The role of Plasmodium falciparum var genes in malaria in pregnancy

    No full text
Sequestration of Plasmodium falciparum-infected erythrocytes in the placenta is responsible for many of the harmful effects of malaria during pregnancy. Sequestration occurs as a result of parasite adhesion molecules expressed on the surface of infected erythrocytes binding to host receptors in the placenta such as chondroitin sulphate A (CSA). Identification of the parasite ligand(s) responsible for placental adhesion could lead to the development of a vaccine to induce antibodies to prevent placental sequestration. Such a vaccine would reduce the maternal anaemia and infant deaths that are associated with malaria in pregnancy. Current research indicates that the parasite ligands mediating placental adhesion may be members of the P. falciparum variant surface antigen family PfEMP1, encoded by var genes. Two relatively well-conserved subfamilies of var genes have been implicated in placental adhesion, however, their role remains controversial. This review examines the evidence for and against the involvement of var genes in placental adhesion, and considers whether the most appropriate vaccine candidates have yet been identified

George Rowe Smith, LL.B. Central City, Kentucky Heavyweight Boxing Championship \u2712; President Freshman Class; Varsity Track (1); Varsity Football (4) ; Union Literary Society; Democratic Club; Y. M. C. A. Cabinet; Prohibition Club. If well thou hast begun, go on fore-right; It is the end that crowns us, not the fight. George R. began life in the Prep and has had many a delightful day since he could first call himself a Freshman. He admits that he is differently constituted from the rest since no one can hurt him. He has measured arms with some sturdy fighters but still holds his championship in boxing. His crowning glory came when he received his letter in football after many years of faithful toil. -The Kentuckian, 1915--------------------------------- George Rowe Smith (January 16, 1890 - December 18, 1975) was born in Central City, Kentucky to George Smith and Martha Eleanor Rowe. Smith stayed in Lexington and entered into a law partnership with fellow class of 1915 student Jacob Owen Reynolds. Smith practiced law in Lexington for over 50 years before retiring in 1972. Smith was also involved in Lexington\u27s motel industry. He married twice--his wives were Ella Mae Clark and Mattie Stella Clark.https://uknowledge.uky.edu/klapp_1915/1029/thumbnail.jp

IDH1 gene mutations are revealed in up to 20% of the patients with acute myeloid leukemia (AML). A mutant IDH enzyme, existing in the cell cytoplasm and possessing neomorphic activity, converts αKG into oncometabolite R-2-hydroxyglutarate (R-2-HG) that accumulates in high amounts in the cell and inhibits αKG-dependent enzymes, including epigenetic regulators. The resultant alteration in gene expression and blockade of differentiation ultimately lead to leukemia development. Myeloid differentiation capacity can be restored by obstruction of the mutant enzyme, inducing substantial reduction in R-2-HG levels. Ivosidenib, a potent selective inhibitor of mutant IDH1, is a differentiating agent shown to be clinically effective in newly diagnosed AML (ND-AML) and relapsed/refractory (R/R) AML harboring this mutation. The drug is approved by the Food and Drug Administration (FDA) as a single-agent treatment for R/R AML. Significance of mutated IDH1 targeting and a potential role of ivosidenib in AML management, when used either as a single agent or as part of combination therapies, will be reviewed herein. Keywords: mutant IDH1, acute myeloid leukemia, ivosideni

The article addresses the intellectual !gure of the Anglo-American Colin Rowe, in a fragmented way, focusing on some aspects that have marked milestones in the historiography and architectural interpretation of the 20th century. Plasmodium falciparum:Rosettes do not protect merozoites from invasion-inhibitory antibodies

    No full text
Rosetting is a parasite adhesion phenotype associated with severe malaria in African children. Why parasites form rosettes is unknown, although enhanced invasion or immune evasion have been suggested as possible functions. Previous work showed that rosetting does not enhance parasite invasion under standard in vitro conditions. We hypothesised that rosetting might promote invasion in the presence of host invasion-inhibitory antibodies, by allowing merozoites direct entry into the erythrocytes in the rosette and so minimising exposure to plasma antibodies. We therefore investigated whether rosetting influences invasion in the presence of invasion-inhibitory antibodies to MSP-1. We found no difference in invasion rates between isogenic rosetting and non-rosetting lines from two parasite strains, R29 and TM284, in the presence of MSP-1 antibodies (P = 0.62 and P = 0.63, Student's t test, TM284 and R29, respectively). These results do not support the hypothesis that rosettes protect merozoites from inhibitory antibodies during invasion. The biological function of rosetting remains unknown
