TaqMan qPCR for Quantification of Clonostachys rosea Used as a Biological Control Agent Against Fusarium graminearum
Clonostachys rosea is a biological control agent against Fusarium graminearum in small grain cereals and maize. Infections with F. graminearum do not only reduce the yield but, due to the production of mycotoxins, also affect the entire value chain of food and feed. In addition, production of other secondary metabolites such as hydrophobins, also known as gushing inducers, may cause quality challenges for the malting and brewing industry. Sustainable disease control strategies using C. rosea are treatment of infected residues of the previous crop, direct treatment of the actual cereal crop or post-harvest treatment during malting processes. Follow-up of growth and survival of biocontrol organisms during these different stages is of crucial importance. In the current study, we developed a quantitative real-time PCR detection method that amends the currently available culture-dependent techniques by using TaqMan chemistry with a highly specific primer and probe set, targeting the actin gene. We established a sensitive assay that detects the biological control agent down to 100 genome copies per reaction, with PCR efficiencies between 90 and 100%. The specificity of the assay was confirmed against a panel of 30 fungal and 3 bacterial species including 12 members of the Fusarium head blight complex and DNA of barley, maize and wheat. The DNA of C. rosea was detected in Fusarium-infected maize crop residues that were either treated in the laboratory or in the field with C. rosea and followed its DNA throughout the barley malting process to estimate its growth during grain germination. We used a standardized DNA extraction protocol and showed that C. rosea can be quantified in different sample matrices. This method will enable the monitoring of C. rosea during experiments studying the biological control of F. graminearum on cereal crop residues and on cereal grains and will thus contribute to the development of a new disease control strategy

Metamodern sensibility in Jenni Fagan's 'The Waken'
Jenni Fagan is a critically acclaimed author whose works explore marginalized girlhood in Scottish cultural contexts. "The Waken" depicts the story of a young girl brought up by a violent father on a Hebridean island in which a Stevensonian representation of evil acquires a feminist flavour. Through allusions to Stevenson's Gothic, Fagan draws on the antisyzygy element in Scottish literature but instead of reproducing an idea of duelling polarities within one entity, she uses binary oppositions in a polysyzygiacal way. This article shows that multiple alignments and plural connections of Fagan's polysyzygy indicate her text's participation in a metamodern sensibility. Creating a metamodern simultaneity in which a feminist deconstruction of patriarchal narratives coexists with the employment of the reconstructive potential of myth, Fagan's narrative presents a more dialogical approach to male literary traditions than the works of postmodern feminists.1Jenni Fagan is a critically acclaimed author whose works explore marginalized girlhood in Scottish cultural contexts. Jenni Brandon: Colorist Landscape Composer Three Selected Works for Oboe
This project examines three contrasting works, featuring the oboe by award-
winning American composer, Jenni Brandon. A champion of writing new music for the
oboe, Brandon has composed thirteen works from 2003 to 2016, ranging from
unaccompanied solo works to woodwind chamber ensemble.
Brandon is highly influenced by her surrounding landscapes and paints vivid
musical pictures. Apart from being emotionally impactful, Brandon’s works are enriched
with color, lyricism, and technical demand. This document focuses on three selected oboe
works by Brandon: On Holt Avenue (2006), In the City at Night (2008), and The Sequoia
Trio (2008). It presents a biographical sketch of the composer, surveys of the works,
compositional process and provide performance practice considerations. It is hoped that
this research will continue to increase awareness of the composer, provide performers a
better understanding on approaching the selected work, while promoting individual

Tässä työssä tarkastellaan kunnan väestön demografisia muuttujia sekä kartoitetaan kirjaston palvelut ja käyttäjät. Tutkimuksessa kirjaston kohderyhmänä tarkastellaan kaikkia kuntalaisia. Kirjaston palveluita sekä olemassa olevia asiakkaita tarkastellaan kunnan väestöön verraten. Opinnäytetyön on tilannut Oulunsalon kirjasto.
Tutkimuksessa on käytetty valmiita tilastoja sekä dokumentteja kunnasta ja kirjastosta. Tilastokeskus on suurin aineiston lähde Oulunsalon kunnan ohella. Lisäksi menetelmänä käytettiin teemahaastattelua. Haastateltavina olivat Oulunsalon kirjaston työntekijät.
Työn viitekehyksessä käsitellään kuntalaista kirjaston asiakkaana sekä kirjastonkäyttöä ja sen tutkimista. Oulunsalon historiaa tarkastellaan hieman ja uuden Oulun kirjastoverkko esitellään. Kulttuuriympäristön selvityksessä käydään läpi kunnan aktiivista harraste-, järjestö- ja kulttuuritoimintaa.
Väestöselvityksessä saatuja tietoja pyritään havainnollistamaan graafisin esityksin. Palvelu- ja käyttäjäryhmäselvityksen sekä aiempien yleisistä kirjastoista tehtyjen tutkimusten perusteella Oulunsalon kirjasto on piirteiltään tyypillinen suomalainen kunnankirjasto.ABSTRACT

