ENZYMATIC-SYNTHESIS OF D-P-HYDROXYPHENYLGLYCINE FROM DL-P-HYDROXYPHENYLHYDANTOIN IN THE PRESENCE OF ORGANIC COSOLVENT
Bacterial strain KBEL 101 isolated from soil was immobilized within polyacrylamide gel and used for the synthesis of D-p-hydroxyphenylglycine from DL-5-substituted hydantoin. The optimum reaction conditions for immobilized whole cell enzyme were pH 8.0 and temperature 30-degrees-C. Various water-miscible organic solvents were tested with respect to both the activity and the stability of whole cell enzyme, and 5% dimethylsulfoxide (DMSO) was chosen to be a proper solvent: hydantoinase activity increased twofold in the presence of 5% DMSO. From a practical standpoint, it is advantageous to obtain the highest conversion yield, and enzymatic synthesis of N-carbamoyl-D-p-hydroxyphenylglycine was carried out in repeated batch operation in the presence of 5% DMSO. Enhancement of operational stability of immobilized whole cell D-hydantoinase
A bacterium Pseudomonas sp. KBEL 101 isolated from soil was immobilized within polyacrylamide gel and used for the synthesis of D-p-hydroxyphenylglycine from DL-5-substituted hydantoin. The half-life of immobilized whole cell D-hydantoinase was found to be about 50 hrs. In order to increase the operational stability of immobilized whole cell D-hydantoinase, a carbon or nitrogen source was supplied with the reaction mixture in the continuous stirred tank reactor. As a sole source of carbon, glycerol was found to be most effective, and the activity of immobilized whole cell enzyme was maintained stably during 7 days when O.1%(W/V) glycerol solution was provided. In the case of nitrogen source, supplying of O.1%(W/V) yeast extract prolonged the half-life of immobilized whole cell D-hydantoinase to about 25 days
Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Modeling of branching density and branching distribution in low-density polyethylene polymerization
Low-density polyethylene (ldPE) is a general purpose polymer with various applications. By this reason, many publications can be found on the ldPE polymerization modeling. However, scission reaction and branching distribution are only recently considered in the modeling studies due to difficulties in measurement and computation of scission effect and branchings of polymer. Our previous papers [Kim, D.M., et al., 2004. Molecular weight distribution modeling in low-density polyethylene polymerization; impact of scission mechanisms in the case of CSTR. Chemical Engineering Science 59, 699-718; Kim, D.M., Iedema, P.D., 2004. Molecular weight distribution modeling in low-density polyethylene polymerization; impact of scission mechanisms in the case of a tubular reactor. Chemical Engineering Science, submitted for publication] are concerned with the scission reaction during ldPE polymerization and its effect on molecular weight distribution (MWD) of ldPE for various reactor types. Here we consider branching distributions as a function of chain length for CSTR and tubular reactor processes. To simultaneously deal with chain length and branching distributions, the concept of pseudo-distributions is used, meaning that branching distributions are described by their main moments. The computation results are compared with properties of ldPE samples from a CSTR and a tubular reactor. Number and weight average branchings and branching density increase as chain length increases until the longest chain length. The concentrations of long chain branching (LCB) are close to those of first branching moment in both CSTR and tubular reactor systems. The branching dispersity, a measure for the width of the branching distribution at a certain chain length, has the highest value at shorter chain length and then monotonously decreases approaching to 1.0 as chain length increases. Excellent agreements in branching dispersities between calculation with branching moments and prediction with assumption of binomial distribution for a tubular reactor and CSTR processes show that the branching distribution follows a binomial distribution for both processes
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Gluconic acid and sorbitol were simultaneously produced from glucose and Jerusalem artichoke using a glucose-fructose oxidoreductase of Zymomonas mobilis and inulinase. Inulinase was immobilized on chitin by cross-linking with glutaraldehyde. Cells of Z. mobilis permeabilized with toluene were coimmobilized with chitin-immobilized inulinase in alginate beads. The optimum amounts of both chitin-immobilized inulinase and permeabilized cells for coimmobilization were determined, and operational conditions were optimized. In a continuous stirred tank reactor operation, the maximum productivities for gluconic acid and sorbitol were about 19.2 and 21.3 g/L/h, respectively, at the dilution rate of 0.23 h-1 and the substrate concentration of 20%, but operational stability was low because of the abrasion of the beads. As an approach to increase the operational stability, a recycle packed-bed reactor (RPBR) was employed. In RPBR operation, the maximum productivities for gluconic acid and sorbitol were found to be 23.4 and 26.0 g/L/h, respectively, at the dilution rate of 0.35 h-1 and the substrate concentration of 20% when the recirculation rate was fixed at 900 mL/h. Coimmobilized enzymes were stable for 250 h in a recycle packed-bed reactor without any loss of activity, while half-life in a continuous stirred tank reactor (CSTR) was observed to be about 150 h
Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more
