Tracing time through cuticular clues: the role of rearing conditions and generational divergence in Lucilia sericata (Meigen, 1826) (Diptera: Calliphoridae)

    No full text
The chemical profiles of the cuticle of adult flies are highly influenced by environmental factors and generational variation, although the extent and mechanisms of these influences are still poorly understood. This research rigorously investigates the influence of rearing environment and generational changes on the cuticular hydrocarbon (CHC) profiles of adult flies collected from the natural environment in Swindon (UK), in June 2019. Gas chromatography-mass spectrometry (GC-MS) was used to analyze the hydrocarbon profiles. Then, chemometric analysis was applied to determine the chemical variation patterns, allowing the samples to be classified according to their chemical fingerprints. Significant differences in hydrocarbon composition were found between laboratory-maintained and field-collected specimens, underscoring the impact of environmental conditions on CHC expression. Additionally, gradual modifications in hydrocarbon content were detected across generations raised in the controlled environment, suggesting the involvement of adaptive physiological or epigenetic mechanisms. These findings contribute valuable insights into cuticle plasticity, highlighting its relevance in forensic entomology, chemical ecology, and insect evolutionary biology. The implications also extend to forensic investigations, where cuticular hydrocarbon profiles (CHCs) demonstrate potential for enhancing postmortem interval (PMI) estimation accuracy and species identification in criminal cases. By demonstrating quantifiable differences in CHC composition across rearing conditions and generations (AUC values ≥0.92 for all comparisons), this study provides a foundation for the broader application of chemical markers in forensic investigations.First author (Dr. Canan Kula) received sponsorship from the Ministry of National Education of the Republic of Türkiye.Turkish Journal of Entomolog

© Cranfield University 2022. All rights reserved. No part of this publication may be reproduced without the written permission of the copyright ownerCalliphoridae (blowflies) are important for forensic entomology as they are often the first group of insects to inhabit decomposing remains. To estimate the minimum post-mortem interval (PMImin), blowfly samples are collected from the crime scene and identified, and then the age needs to be determined for each species. Accurate species identification is crucial, as varied species have different life-stage timings. Previous studies on Diptera have shown the possibility of using cuticular hydrocarbon (CHC) analysis to aid species identification, ageing and ultimately calculating PMImin. However, since prior research was conducted at constant temperatures, the effect of fluctuating temperature on chemical profiles and the stability of CHCs in the field remains unclear. This thesis aims to examine the CHC profiles of Lucilia sericata and Calliphora vicina reared outdoors in the United Kingdom, Germany and Turkey in terms of species, age and inter-population differences. The first study investigates whether the stability of the CHC profiles of two colonies reared outdoors can be used to identify species-specific characteristics. The second examines the effects of fluctuating and constant temperatures on the CHC profiles of the life cycle stages and the use of their chemical profiles for age estimations. The final study aims to determine the differences in CHC profiles of the same species living in different habitats and whether they can be used to distinguish populations. The CHC profiles of the samples were extracted and analysed using Gas Chromatography-Mass Spectrometry (GC-MS). The statistical interpretation was carried out using Principal Component Analysis (PCA). The results showed that CHC profiles differed significantly between developmental stages by species, age, rearing temperature and geographical location. This study highlights that despite environmental factors affecting the stability of CHC compositions, the CHC analysis can be used as a complementary tool for identifying and ageing the species. Furthermore, the effects of ambient temperature and geographical variation on CHC profiles may be useful for determining inter-population variations.Ph

It can be seen that prison theme is mostly used by the authors who are sentenced to short or long term punishments for political reasons in Turkish literature. Orhan Kemal, Kemal Tahir, Kerim Korcan, Çetin Altan, Nihal Atsız, Erdal Öz, Rıfat Ilgaz, Abdülkadir Billurcu, Remzi Çayır, Sevgi Soysal, Feride Çiçekoğlu are some examples of the Turkish authors who carried their experiences in this harsh environment to their literary works. Füruzan and Canan Tan differ from other writers who used prison theme in their novels by not knowing this place by a consequence of legal punishment. In her novel 47’liler, Füruzan tells the story of the women who lives in this place which she herself did not live. Yet, she scrutinizes the relationship between woman and prison in 12 Mart era instead of focusing to prison. One of the bestselling writers of our time and the author of novel Kelepçe which is subject of our study, Canan Tan, set foot in prison not as a prisoner but as an observer with the purpose of gathering material for her novel. In this paper, the novel Kelepçe, which is written by Canan Tan and published in April 2016, will be reviewed and its portrayal of psychological – sociological background of crime will be examined. Kelepçe presents a view to the life of the women inmates, and also narrates the process which ends up with stigmatizing of them in public opinion