In this thesis I examine demographical variables of local population and survey the library’s services and users. Every habitant of Oulunsalo is considered as library’s target group. Library services and existing customers are assimilated on local population. Thesis is made for the library of Oulunsalo.
As research material is used statistics and documents. Majority of the material is produced by Statistics Finland and Oulunsalo municipal. Thesis contains also theme interview where the workers of the library is interviewed.
I review citizen as library user, library use and library research in the framework. I examine the history of Oulunsalo and its library briefly. Library services of new Oulu are also reviewed. In survey of culture environment I look in to lively organization and culture activities of Oulunsalo.
In report of population structure information is clarified with graphics. Library of Oulunsalo didn’t depart from other public libraries in the service and customer group survey, if compared on other research made from public libraries
What gives? How companies invest in communities
Against a backdrop of growing public interest in corporate citizenship and responsibility, this report investigates the approach that leading firms are taking to investing in communities. It looks at how, and how well, companies are performing against leading indicators for community investment.
Significantly, it shows there is a lack of guidance about what constitutes good practice in community investment. Most companies undertake their community investment activities without a strong framework, strategy or tools to measure the performance, impact or effectiveness of their approach. In many cases, this is because tools and frameworks are poorly developed and applied in the Australian context. This has implications for the effectiveness of the programs companies support and, ultimately, may prevent programs reaching their full potential for communities.
The report presents some valuable insights for corporations and communities to inform better practice, reporting and outcomes in this important area.

Tämä opinnäytetyö on tuotettu Kainuun hyvinvointialueen aikuisten mielenterveyspalveluiden ja riippuvuuksienhoidon avohoitoyksikköön Kajaanin ryhmätalolle. Opinnäytetyöllemme oli työelämälähtöinen tarve. Kajaanin ryhmätalolla työskennellään asiakaslähtöisesti ja kuntouttavan työotteen mukaisesti. Sairaanhoitajana työskentelevä tiimivastaava kuvaili ongelmana olevan tämän työtavan näkyväksi saattaminen. Käytössä oleva materiaali kuntouttavasta työotteesta oli aiheen laajuuteen nähden kovin suppea, eikä käytössä olevassa materiaalissa tuoda esiin menetelmiä työn tueksi. Kuntouttavaa työotetta oli vaikeaa konkretisoida ja tueksi tarvittiin materiaalia, johon työntekijä voisi tarvittaessa palata jälkeenkin päin.
Opinnäytetyömme avaa kirjalliseen muotoon asiakaslähtöisen kuntouttavan työotteen perusajatusta mielenterveystyön näkökulmasta. Lisäksi tuomme esille menetelmiä työn tueksi ryhmätalon henkilö-kunnan käyttöön. Opinnäytetyömme palvelee myös uuden työntekijän perehdyttämisen tukimateriaalina. Menetelminä opinnäytteeseen on avattu Spiral-menetelmä ja Elämäntilanteen selvittämisen ympyrä, ESY. Opinnäytteeseen valikoituneet menetelmät eivät olleet vielä käytössä Kajaanin ryhmätalolla.
Opinnäytetyömme tuotoksena syntyi infograafi, jonka tarkoituksena oli avata asiakaslähtöistä kuntouttavaa työotetta ja kuvata sen toteuttamista kuntoutusprosessin vaiheissa. Halusimme sisällyttää infograafiin teoriaosuuden käsitteistä tiiviissä muodossa, sekä esitellä valitsemamme konkreettiset menetelmät työn toteutukseen.Abstract

This thesis was commissioned by Ryhmätalo, an adult mental health outpatient unit located in Kajaani, which is part of Kainuu Wellbeing Services County. This thesis is based on the needs of working life. Work at Ryhmätalo is client- and rehabilitee-oriented. The team leader, a nurse by education, described that the problem was making the rehabilitative approach visible. The currently used rehabilitative material was very narrow compared to the scope of the issue and did not include methods that supported the approach. According to the team leader, it was difficult to concretize the rehabilitative approach. New material to support the approach was needed.
This thesis discusses the basic idea of client-oriented rehabilitative approach from the perspective of mental health care. In addition, it presents methods to support the work of the staff at Ryhmätalo. This thesis can also be used in the orientation of a new employee. Methods described in this thesis include the Spiral method and the circle of life situation exploration, ESY. These methods were not yet used at Ryhmätalo.
This thesis resulted in an infographic, the purpose of which was to open up a client-oriented rehabilitative approach and describe its implementation in the phases of the rehabilitation process. The infographic also includes a theoretical part of the key concepts in a compact form and presents selected methods for the implementation of the work

