Highly enantioselective sulfa-Michael addition reactions using N-heterocyclic carbene as a non-covalent organocatalyst
We report the first asymmetric sulfa-Michael addition (SMA) reactions using a chiral N-heterocyclic carbene (NHC) as a non-covalent organocatalyst. We demonstrate that a triazolium salt derived NHC functions as an excellent Bronsted base to promote enantioselective carbon-sulfur bond formation. The reaction is applicable to a wide range of thiols and electrophilic olefins. Notably, quaternary chiral centers bearing both an S atom and a CF3 group were synthesized with excellent asymmetric control. Mechanistic studies suggest that the facial discrimination is likely to be guided by non-covalent interactions: hydrogen bonding and pi-pi stacking.National Natural Science Foundation of China [21372013]; Shenzhen Peacock Program [KQTD201103]SCI(E)[email protected]
Experimental Studies of Sulfa Drugs on Renal Function Report 2. Influences of Sulfadiazine on Renal Function
Renal complications of sulfadrugs are the result of poor solubility of these drugs in their acetyl derivatives and moreover, there is an intimate relationship between the degree of acetylation and the solubilities of the various sulfa drugs. Experimental studies have shown that sulfadiazine is absorbed rapidly after oral administration, gave relatively high blood concentration, leaves the blood tream slowly with less acetylation, and produced very little toxic reaction on tissues, especially in the urinary tract. In this studies, the author made an experiment of sulfadiazine on rabbits using the renal clearance test and obtained the following results. 1. The amounts of urine, GFR, RPF, RBF, hematocrit showed obviously decreases by the administration of 0.05 g, 0.10 g, 0.20 g per 1 kg weight of rabbits for 10 days. 2. The renal damages paralleled to the amount of sulfadiazine administrated on rabbits and acute renal insufficiency cound be observed
Asymmetric Sulfa-Michael Addition of ,-Unsaturated Esters/Amides Using a Chiral N-Heterocyclic Carbene as a Noncovalent Organocatalyst
We report an asymmetric sulfa-Michael reaction of ,-unsaturated amides and esters using a chiral N-heterocyclic carbene as the HOMO-raising organocatalyst. We discovered an interesting correlation between C-13 NMR shifts of substrates and ee of their products. More electron-deficient Michael acceptors afforded higher enantioselectivity.National Natural Science Foundation of China [21372013, 21572004]; Shenzhen Peacock Program [KQTD201103]; Disciplinary Development Program for Chemical Biology by the Shenzhen Municipal Development and Reform CommissionSCI(E)[email protected]; [email protected]
Drug reaction with eosinophilia and systemic symptom induced by carbamazepine: a case report
Drug reaction with eosinophilia and systemic symptoms (DRESS) syndrome is a severe, potentially life-threatening acute adverse drug reaction (ADR), typically characterized by a long latency period (2-6 weeks to 3 months) from drug exposure. DRESS syndrome is defined by the presence of fever, cutaneous eruption, lymphadenopathy, systemic or asymptomatic internal organ involvement (e.g. Hepatitis, carditis, interstitial nephritis, interstitial pneumonitis, etc.) and haematological abnormalities, mainly leucocytosis, eosinophilia and sometimes atypical lymphocytosis. There are around 50 culprit drugs which cause DRESS syndrome e.g. carbamazepine, phenytoin, allopurinol, sulfa derivatives, antidepressants, antiepileptics, non-steroidal anti-inflammatory drugs and antimicrobials. The incidence of DRESS syndrome has been estimated to be between 1 in 1,000 and 1 in 10,000 drug exposures. There are many reported cases of DRESS syndrome due to carbamazepine, phenytoin, vancomycin, levitiracitam, ceftriaxone etc. Author presented a case of DRESS syndrome by carbamazepin. RegiSCAR and Japanese consensus group have developed specific criteria for making the diagnosis of DRESS syndrome. The patient described here met the majority of criteria according to RegiSCAR scoring guidelines for a diagnosis of DRESS syndrome induced by carbamazepine. As per RegiSCAR diagnostic criteria author had concluded that this was a “Definite” case of DRESS (Drug Reaction with Eosinophilia and Systemic Symptom) syndrome induced by carbamazepine. Carbamazepine is most common broad-spectrum antiepileptic drugs so, this case report will raises awareness among physician to suspect DRESS syndrome in patients who present unusual complaints and skin findings after starting antiepileptic drugs

New applications of Ester/Amide surrogates in organic synthesis
2019 - 2020Masked esters/amides are scaffolds endowed with a great potential in the field of organic synthesis. This PhD
project has been conceived, in the context of non-covalent organocatalysis, with the aim to exploit unsaturated and saturated masked esters/amides as starting materials to accomplish the synthesis of
different classes of organic compounds in a one-pot fashion.Masked esters/amides exhibit some important features which make them valid substrates for
organocatalytic one-pot sequences. Firstly, the presence of a nitrogen-based heterocycle renders them more
reactive if compared with esters or amides: the heterocycle “steals” electron density from the molecule,
resulting in an enhanced electrophilicity at the β position of reagent a or an enhanced acidity of α-proton in
type b substrates. Secondly, nitrogen atoms of the heterocycle offer to these substrates more possibilities of
interaction with an organocatalyst through further H-bonds formation. This provides a major rigidity in the
transition state and a subsequent increase in the stereochemical outcome of the reaction. Finally, another
important property of these compounds, due to the ability of the aza-heterocycle as leaving group, is the
possibility to obtain ester or amide functionality through simple treatment with alcohols or amines via typical
addition/elimination mechanism (hence the name “ester/amide surrogates”).
We tried to achieve the stereocontrolled formation of carbon-carbon and carbon-heteroatom bonds to obtain
cyclic compounds of different nature and size (such as benzothiazepines and bicyclic pyrazolidinones) and
non-cyclic compounds, such as imines, β-aminoalcohols and nitrones.
In this doctoral thesis, the first stereoselective cascade sulfa-Michael/lactamization sequence for the synthesis
of cis- and trans-2,3-diaryl substituted 1,5-benzothiazepines has been developed, starting from α,β-unsaturated
N-acylpyrazoles and 2-aminothiophenols. The two steps are promoted by catalytic amounts of a readily
available bifunctional thiourea and p-toluenesulfonic acid, respectively. Our work provides access to both N unprotected diastereoisomers of the product with satisfactory results.
Moreover, we demonstrated that these products can be easily elaborated to prepare libraries of compounds for
biological tests. [edited by Author]XXXIII cicl
