    RNA-dependent association with myosin IIA promotes F-actin-guided trafficking of the ELAV-like protein HuR to polysomes

    The role of the mRNA-binding protein human antigen R (HuR) in stabilization and translation of AU-rich elements (ARE) containing mRNAs is well established. However, the trafficking of HuR and bound mRNA cargo, which comprises a fundamental requirement for the aforementioned HuR functions is only poorly understood. By administering different cytoskeletal inhibitors, we found that the protein kinase Cδ (PKCδ)-triggered accumulation of cytoplasmic HuR by Angiotensin II (AngII) is an actin-myosin driven process functionally relevant for stabilization of ARE-bearing mRNAs. Furthermore, we show that the AngII-induced recruitment of HuR and its bound mRNA from ribonucleoprotein particles to free and cytoskeleton bound polysomes strongly depended on an intact actomyosin cytoskeleton. In addition, HuR allocation to free and cytoskeletal bound polysomes is highly sensitive toward RNase and PPtase and structurally depends on serine 318 (S318) located within the C-terminal RNA recognition motif (RRM3). Conversely, the trafficking of the phosphomimetic HuRS318D, mimicking HuR phosphorylation at S318 by the PKCδ remained PPtase resistant. Co-immunoprecipitation experiments with truncated HuR proteins revealed that the stimulus-induced association of HuR with myosin IIA is strictly RNA dependent and mediated via the RRM3. Our data implicate a microfilament dependent transport of HuR, which is relevant for stimulus-induced targeting of ARE-bearing mRNAs from translational inactive ribonucleoprotein particles to polysomes

    Modified potential for atomistic simulation of the growth of carbon materials from binary alloy catalysts

    © 2019 the Owner Societies.A new hybrid bond order potential has been developed and implemented to describe carbon-bimetallic alloy interactions, which are involved in the catalytic growth of carbon materials such as graphene and carbon nanotubes on the surface of binary alloy catalysts. In carefully adjusting the parameters, the potential energy fitting correlated with the results calculated from the density functional theory (DFT) method leads to a high quality empirical force field with an average error of <4.5% only. With the PES accuracy, in total 16 (n,m) have been successfully obtained from the MD trajectories in this work, and the structural evolution including random chirality and diameter formation has been identified. The newly modified force field is expected to be useful for modelling the spontaneous growth of carbon materials, particularly tubes on binary alloy clusters, giving an idea of how these C-C, C-M, and M-M interactions affect the growth behavior of carbon nanotubes. In addition, the new FF is only valid for liquid alloy nanoparticles at this time, but the use of solid alloy nanocatalysts with the new FF can be further employed for 2-D material growth such as graphene layer growth11sciescopu

    Kaposi's sarcoma (KS) is the most common cancer among HIV-positive patients. Histogenetic origin of KS has long been elusive due to a mixed expression of both blood and lymphatic endothelial markers in KS tumor cells. However, we and others discovered that Kaposi's sarcoma herpes virus (KSHV) induces lymphatic reprogramming of blood vascular endothelial cells by upregulating PROX1, which functions as the master regulator for lymphatic endothelial differentiation. Here, we demonstrate that the KSHV latent gene kaposin-B enhances the PROX1 mRNA stability and plays an important role in KSHV-mediated PROX1 upregulation. We found that PROX1 mRNA contains a canonical AU-rich element (ARE) in its 3'-untranslated region that promotes PROX1 mRNA turnover and that kaposin-B stimulates cytoplasmic accumulation of the ARE-binding protein HuR through activation of the p38/MK2 pathway. Moreover, HuR binds to and stabilizes PROX1 mRNA through its ARE and is necessary for KSHV-mediated PROX1 mRNA stabilization. Together, our study demonstrates that kaposin-B plays a key role in PROX1 upregulation during lymphatic reprogramming of blood vascular endothelial cells by KSHV

    Comparison of the i-gel with the AuraGain laryngeal mask airways in patients with a simulated cervical immobilization: a randomized controlled trial

