    Pathological gambling and romantic relationships: The perception of the members of the couple regarding the quality of their relationship

    Pathological gambling is a relevant condition that negatively affects the family system of gamblers. The aim of the present study was to explore the specific qualitative aspects of romantic relationship and the style of romantic attachment of pathological gamblers and their partners. Thirty-five couples with a partner that had been diagnosed with Gambling Disorder and 45 control couples were recruited for the present study. All participants filled out a battery of questionnaires aimed at assessing the quality of their couple relationship and their romantic attachment. A series of multivariate analyses of variance (MANOVA) were conduct- ed. Results showed that both pathological gamblers and their partners reported a worse quality of their romantic relationship than partners of a control group, but this is stronger especially for partners of pathological gamblers. In fact, pathological gamblers reported higher levels of conflict and lower levels of help and security than partners of the control group. Indeed, partners of pathological gamblers reported a worse quality of couple relationship on all dimensions investigated: they reported higher levels of conflict and lower levels of companionship, help, security, and closeness than partners involved in a relationship with non- pathological gamblers. Moreover, both pathological gamblers and their partners reported a more insecure romantic attachment, both anxious and avoidance, than did partners of the control group. Limitations, strengths, and theoretical and clinical implica- tions are discussed

    Iron incorporation into Escherichia coli Dps gives rise to a ferritin-like microcrystalline Core

    Escherichia coli Dps belongs to a family of bacterial stress-induced proteins to protect DNA from oxidative damage. It shares with Listeria innocua ferritin several structural features, such as the quaternary assemblage and the presence of an unusual ferroxidase center. Indeed, it was recently recognized to be able to oxidize and incorporate iron. Since ferritins are endowed with the unique capacity to direct iron deposition toward formation of a microcrystalline core, the structure of iron deposited in the E. coli Dps cavity was studied. Polarized single crystal absorption microspectrophotometry of iron-loaded Dps shows that iron ions are oriented. The spectral properties in the high spin 3d5 configuration point to a crystal form with tetrahedral symmetry where the tetrahedron center is occupied by iron ions and the vertices by oxygen. Crystals of iron-loaded Dps also show that, as in mammalian ferritins, iron does not remain bound to the site after oxidation has taken place. The kinetics of the iron reduction/release process induced by dithionite were measured in the crystal and in solution. The reaction appears to have two phases, with t1/2 of a few seconds and several minutes at neutral pH values, as in canonical ferritins.     Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Trypanothione reductase from leishmania infantum: cloning, expression, purification, crystallization and preliminary x-ray data analysis

    The most promising targets for Leishmania-specific drug design are two key enzymes involved in the unique thiol-based metabolism, common to all parasites of the Trypanosomatidae family: trypanothione synthetase (TryS) and trypanothione reductase (TR). Recently, new inhibitors of TR have been identified such as polyamines and tricyclic compounds. The knowledge of the three-dimensional structure of Leishmania TR will shed light on the mechanism of interaction of these inhibitors with TR and will be the starting point to design novel lead candidates to facilitate the development of new effective and affordable drugs. Trypanothione reductase from Leishmania infantum has been cloned, expressed in E. coli and purified. Crystals were obtained at 294 K by the hanging drop vapour diffusion method using ammonium sulfate as precipitant agent and diffract to better than 2.95 Å resolution using a synchrotron radiation source. The crystals exhibit an unusually high solvent content of 74 %, belong to the tetragonal space group P41 with units cell parameters a=b=103.45 Å, c=192.62 Å and two molecules in the asymmetric unit. The protein X-ray structure has been solved by Molecular Replacement and the model is under construction. © 2009 Bentham Science Publishers Ltd

    Molecular basis of antimony treatment in leishmaniasis

    Leishmaniasis is a disease that affects 2 million people and kills 70000 persons every year. It is caused by Leishmania species, which are human protozoan parasites of the trypanosomatidae family. Trypanosomatidae differ from the other eukaryotes in their specific redox metabolism because the glutathione/glutathione reductase system is replaced by the unique trypanothione/trypanothione reductase system. The current treatment of leishmaniasis relies mainly on antimonial drugs. The crystal structures of oxidized trypanothione reductase (TR) from Leishmania infantum and of the complex of reduced TR with NADPH and Sb(III), reported in this paper, disclose for the first time the molecular mechanism of action of antimonial drugs against the parasite. Sb(III), which is coordinated by the two redox-active catalytic cysteine residues (Cys52 and Cys57), one threonine residue (Thr335), and His461' of the 2-fold symmetry related subunit in the dimer, strongly inhibits TR activity. Because TR is essential for the parasite survival and virulence and it is absent in mammalian cells, these findings provide insights toward the design of new more affordable and less toxic drugs against Leishmaniasis. © 2009 American Chemical Society

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Pellet production from residual biomass of greenery maintenance in a small-scale company to improve sustainability

    Replacing fossil energy sources with renewable energy sources is a key strategic action to limit environmental issues. To achieve this goal, substitution with biomass is beneficial due to its versatility in various fields. In terms of circular economy and sustainability, the possibility of energy exploitation of residual biomass is particularly desirable in small-medium enterprises. The use of supply chain by-products can improve sustainability and create opportunities for companies. The purpose of this study is to evaluate the suitability of residual biomass of conifers and broad-leaved trees to produce quality pellets using an agri-pellet machine activated by the power take-off of a tractor. This system can be employed at the farm level. Wood biomass of four species was tested; poplar, stone pine, black locust, and oak. Wood chips samples were analyzed to determine their qualitative characteristics following the technical standard ISO 17225-4. Based on the results, different wood blends were created to produce pellets, subsequently characterized according to ISO 17225-2. The analyses carried out on wood chips and pellets were bulk density, moisture, ash content, calorific value, elemental composition, chlorine, sulfur, and heavy metals. In addition, particles size was measured only for wood chips, while the length, diameter, mechanical durability, and ash melting behaviors were determined only for pellets. Some of the analyzed mixtures show acceptable values according to the current ISO technical standards. The values related to the apparent pellet bulk density and the durability test highlight that not all the mixtures are suitable to produce quality pellets. Results also represent a good starting point for future studies

    Huntingtin ubiquitination mechanisms and novel possible therapies to decrease the toxic effects of mutated huntingtin

    Huntington Disease (HD) is a dominant, lethal neurodegenerative disorder caused by the abnormal expansion (>35 copies) of a CAG triplet located in exon 1 of the HTT gene encoding the huntingtin protein (Htt). Mutated Htt (mHtt) easily aggregates, thereby inducing ER stress that in turn leads to neuronal injury and apoptosis. Therefore, both the inhibition of mHtt aggregate formation and the acceleration of mHtt degradation represent attractive strategies to delay HD progression, and even for HD treatment. Here, we describe the mechanism underlying mHtt degradation by the ubiquitin–proteasome system (UPS), which has been shown to play a more important role than the autophagy–lysosomal pathway. In particular, we focus on E3 ligase proteins involved in the UPS and detail their structure–function relationships. In this framework, we discuss the possible exploitation of PROteolysis TArgeting Chimeras (PROTACs) for HD therapy. PROTACs are heterobifunctional small molecules that comprise two different ligands joined by an appropriate linker; one of the ligands is specific for a selected E3 ubiquitin ligase, the other ligand is able to recruit a target protein of interest, in this case mHtt. As a consequence of PROTAC binding, mHtt and the E3 ubiquitin ligase can be brought to a relative position that allows mHtt to be ubiquitinated and, ultimately, allows a reduction in the amount of mHtt in the cell
