No full text학위논문(석사)--아주대학교 일반대학원 :분자과학기술학과,2011. 2약물대사 (drug metabolism) 혹은 생체내변화 (biotransformation)란 약물이 다른 화학 구조로 전환되는 생리화학적 과정을 말한다. 체내의 많은 조직도 약물대사능력이 있지만 대부분의 약물은 세망내피계 (endoplasmic reticulum)로부터 파생된 세포부분인 마이크로좀 (microsome) 내에 존재하는 효소인 cytochrome P450 (CYP)에 의해 간에서 주로 대사된다. 본 논문에서는 fenbendazole과 diclofenac의 대사산물을 합성하고 약물대사 후 분석에 대해 연구를 하였다.
Fenbendazole은 구충제로 파충류 및 포유류들에게 사용되고 간에 있는 CYP와 마이크로좀 효소에 의해 산화대사가 일어지만 주 대사산물인 oxfendazole에 의해 간에 있는 cytochrome P450-isozyme의 활성이 커지면서 간암을 유발하는 것으로 알려져 있고 diclofenac은 인간에게 사용하는 비스테로이드성 항 염증 약물로 간에 있는 CYP3A4와 CYP2C9에 의해 주요 대사작용이 진행되지만 대사산물인 4’-hydroxydiclofenac과 5-hydroxydiclofenac이 간 독성을 유발한다는 결과들이 보고되었다.
이에 fenbendazole과 diclofenac의 대사산물들을 합성하여 S9 enzyme에서 in vitro 실험을 통해 CYP에 의해 분해되는 대사산물을 확인하고, 이를 LC-MS를 사용하여 정성 및 정량 분석을 진행한 분석조건을 정립하였다. 또한, diclofenac의 대사산물인 4’-hydroxydiclofenac의 합성 방법을 개선하여 금속 및 수소가스와 같은 위험한 시약을 사용하지 않고, 중간체를 바꾸어 반응을 쉽고 높은 수율로 화합물을 얻을 수 있었다. 결과적으로 수율을 기존보다 약 3.6배 개선할 수 있었다. 새로운 반응조건에서는 중간체 합성 시 반응용매를 MeOH 대신 EtOH을 사용하여 수율을 45%에서 94%로 개선할 수 있었고, 마지막 가수분해 반응에서는 radical을 제거해주는 CuCl을 사용하여 indoline 부산물의 생성을 피할 수 있었다. 이러한 새로운 방법을 적용하여 더욱 안전하고 용이한 방법으로 대사산물들을 합성하였다.Syntheses and Quantifications of Fenbendazole, Diclofenac and their Metabolites 1
