    SummaryTo complete mitosis, Saccharomyces cerevisiae needs to activate the mitotic phosphatase Cdc14. Two pathways contribute to Cdc14 regulation: FEAR (Cdc14 early anaphase release) and MEN (mitotic exit network). Cdc5 polo-like kinase was found to be an important mitotic exit component. However, its specific role in mitotic exit regulation and its involvement in Cdc14 release remain unclear. Here, we provide insight into the mechanism by which Cdc5 contributes to the timely release of Cdc14. Our genetic and biochemical data indicate that Cdc5 acts in parallel with MEN during anaphase. This MEN-independent Cdc5 function requires active separase and activation by Cdk1-dependent phosphorylation. Cdk1 first phosphorylates Cdc5 to activate it in early anaphase, and then, in late anaphase, further phosphorylation of Cdc5 by Cdk1 is needed to promote its MEN-related functions

    Risk mitigating versus risk shifting : evidence from banks security trading in crises

    We show that risk-mitigating incentives dominate risk-shifting incentives in fragile banks. We study security trading by banks, as banks can easily and quickly change their risk exposure within their security portfolio. For identification, we exploit different crisis shocks and supervisory ISIN-bank-month-level data. Less capitalized banks take relatively less risk after financial stress shocks. Results hold within identical regulatory capital risk weights categories. Moreover, additional tests suggest that banks’ own incentives, rather than supervision, are the main drivers. Results hold for the different crisis shocks since 2007/08, including the COVID-19 one. A model of bank behavior rationalizes our findings

    Objective: To assess injury incidence, nature, severity, and mechanisms in amateur and professional polo players in Germany and Switzerland during the 2017 season.Methods: Questionnaire-based survey, where players were interviewed by the first author and injuries over the last two years recorded. The study was conducted from May-July 2017. Players were interviewed during tournaments and practice matches.Results: A total of 103 polo players were interviewed. Forty percent (n=41) of the players experienced a total of 55 injuries. The injury rate was 5.3-6.1/1000 player-game hours. Sixty-seven percent of all injuries were classified as major. Trauma was the main (84%; 46 cases) cause of injury. Most cases (42%) involved the upper extremity, followed by the lower extremity in 14 (25%) cases. The trunk was affected in 12 cases (22%) and the head in 6 (11%). Overall, a typical pain pattern after playing polo (without missing play) was observed in the lumbar spine, groin, and hitting arm.Conclusions: Although the injury rate was low, the severity was quite high. Typical injury and overuse patterns are presented in this study. Life-threatening injuries do occur, and medical staff trained in trauma management should be present at polo matches. Safer helmets according to the newest standards should be worn and the face should be protected to avoid fatal injuries.Clinical Relevance: Polo is a dangerous team sport that can lead to major injuries and suitable preventive methods should be developed.KEY WORDS: Polo, Injury Rate, Injury Severity, Polo Rider

    Crystal structure of the polo-box domain of polo-like kinase 2

    Polo-like kinase 2 (PLK2) is a crucial regulator in cell cycle progression. DNA damage response, and neuronal activity. PLK2 is characterized by the conserved N-terminal kinase domain and the unique C-terminal polo-box domain (PBD). The PBD mediates diverse functions of PLK2 by binding phosphorylated SerpSer/pThr motifs of its substrates. Here, we report the first crystal structure of the PBD of PLK2. The overall structure of the PLK2 PBD is similar to that of the PLK1 PBD, which is composed by two polo boxes each contain beta 6 alpha structures that form a 12-stranded beta sandwich domain. The edge of the interface between the two polo boxes forms the phosphorylated Ser-pSer/pThr motifs binding cleft. On the hand, the peripheral regions around the core binding cleft of the PLK2 PBD is distinct from that of the PLK1 PBD, which might confer the substrate specificity of the PBDs of the polo-like kinase family.
