    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     Humane Papillomviren (HPV) induzieren benigne Tumoren der Haut oder Schleimhaut, die nach langjähriger viraler Persistenz maligne entarten können. Je nach Entartungsrisiko der induzierten Primärläsion werden die humanen Papillomviren in hoch-karzinogene („high-risk“) und niedrig-karzinogene („low-risk“) Virustypen eingeteilt. Das onkogene Potential humaner genitaler Papillomviren wird durch die Aktivität der viralen Onkogene E6 und E7 bestimmt, die bei HPV6 als Vertreter der niedrig-karzinogenen Virustypen über zwei unabhängige Promotoren P1 und P2 reguliert werden, während bei den hoch-karzinogenen Typen die Regulation über nur einen Promotor erfolgt. Die Regulation der beiden frühen Promotoren von HPV6 erfolgt differentiell und zelltypspezifisch über den viralen Transkriptionsfaktor E2 und seine Bindung an vier spezifische Erkennungssequenzen in der nicht-kodierenden Region des HPV6-Genoms sowie vermutlich durch zelluläre Proteine. So zeigen RTS3b-Zellen in Anwesenheit von E2 ein E6/E7-Expressionsmuster, das demjenigen natürlich infizierter, gutartiger Kondylomata ähnelt, während in der Karzinom-Zelllinie HT3 eine Umkehrung der E2-vermittelten Repression des E6-Promotors zu einer Aktivierung beobachtet wird. Diese unterschiedliche E2-vermittelte Regulation der frühen Promotoren in den beiden Epithelzelllinien deutet auf eine Beteiligung zellulärer Faktoren hin, die durch Bindung an die E2-Bindestellen oder deren unmittelbare Umgebung mit der Funktion von E2 interferieren könnten. In der vorliegenden Arbeit konnte mittels des „Yeast One-Hybrid“-Systems ein humanes Protein, das „cervical cancer suppressor 3“-Protein, identifiziert werden, das potentiell mit der Bindung von E2 an die E2-Bindestellen 3 und 4 von HPV6 interferiert und so an der Regulation der frühen Promotoren von HPV6 in der Zelllinie HT3 beteiligt sein könnte. Eine endgültige Beurteilung, ob es sich bei dem identifizierten humanen Protein tatsächlich um einen Faktor handelt, der mit der DNA-Bindung von E2 und damit der E2-vermittelten Transkriptionsregulation von HPV6 interferiert, kann erst nach Verifizierung der Expression eines Fusionsproteins und gegebenen-falls Überprüfung der DNA-Bindung durch unabhängige Methoden erfolgen.Human papillomaviruses (HPV) induce benign tumours of the skin or mucosa, which can progress into malignant lesions after long-term viral persistence. According to the risk of progression of the induced primary lesion, human papillomaviruses are grouped into ”high-risk” and ”low-risk” virus types. The oncogenic potential of human genital papillomaviruses is determined by the activity of the viral oncogenes E6 and E7. These are regulated by two independent promoters P1 and P2 in HPV as a representative of the low-risk virus types, whereas the regulation in high-risk types is mediated by only one promoter. Both early promoters of HPV6 are differentially and cell type-specific regulated by the viral transcription factor E2 and its binding to four specific recognition sequences in the non-coding region of the HPV6 genome and probably by cellular proteins as well. Accordingly, RTS3b cells show an E6/E7 expressional pattern in presence of E2 resembling naturally infected benign condylomata, whereas in the carcinoma cell line HT3 a reversion of the E2-mediated repression of the E6-promoter towards activation can be found. This differential E2-mediated regulation of the early promoters in both epithelial cell lines indicates an involvement of cellular factors that are presumed to interfere with the E2 function by binding to the E2-binding sites or directly adjacent sites. In the present study, a human protein, the ”cervical cancer suppressor 3” protein, has been identified with the help of the ”Yeast One-Hybrid” system. This protein potentially interferes with the binding of E2 to its recognition sites E2BS-3 and -4 of HPV6 and thereby it is likely to be involved in the regulation of the early promoters of HPV6 in the HT3 cell line. However, a final conclusion whether the identified human protein indeed is a factor interfering with DNA-binding of E2 and thereby with the E2-mediated transcriptional regulation of HPV6 can only be drawn after the verification of the expression of the protein and confirmation of its DNA-binding abilities by independent methods

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Charakterisierung von humanen Milchzellen während der Laktation und der mögliche Einfluss der Reaktivierung von humanem Zytomegalievirus

