Cancer cell adhesion measurement on the collagen layer in multiple shear stress levels
This paper presents a cell adhesion measurement chip with multiple shear stress zones in a single microchannel. In the experimental study, we used epithelial-like H358 and epithelial/mesenchymal-like A549 lung cancer cells to measure cell adhesion on the collagen layer. The detachment rate of H358 cells is 2∼4 times higher than that of A549 cells in the shear stress range of 34∼60dyne/cm2. Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. Properties of amino-terminal region of lipin-1
의과학과/석사[한글]전사 활성자로써의 lipin-1은, 현재까지 유전자의 구조 내에
DNA와 결합할 수 있는 부분이 발견된 바 없지만 카르복시말단
LXXIL부분이 다른 전사 인자들과 상호작용하여 전사 활성 기능
을 낸다고 보고되었다. 그러나 LXXIL을 포함하는 lipin-1 카르
복시말단 부위(423-947)에서는 yeast와 동물세포 모두에서 전사
활성화 기능을 관찰할 수 없었으며, 반대로 미노말단 부위
(1-441)에서 강한 전사 활성 기능을 확인할 수 있었다. 세분화
된 lipin-1의 전사 활성 기능 부위는 217-399번 아미노산 부위
이며, 이 부위 내에 존재하는 βisoform의 242-277번 아미노산
부위는 전사 활성 기능에 큰 영향이 없었다. Lipin-1의 전사 활
성화 기능 부위는 PGC-1α에 의해 그 활성이 증가되며, 다른
CBP, P/CAF, SRC-1, SRC-2, SRC-3에 의해서는 영향을 받지 않았
다. 전사 활성화 기능이 거의 제거된 lipin-1 아미노말단 부위
(1-327)와 단백질 상호작용을 가지는 유전자를 yeast
two-hybrid 방법으로 골격근 cDNA library를 선별한 결과 16개
의 유전자 cDNA를 밝히었다. 이 중에는 actin α1을 포함하는
cytoskeleton의 유전자가 7가지이었으며 PGM1, AGK, ECHS1,MAT2B
의 대사 효소들이 선별되었다. 그 외 5개 유전자들 중 흥미롭게
도 RNA polymerase Ⅱ의 소단위인 POLR2J와 단백질 합성에 관여
하는 EIF2A2, L30 및 ubiquitination에 관여하는 CUL1이 존재하
였다. 본 연구는 lipin-1의 아미노말단 부위의 전사 활성화 기
능 부위를 규명하였으며, 단백질 상호 작용이 있는 유전자들을
찾아내어 앞으로 진행해야 할 중요한 많은 연구과제들을 도출하
였다
[영문]Lipin-1 has been reported to interact with transcription factors and coactivator through its carboxy-terminal region containing LXXIL motif and to carry out transcriptional activation function. However, carboxy-terminal region (423-947), including DXDXT (active site for PAP1 activity) and LXXIL motives, did not show any transcriptional activation function in yeast and mammalian cells. In contrast, amino-terminal region (1-441) has strong transcription activation function in form of fusion protein with GAL4 DNA-binding domain. The activation domain could be delimitated from residue 217 to 399. The removal of residue 242 to 277, which is present in β isoform, did not affect the activities of its transcriptional activation function. The
transcriptional activation function of lipin-1 was enhanced by PGC-1α, but not by CBP, P/CAF, SRC-1, SRC-2 and SRC-3. In addition to transcription activation function, lipin-1 has several physiological roles in 35 skeletal muscle, which might be linked to protein-protein interactions.
For these reasons, skeletal cDNA library was screened to find out the genes showing the protein-protein interaction with lipin-1 amino-terminal region (1-327) by yeast two-hybrid system. Finally, 16 genes were revealed from 35 isolated clones. Among these genes, there are 7 cytoskeletal genes and 4 metabolic enzyme genes. The functional role of interactions with cytoskeletal proteins of lipin-1 should be addressed in
future. In metabolic enzymes, eight clones were isolated, containing the cDNA for carboxy-terminal region of PGM1. The strong binding of lipin-1 in skeletal muscle might affect the physiological activities of PGM1, resulting in changes of glucose and glycogen metabolisms. Lipin-1 can bind to ECHS1, which is one of the enzymes involved in mitochondrial fatty oxidation. The interaction with subunit J of RNA polymerase II, the ortholog of RPB11 of yeast, needs the further study in the view of the role of lipin-1 as a linker between transcriptional factors and RNA polymerase II.
The present study characterized and delimitate the activation function domain of lipin-1. I also identified the interactions of several proteins with lipin-1 amino-terminal regions and provide the important themes about their functional roles, which should be investigated.ope