Forensic entomology has become increasingly important in the last decade. In cases of homicide, suicide or suspicious death, insect specimens are taken into consideration as acceptable evidence like blood, fingerprint or any other biological materials. When solving criminal cases, it is essential to determine the time since death or post-mortem interval (PMI). Entomological evidence can provide valuable information about the prediction of the post-mortem interval and where the death occurred.Forensic entomology, like many other fields in forensic sciences, has been developing using new technologies. Cuticular hydrocarbon (CHC) analysis has become a useful tool for identifying and ageing forensically important blowfly species. Variation in hydrocarbon profiles is significant for climatic adaptation, protection against dehydration and desiccation. The effect of other variables, such as environmental climate and geographic region, and how this may change the CHC structure is not known. In the literature, there are some studies showing differences in the hydrocarbon structure of beetles, fruit flies and house flies when reared at different temperatures. Although there are many studies on the relationship between temperature effect and development time of blowfly species using classical entomology methods, no studies have been conducted using cuticular hydrocarbon analysis. In this regard, this study has been a first in the field of forensic entomology. In this study, L. sericata collected from the wild were used for oviposition. Once eggs were laid on the oviposition medium, the Petri dish containing the meat was transferred to incubators at 14 ºC, 25 ºC and 34ºC. The cuticular hydrocarbons were chemically extracted and analysed for post-feeding larvae. The effect of temperature changes on the cuticular structure was investigated by using Gas Chromatography-Mass Spectrometry.Canan Kula is supported by the Turkish Ministry of National Education for her PhD research

Literary works, depending on the content of individual or social issues can reach a wider audience. In a novel, poetry or essay, the matter in hand can change the prejudices and existing perceptions by creating awareness in the reader. In the same way, it can help the reader to obtain prior knowledge in a subject that he/she has no idea about before. The literary works referring to social problems rather than individual issues are also a sociological document. Issues that are subject to works can either directly or indirectly affect a significant part of society. In this context, novels with broad expression have covered many issues ranging from children's rights to exile in both Turkish literature and world literature. Canan Tan, with her novels published in the 2000s and after, puts some ongoing social problems on her agenda. Various issues are addressed in the novels written by the author, employing her own experiences and observations. Piraye and Pembe and Yusuf draw attention as the novels in which elements of morals were intensely discussed. In this study, it is examined how the ceremonies were performed in Canan Tan's Piraye and Pembe and Yusuf novels and the concerns of the people who are effective in these practices. Negative examples are emphasized more and how some of these morals caused the people to suffer is explained by means of the texts

Determination of the Objective Reason Condition in the Conclusion and Renewal of Fixed Term Employment Contracts

    No full text
An employment contract may be executed for a fixed or indefinite term. An indefinite-term employment contract is the rule of thumb, whereas a fixed-term contract is an exception. According to Article 11 of the Employment Code No 4857, a fixed-term contract may be signed when it is based on an objective reason, such as fixed-term work, the completion of a certain job, or the emergence of a relevant fact. There is no similar provision in other employment codes, which is why concluding a fixed-term employment contract with a worker covered by the Employment Code is more difficult than terminating workers excluded from the said code. This strict structure to conclude a fixed-term employment contract of the Employment Code has been criticized by the doctrine. According to Article 11, paragraphs 2 and 3, of the Employment Code and Article 430 of the Obligation Code No. 6098, to renew a fixed-term employment contract with all workers, an objective reason is required. This article explains what objective reason means and if such a condition is suitable and evaluates the debates in the doctrine and legal studies regarding the objective reason condition in the conclusion of fixed-term employment contracts, regulated in Article 11 of the Employment Code. Further, the author discusses whether an exception should be introduced into the objective cause condition. This article next examines the objective reason condition for the renewal of fixed-term employment contracts. In this framework, in addition to the objective reason condition, the application of a time limit and a number limit will be evaluated to prevent abuse in the renewal of employment contracts by using successive fixed-term employment agreements