    Muttermilch als optimale Versorgung des Neugeborenen stellt durch die hohe Exkretion von humanem Cytomegalievirus die Hauptinfektionsquelle von Säuglingen dar. Obwohl in früheren Studien immer wieder die Reaktivierung sowie der Zelltropismus von HCMV untersucht wurden, ist bis heute der Ort der HCMV Persistenz und Reaktivierung in Muttermilch weitgehend unbekannt. In den letzten Jahren wurde die Kinetik der HCMV-Reaktivierung in Muttermilch weiter erforscht. Auch im Rahmen dieser Arbeit konnte eine hohe Rate an humanen Zytomegalieviren in Muttermilch bei HCMV-seropositiven Spenderinnen erfolgen, welche der Kinetik von Maschmann beschrieben folgt (Maschmann et al., 2015). Trotz allgemeiner Kinetik ist der Verlauf der Reaktivierung mit Erstnachweis von HCMV sowohl in zellfreier Molke als auch in Milchzellen intraindividuell unterschiedlich. Die zelluläre Zusammensetzung von Muttermilch zeigte sich auch im Rahmen dieser Arbeit individuell spezifisch und Laktationszeitpunkt abhängig. Die morphologische Darstellung aller Zellen mittels Panoptischer Färbung nach Pappenheim sowie spezifisch von Monozyten/Makrophagen mittels alpha-Naphthylacetatesterase-Färbung zeigte typische Unterschiede von Milchzellen zu Blutzellen. Die charakteristischen lipidhaltigen Vakuolen in phagozytosefähigen Zellen, welche im Verlauf der Laktation zunahmen, sind in Muttermilch ebenso einzigartig, wie die schwankende Zellgröße einzelner Zellen, vor Allem von Monozyten/Makrophagen. Die Untersuchung von Leukozytensubpopulationen mittels Durchflusszytometrie lieferte eine eindeutige Immunsignatur jeder Spenderin zu einem spezifischen Laktationszeitpunkt. Im Vergleich der Muttermilchzellen und Immunzellsignaturen von HCMV-seronegativen zu HCMV-seropositiven Spenderinnen fanden sich prominente Unterschiede. HCMV-seropositive Spenderinnen zeigten im Verhältnis zu HCMV-seronegativen Spenderinnen eine durchschnittlich deutlich erhöhte Zellzahl über den gesamten Verlauf der Laktation. Es zeigte sich weiterhin, dass um den Zeitpunkt der HCMV-Reaktivierung die Zellzahl stark anstieg. Im Verlauf der Laktation waren antigenpräsentierende Zellen wie T-Zellen und Makrophagen vor allem bei HCMV-seropositiven Spenderinnen prominent. Die Gesamtzahl der Monozyten/Makrophagen fiel in den durchflusszytometrischen Untersuchungen bei HCMV-seropositiven Spenderinnen signifikant ab. Die fortlaufende Analyse einer HCMV-seropositven Mutter zeigte, dass der Abfall der Monozyten/Makrophagen womöglich nur ein temporäres Absinken während dem Maximum der HCMV-Reaktivierung darstellt und im Verlauf, wie teilweise in früheren Studien ersichtlich, wieder ansteigt. Eine mögliche Erklärung könnte die Lyse von infizierten Monozyten/Makrophagen im Rahmen der Virusreplikation und Exkretion darstellen. Dies würde präliminäre Befunde, dass Monozyten/Makrophagen der Ort der Viruspersistenz und -vermehrung darstellen unterstützen (Maschmann et al., 2015). Möglicher Ansatzpunkt folgender Studien könnte die separate Analyse dieser Zellpopulation auf Vorliegen von HCMV darstellen. Im Rahmen dieser Arbeit wurden erstmals Milchzellen mittels ImageStream-Verfahren untersucht. Muttermilchbestandteile konnten so mittels Auflichtbild identifiziert werden und Muttermilchzellen anhand ihrer Antikörperexpression mittels Zytofluorometrie in klare Subpopulationen unterschieden werden. Dies ergänzt und erweitert die klassische Analyse der Zellen mittels Durchflusszytometrie. Weiterhin bietet dieses Verfahren neue Möglichkeiten auch Subpopulationen mit geringer Größe zu untersuchen und auf Grund ihrer Morphologie und der Antikörperverteilung ihrer Oberfläche zu kategorisiert. Diese Arbeit soll dazu beitragen, das Kompartiment Muttermilch während der Laktation besser analysieren zu können sowie den möglichen Einfluss von HCMV auf die zelluläre Zusammensetzung der Muttermilch, den Ort der Persistenz und Übertragungswege auf den Säugling zu verstehen

    Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
