    Comparison of a Formulated Transport Medium and EDTA Anticoagulant in Kenyan Field Isolates from Maseno Division against a Panel of Antimalarials Using SYBR Green I technique

    In vitro drug sensitivity relies on the growth of plasmodium in the presence of the antimalarials. These fresh isolates brought in for assay from collection sites normally not near the laboratory require a medium to keep the parasites viable in order to produce results which are a representation of the field situation in terms of drug susceptibility. This study validated the use of a formulated transport medium (TM) by assaying 322 Plasmodium falciparum field isolates from patients visiting Chulaimbo sub county hospital in Maseno division, for 50% inhibitory concentrations (IC50) against four antimalarials using SYBR Green 1 in vitro assay. These samples were transported using a formulated transport medium (TM) and the conventional EDTA anticoagulant tubes to act as control. The similarity which was detected with no significant difference (P >0.05) observed in the medians inhibitory concentration 50 (IC50) between samples in the EDTA anticoagulant and TM warrants its continued use. However, dihydroartemisinin (DHA) which showed to have a correlation significance (p<0.05) with most of the drugs used, calls for more research to expound on this finding

    Background: Paracetamol is the most common analgesic that is readily available with or without prescription over the counter and its toxicity is due to misuse or overdose associated with liver damage. Silymarin milk thistle is an edible herb believed to have antioxidant, scavenging and regulation of glutathione contents within the cell. Aim of the study was to determine the effects of different doses of silymarin milk thistle on liver biomarkers of paracetamol induced liver toxicity among albino rats. Methods: Twenty-four adult albino rats were randomly grouped into four groups each group consisting of six animals. Paracetamol-only group given high dose paracetamol-750 mg/kbwt and control group received water and food only. The experimental group, were divided into three subgroups with all receiving high dose paracetamol for five days and thereafter given varying doses of silymarin milk thistle. SIL-G1 given 200 mg/kbwt of silymarin milk thistle, SIL-G2 given 400 mg/kbwt of silymarin milk thistle and SIL-G3 was given 600 mg/kbwt of silymarin milk thistle.  All the animals were fed with standard rodent foods and water ad libitum. After completion of 24 hours of the last drug administration, they were humanely sacrificed, serum biochemical analysis of liver biomarkers (ALP, ALT and AST) was determined. Results: High dose of paracetamol induced liver toxicity. Upon administration of high dose of silymarin milk thistle, there was restoration of ALP, AST and ALP of all these biomarkers in relation to the control group. Conclusions: These findings showed that high dose (600 mg/kbwt) of silymarin milk thistle was found to have restorative effects and normalized liver biomarkers of ALP, AST and ALT

Uno studio di caso che s’inserisce pertanto all’interno della storia della professione docente e direttiva in Italia, tramite l'approccio microstorico. The essay traces the professional career of Maria Bartolini, a multi-faceted woman who lived to a long age. Author of children’s literature, teacher, but also director of studies and school inspector, he career crossed different scholastic seasons running from the late 1800s, the Giolittian age, Fascism and Republican Italy. Consequently, this case is part of the history of teaching and school management profession in Italy

Since factors  such  as  mixed  infection  of  drug resistant and  sensitive  parasites occurring in most field isolates can  influence  drug  test  outcome because they have different drug susceptibilities, Method: This study validated the use of a formulated transport medium (TM) by assaying clones which have been culture adapted in the lab for 50% inhibitory concentrations (IC50) against four antimalarials using SYBR Green 1 in vitro assay. These clones (3D7 and W2) were revived and recultured for assay. Prior to subjecting them to drugs, they were exposed to 2ml formulated TM and the conventional EDTA anticoagulant in 5ml vacutainer tubes to act as control.Results: Similarity was detected with no significant difference (P >0.05) observed in the medians inhibitory concentration 50 (IC50) between samples in the EDTA anticoagulant and TM holds however, samples in EDTA generally had higher IC50 values.Conclusion: This study concludes that the formulated TM holds promise for future use, Keywords: clones, transport medium, EDTA, susceptibility, inhibitory concentration 50, isolate
