    Analysis of 6-methoxy podophyllotoxin and podophyllotoxin in hairy root cultures of Linum album Kotschy ex Boiss

    Linum album is a herbaceous medicinal plant that contains some lignans with antiviral and anticancer properties such as podophyllotoxin (PTOX) and 6-methoxy podophyllotoxin (MPTOX). In this research, hairy root cultures of L. album were established by transformation with Agrobacterium rhizogenes strains LBA9402, A4, AR15834 and A. tumefaciens strain C58C1 (pRiA4). The presence of PTOX and MPTOX in the hairy roots was verified by ESI/MS in positive ion mode. MPTOX was confirmed and its enantiomer determined by nuclear magnetic resonance spectroscopy and circular dichroism spectroscopy, respectively. PTOX and MPTOX production was determined by HPLC, in different lines of hairy roots. The results showed that all obtained hairy root lines produced higher yield of lignans than mother plant roots. In addition, the lignans content in the roots derived from A. rhizogenes strain LBA9402 was higher than in those obtained from A. tumefaciens strain C58C1.Fil: Ahmadian Chashmi, Najmeh.     [ES] En este Trabajo Final de grado (TFG) se exponen los resultados de un estudio técnico y de estado de conservación de una obra perteneciente a la colección de pinturas del Museo Escolar de Pusol en Elche. La obra, realizada al óleo sobre lienzo es de autor anónimo y en ella se representa una Epifanía. Este TFG se elabora a partir de los estudios histórico, iconográfico, de aspectos técnicos y estructurales, diagnóstico de patologías y el estudio del estado de conservación de la obra. Tras la recogida de la información pertinente, se elabora una propuesta de intervención adecuada para la pieza en cuestión. La propuesta de intervención contempla y especifica los materiales y procesos necesarios para asegurar su integridad. Finalmente se propone un plan de conservación preventiva para minimizar los riesgos de exposición de la obra a factores perjudiciales para su adecuada conservación. Palabras clave: Lienzo, pintura, Epifanía, Museo Escolar de Pusol, Elche.[EN] This Final Degree Work (FDW) sets out the results of a technical study and conservation status of a work belonging to the collection of paintings of the Pusol School Museum in Elche. The artwork, made in oil on canvas is by an anonymous author and represents an Epiphany. This TFG is elaborated from historical, iconographic studies, technical and structural aspects, diagnosis of pathologies and the study of the conservation status of the work. Following the collection of relevant information, an appropriate intervention proposal is prepared for the artwork in question. The intervention proposal contemplates and specifies the materials and necessary procedures to ensure their integrity. Finally, a preventive conservation plan is proposed to minimize the risks of exposure of the work to factors detrimental to its proper conservation.     Effect of in vitro morphogenesis on the production of podophyllotoxin derivatives in callus cultures of Linum album

    34 p.-9 fig.The anticancer compound podophyllotoxin and other related lignans can be produced in Linum album in vitro cultures, although their biosynthesis varies according to the degree of differentiation of the plant material. In general, L.album cell cultures do not form the same lignans as roots or other culture systems. Our aim was to explore how the lignan-producing capacity of organogenic cell masses is affected by the conditions that promote their formation and growth. Thus, L.album biomass obtained from plantlets was cultured in darkness or light, with or without the addition of plant growth regulators, and the levels of podophyllotoxin, methoxypodophyllotoxin and other related lignans were determined in each of these conditions. The organogenic capacity of the cell biomass grown in the different conditions was studied directly and also with light and scanning electronic microscopy, leading to the observation of.several somatic embryos and well-formed shoots. The main lignan produced was methoxypodophyllotoxin, whose production was clearly linked to the organogenic capacity of the cell biomass, which to a lesser extent was also the case for podophyllotoxin.Peer reviewe

    International audienceWhen creating a real-time character animation (e.g. for video games), fine grain details such as cloth wrinkles are very often omitted due to the limitation of available resources. However, we believe that they are crucial to increase realism, hence user immersion. In this paper, we present a perceptual study of dynamic wrinkles on clothes to show that they indeed have an important impact on realism. The models, animations and textures we used are game realistic and the wrinkling algorithm is fast enough to be used in a game. We have found that parameters influencing the perception of dynamic wrinkles include movement type and speed, viewing angle, and texture colors. Our results should be useful for animators or game designers to help them add wrinkles only where they are necessary, and activate them only when they have an impact on realism