Microstructural Evolution and Physical Properties of Polymer-Modified Mortars
Polymer-modified cementitious materials provide the base for building materials commonly used on modern
construction sites. By adding polymers, the properties of cementitious materials can be extended to suit a variety
of applications. With respect to adhesion properties, for example, the first patent of latex-modified hydraulic
cement systems was issued in 1924 (Lefebure 1924). In the field of tile adhesives, latex-modification allowed thinbed
application, a technique that is still standard because of its economic advantages with respect to application
time and resource costs. Before invention of redispersible powders, the appropriate latex was only available in the
form of dispersions. Mortar mixing was an important issue, which when improperly performed, often caused cases
of damage at construction sites. The development of latex in the form of redispersible powders drastically reduced
this problem, because it allowed the production of one-component systems or so-called “dry mortars”, which only
require the appropriate amount of water to be added before application.
Mortar properties were continuously improved by optimising the formulation or enhancing the system's
components. Empirical approaches dominated, in which numerous formulations were compared with each other,
in terms of physical properties of the resultant mortars. To further improve these properties at the present stage, an
extended understanding of the mechanisms active during mortar evolution is required. Many of these mechanisms
leave characteristic marks on the mortar microstructures, which, once recognised and related to the corresponding
mechanism, can be linked with the physical properties. Therefore, the microstructure represents a major key to an
improved understanding of the highly complex system of polymer-modified mortars.
The cementitious, mineralic microstructures can be investigated by methods commonly applied in earth and
material sciences. In contrast, organic compounds like polymers can form delicate and fragile structures requiring
specific techniques that originated in the field of organic chemistry and biology. Therefore, the investigation of tile
adhesive requires an interdisciplinary approach, in which methods from different fields of research are adapted and
combined.
As is common in applied research, the investigation of polymer-modified mortars is a tightrope walk between the
complex, commercial system and model systems usually based on crude simplifications. The combination of both
approaches might result in large forward steps in understanding, and new insights.
The present study on polymer-modified, cementitious mortars tries to incorporate the previously mentioned
requirements and is organised in the following manner: (a) methods of quantitative investigations, (b) influence of
polymers on microstructure and physical properties, (c) changes of microstructures and physical properties during
wet storage.
a) The first chapter describes the new methods developed to quantitatively investigate microstructures in
polymer-modified mortars. A combination of digital light, fluorescence and electron microscopy allowed the
visualisation of different mortar components such as specific polymer components, air voids, cement phases,
and filler minerals. In a second step, their occurrence and spatial distribution was quantified by image
analysis requiring appropriate program routines, whose use and functionality is explained. To demonstrate the
power of the new quantitative approach in the field of polymer-modified tile adhesives, a selected mortar
formulation was analysed as an example. The results show that the mortar fractionated during application and
hardening, inducing a variety of phase enrichments or depletions. The occurrence of these microstructural
heterogeneities suggests the major influence that the microstructure has on the physical properties of the
mortar system.
b) In the second chapter, the microstructural evolution of the mortar and the mechanisms involved were
investigated by using the methodology developed above. It is shown that water flux, induced by evaporation

and capillary forces of the porous substrate, played the most important role in mortar fractionation. It
transported cellulose ether, polyvinyl alcohol, and cement ions to the mortar interfaces, where they became
accumulated. In contrast, latex components did not migrate and remained homogeneously distributed within
the microstructure. Combination of quantitative with qualitative investigations allowed a reconstruction of the
mechanisms forming the microstructure during the different mortar stages. By correlating microstructural
observations with physical properties (e.g., adhesive strength), skinning on the mortar surface of the applied
fresh paste was found to decrease adhesion strength to the tile. As a consequence, it is the mortar-tile interface
that dominates the properties of the entire hardened substrate-mortar-tile system.
c) In chapter three the influence of wet storage on the microstructure and its physical properties are investigated.
Wet storage represents an important test criteria on the durability of polymer-modified systems exposed to
wet conditions in case of outdoor or bath room applications. Tests on individual polymer structures revealed
that cellulose ether and polyvinyl alcohol redissolved in the pore water, whereas latices were water-resistant.
Consequently, latex distributions in the mortar measured before and after wet storage were identical because
latex remained immobile, but cellulose ether and polyvinyl alcohol distributions changed. By combining
these observations with microstructural investigations of the failure surfaces, pore size, shrinkage and
physical test data, we were able to show that changes in the mortar volume and reinitiated cement hydration
caused a decrease of the mechanical properties during wet storage. Although they remained immobile, the
latex films also weakened due to water uptake and swelling, which was shown to be a reversible mechanism.
