region may regulate the HIF3A expression. Conclusions: Our results show that about 60% of LMs exhibit LOH in the 2.7 Kb region surrounding intron 8 of the HIF3A locus. Deletion within this region may lead to upregulation of HIF2A and ANGPT2 by negatively regulating HIF3A expression.     The role of aquaporin-4 in the blood-brain barrier development and integrity: Studies in animal and cell culture models

    Aquaporin-4 (AQP4) is the major water channel expressed in brain perivascular astrocyte processes. Although the role of AQP4 in brain edema has been extensively investigated, little information exists regarding its functional role at the blood-brain barrier (BBB). The purpose of this work is to integrate previous and recent data regarding AQP4 expression during BBB formation and depending on BBB integrity, using several experimental models. Results from studies on the chick optic tectum, a well-established model of BBB development, and the effect of lipopolysaccharide on the BBB integrity and on perivascular AQP4 expression have been analyzed and discussed. Moreover, data on the BBB structure and AQP4 expression in murine models of Duchenne muscular dystrophy are reviewed. In particular, published results obtained from mdX(3cv) mice have been analyzed together with new data obtained from mdx mice in which all the dystrophin isoforms including DP71 are strongly reduced. Finally, the role of the endothelial component on AQP4 cellular expression and distribution has been investigated using rat primary astrocytes and brain capillary endothelial cell co-cultures as an in vitro model of BBB. The author uses a qualitative method which is explained graphically, namely the market balance of the IS-LM model where the focus is on money and goods markets associated with macroeconomics where researchers take the side of investors. The results of this study are that the balance in the economy is the point where the IS and LM curves intersect. This point provides an interest rate (r) and income level (Y) that satisfies the equilibrium conditions that occur in the goods market and money market. In other words, planned spending equals actual spending, and the demand for real money balances equals the supply. So that the IS-LM balance, it is stated that IS=LM

    Pectin As a Rheology Modifier: Recent Reports on Its Origin, Structure, Commercial Production and Gelling Mechanism

    Pectins (also known by the singular pectin) are a diverse family of biopolymers with a complex range of structures. Their common feature is a polysaccharide backbone of 1,4-linked α-d-galacturonic acids. Pectins are derived from dicotyledonous and some monocotyledonous plants and make up one third of the cell wall materials. Pectin has been widely used as a gelling and stabilizing agent in food, as an incipient ingredient in pharmaceuticals, in personal care products and in other polymer products. It is recognized as safe (GRAS) by the United States Food and Drug Administration (FDA). Commercial pectin is extracted using acids from by-products of the food industry such as citrus peel, apple pomace and sugar beet pulp. There are two types of pectin: high methoxyl (HM) pectin with a degree of methylation (DM) > 50% and low methoxyl (LM) pectin. LM pectins are usually manufactured from HM pectins by de-esterification. HM pectins gel by cross-linking homogalacturonan residues through hydrogen bonds and hydrophobic forces between the methoxyl groups, assisted by a high sugar concentration and low pH. In contrast, LM pectins gel by forming ionic linkages via calcium bridges between two carboxyl groups from two different chains in close proximity, known as the ‘egg-box’ model. The viscoelastic behavior of both gels depends on intrinsic and extrinsic factors.     This paper discusses the chaotic pseudo-random number generator (CPRNG), which is used in block cipher mode of operation called PM-DC-LM. PM-DC-LM is one of possible subversions of general PM mode. In this paper is not discussed the design of PM-DC-LM, but only CPRNG as a part of it because designing is written in other papers.     CF/LM-PAEK: Characterisation and sensitivity to critical process parameters for automated fibre placement

    This paper presents an investigation into Automated Fibre Placement process parameters for a novel carbon-fibre-reinforced thermoplastic material based on a low-melt polyaryl ether ketone polymer matrix (CF/LM-PAEK). The primary focus of this investigation is in-situ consolidation, with parameter sets for sample manufacturing selected based on a Design of Experiment method and analysed by means of mechanical and thermal testing. For in-situ consolidation, a maximum shear strength of 33.6 MPa and a crystallinity of 27.3% is achieved. Subsequent tempering increases shear strength up to 46.5 MPa and crystallinity up to 32%. The material was found to be insensitive to layup speed within the tested parameter range (1–15 m/min), achieving similar results for high layup speeds

Tenaga listrik dimanfaatkan untuk menghidupkan atau menjalankan alat-alat elektronika yang mempermudah aktivitas manusia. Agar dapat menggunakan tenaga listrik pada alat-alat elektronik diperlukan pengubah daya dari arus listrik arus kuat ke arus listrik arus lemah. Konsep pengubahan arus kuat menjadi arus lemah diterapkan dalam bentuk adaptor. Dalam pembelajarannya adaptor memanfaatkan konsep dioda sebagai penyearah arus listrik. Pada program studi Pendidikan Teknik Elektronika, khususnya di Universitas Negeri Jakarta, penerapan konsep dioda sebagai penyearah dituangkan dalam pembuatan adaptor DC oleh mahasiswa. Menggunakan IC LM 7805, penulis membuat adaptor DC dengan keluaran 5V sebagai penerapan konsep dioda sebagai penyarah. Hasil dari perancangan adaptor menunjukkan hasil pengukuran output adaptor bernilai 5V. Hal ini berbanding lurus dengan sifat IC LM 7805 yang meregulasi tegangan output yang masuk ke IC menjadi 5V. Electrical power is used to turn on or run electronic devices that make human activities easier. In order to use electrical power in electronic devices, it is necessary to convert the power from strong current to weak current. The concept of converting strong current to weak current is implemented in the form of an adapter. In the lesson, the adapter utilizes the concept of a diode as an electric current rectifier. In the Electronics Engineering Education study program, especially at Jakarta State University, the application of the concept of diodes as rectifiers is outlined in the manufacture of DC adapters by students. Using IC LM 7805, the author made a DC adapter with 5V output as an application of the diode concept as a rectifier. The results of the adapter design show that the adapter output measurement results are 5V.