    BACKGROUND: The use of second generation supraglottic airway devices is recommended for airway rescue in failed tracheal intubation. This study was performed to compare the clinical performance of the i-gel with that of the AuraGain in patients with simulated cervical immobilization. METHODS: We conducted a prospective, randomized controlled trial in 104 patients undergoing general anesthesia from June to September 2018 at the Seoul National University Hospital. Patients were randomly allocated to receive either the i-gel or the AuraGain device. A difficult airway was simulated using a cervical collar limiting the mouth opening and neck movement. The primary outcome was the initial oropharyngeal leak pressure. RESULTS: The rate of successful insertion at the first attempt was 92.3% for the i-gel and 86% for the AuraGain. There were no significant differences in the initial and 5-min oropharyngeal leak pressures between the i-gel and the AuraGain (21+/-4 vs. 22+/-5 cmH2O, P=0.229; and 22+/-5 vs. 23+/-5 cmH2O, P=0.308, respectively). The time to successful device insertion was shorter (20 [16-23] vs. 25 [20-41] s, P&lt;0.001) and device insertion was easier (P&lt;0.001) in the i-gel group than in the AuraGain group. The blood staining of the device was more frequently observed in the AuraGain (3 [5.8%] vs. 12 [23.5%] patients, P=0.003). CONCLUSIONS: The i-gel and the AuraGain showed comparable oropharyngeal leak pressures and success rates in the first attempt in patients with simulated cervical immobilization. However, the i-gel was easier to insert and required less time for insertion than the AuraGain

Emergency medicine builds up the front line to maximize the utility of clinically validated biomarkers and is the cutting edge field to test the applicability of promising biomarkers emerging from thorough translational researches. The role of biomarkers in clinical decision making would be of greater significance for identification, risk stratification, monitoring, and prognostication of the patients in the critical- and acute-care settings. No doubt basic research to explore novel biomarkers in relation to the pathogenesis is as important as its clinical counterpart.     Hur digitalt tillgänglig data kan bidra till avfallsminsking i byggbranschen

    Med bakgrund i att byggbranschen behöver reducera mängden avfall genom högre grad av förebyggande åtgärder, återanvändning och återvinning av byggnadskomponenter har arbetets syfte varit att visa hur digitalt tillgängliggjord relevant data kan bidra till detta. Arbetet har utifrån produktkategorin fönster och förutsättningarna för ett specifikt nybyggnationsprojekt, där involverade parter intervjuats, kartlagt hur det fysiska fönstrets, samt dess informationsmodells, värdekedja ser ut. Projektets förutsättningar har också legat till grund för kartläggning av vilka avfallsminskande åtgärder som redan utförs samt vilken data som vore relevant att ta fram eller använda för att uppnå en högre grad av avfallsminskning. Några av de utmaningar som finns med att ta fram eller använda datan, samt informationshanteringen hos och mellan olika parter har identifierats men även incitament och möjligheter som finns. I arbetets studier har det även ingått att undersöka förutsättningarna för att visa en möjlig lösning på hur relevant data kan presenteras och användas ur en informationsmodell. Detta har resulterat i en Proof-of-Concept där relevant data, utifrån projektets förutsättningar, berikats in i informationsmodellen för att sedan presentera några exempel på hur den kan användas vid de avfallsminskande åtgärderna i olika skeden av fönstrets livscykel. Lösningen som har presenterats tar visserligen inte hand om alla utmaningar som identifierats med de avfallsminskande åtgärderna eller informationshanteringen kring det. Den visar dock att det med relativt enkla medel redan nu går att få till en signifikant förbättring och förenkling vid framtida materialinventeringar och produktanalyser i avfallsminskande syfte. Det har i arbetet också framgått att mycket av det som presenterats är applicerbart även på andra produktkategorier än fönster

    År 2020 deklarerades covid-19 till en pandemi av World Health Organization. Pandemin kom att påverka sjöfarten i stor utsträckning och den här studienundersökerGöteborgs energihamn samt hamnens anlöpande tankfartygs rutiner under covid-19. För att ta reda på hur arbetet i energihamnen har förändrats under pandemin, användes semistrukturerade personintervjueroch en observation av en lossningsledarei arbete. Frågorna till intervjuernavar baserade på frågeställningarna: Hur har anlöp påverkats under covid-19?Hur ser hamnanlöp ut i normala fall? Finns det nya rutiner eller arbetssätt som kommer kvarstå efter pandemin? Informatörerna var allapersonal som dagligen jobbar i antingen hamnen eller på fartyg och behöver anpassa sitt arbete på grund av covid-19.Studien begränsades till Göteborgs energihamn samt anlöpande tankfartyg över 500 BRT. Hamnen och fartygen har åstadkommit ett effektivt och säkert arbete som uppfyller restriktionerna kring covid-19. Resultaten visar att ett hamnanlöp har på grund av den tvingade förändringen effektiviserats. Effektiviseringen har uppnåtts tack vare bättre förberedelser och digitalisering i det dagliga arbetet. Ytterligare enförändring under ett anlöp var att hamnpersonalen har så gott som helt slutat gå ombord på fartygen för att skriva dokument och checklistor. Slutsatserna visar att det är troligt att hamnen kommer fortsätta med det arbetssättsom används under pandemin även efter pandemins slut, med endast ett fåtal förändringar
