Transition to turbulence in a qblique shock-wave/boundary-layer interaction at M=15
Direct numerical simulations are carried out for different forcing techniques to trigger transition during the interaction between an oblique shock-wave and a laminar boundary-layer at M = 1.5. Three forcing methods are used: a) forcing of oblique unstable modes, whose shape and behaviour are determined by the local linear stability theory, b) broadband free-stream acoustic disturbances, and c) a cold plasma flow control device. While the oblique-mode breakdown is dominant for low-amplitude forcing, long streaky structures drive the transition process in a high-amplitude disturbance environment. LES are also performed on the experimental setup by the Institute of Theoretical and Applied Mechanics (ITAM) from Novosibirsk State University with cold plasma actuation. As well as the disturbance type, the effect of Reynolds number and forcing amplitude will be investigated
Characterization of the Buzura suppressaria single-nucleocapsid nucleopolyhedrovirus genome : a (phylo)genetic study
Baculoviruses are attractive biological alternatives to chemical insecticides for insect pest control. So far, more than 600 baculoviruses have been isolated from different insect species which provide a rich resource for developing new viral insecticides. Most baculoviruses are host specific. Baculovirus nomenclature is based on the host from which the virus was first isolated and the taxonomy is based on the morphology of the occlusion body, polyhedra (nucleopolyhedroviruses = NPVs) or granula (granuloviruses = GVs). Until 1995, the NPVs had been further subdivided into single-nucleocapsid (S) and multiple-nucleocapsid (M) NPVs. Phylogeny studies based on the polyhedrin gene indicated that lepidopteran NPVs can be classified into two groups (I and II). Group II NPVs were found to encompass both SNPVs and MNPVs, whereas group I NPVs were composed of only MNPVs. Three Group I NPVs, Autographa californica (Ac) MNPV, Orgyia pseudotsugata (OpMNPV) and Bombyx mori (Bm) NPV, have been entirely sequenced and their genome organization was found to be very similar. Such detailed sequence information from other baculoviruses is scare particularly from SNPVs and GVs. The subject of this thesis is the Buzura suppressaria (Busu) NPV which was isolated from a Geometrid insect (tea moth) and has been used successfully as a bio-insecticide in China. Significant amount of sequencing was conducted on a genomic library of this virus which allowed us to conduct comparative investigations on the genome organization and phylogeny.First, the most conserved baculovirus gene, the polyhedrin gene, was identified by hybridization and then sequenced (Chapter 2). The location of the polyhedrin gene was used to set the zero point of the physical map of the viral genome. Phylogeny research based on the polyhedrin sequence placed BusuNPV into the Group II baculoviruses (see Chapter 8).Ecdysteroid UDP-glucosyltransferase (EGT) is an enzyme encoded by baculoviruses which delays larval molting and allows the virus to extend the production of large numbers of progeny virus. Deletion of the egt gene from the genome of baculoviruses results in an increased speed of kill and a reduced larval feeding activity and therefore enhances viral insecticidal activity. The strategy of deleting egt is now being adopted for the engineering of baculoviruses of economically important insect pests. In Chapter 3, the identification, sequencing and comparative analysis of the egt gene of BusuNPV is reported. This is the first egt gene characterized in a SNPV. Maximum parsimony analysis based on the amino acid sequence of baculoviruses EGTs confirmed that BusuNPV belongs to Group II baculoviruses.In order to study the genome organization of BusuNPV, a plasmid library covering the entire genome was constructed and the inserts were terminally or completely sequenced. In Chapter 4, the sequence analysis of the Hin dIII-J region (3.2 kb) of the BusuNPV genome revealed five ORFs ( pep , ORF117 , ctl , iap and sod ). The genomic arrangement of these ORFs in BusuNPV turned out to be significantly different from that of AcMNPV and other characterized baculoviruses. This indicates that BusuNPV may have a gene arrangement distinct from other baculoviruses identified so far.To further investigate the genomic organization and phylogenetic status of BusuNPV, a detailed physical map was constructed by extensive restriction enzyme analysis and Southern blot hybridization (Chapter 5). The size of the viral genome was estimated to be approximately 120.9 kb. About 43.5 kb of dispersed sequence information was generated from the plasmid library. Fifty-two ORFs, homologous to those of other baculoviruses, were identified and their location on the BusuNPV genome determined (Chapter 5). Although the gene content of BusuNPV, based on these 52 ORFs, is similar to that of AcMNPV, BmNPV and OpMNPV the arrangement is, however, significantly different from the latter, highly co-linear Group I NPVs. A new approach (GeneParityPlot) was developed to represent the differences in gene order among baculoviruses when limited sequence information is available. The method is useful to identify potential conserved gene clusters. The data obtained show that BusuNPV is a distinct baculovirus species and suggest that group II viruses may have a genome organization distinct from Group I viruses. It is concluded that the gene distribution along baculovirus genomes may be used as an independent parameter to study baculoviral phylogeny.Protein P10 is a major component of the fibrillar structures found in the cytoplasm and nucleus of infected insect cells. This small protein is involved in the release of polyhedra from infected cell-nuclei late in infection and plays a role in morphogenesis of the occlusion body. A putative p10 gene was identified for the first time in a SNPV (BusuNPV) based on its size and domain structures. Its function was determined in a 'swap assay' by using an AcMNPV p10 deletion mutant as an acceptor for the putative BusuNPV p10 gene (Chapter 6). The AcMNPV recombinant expressing the BusuNPV P10 formed fibrillar structures in the cytoplasm of Spodoptera frugiperda cells, but was unable to induce nuclear disintegration. This supports the current hypothesis that additional viral factors are required for this process.Polyhedrin is the major component of the occlusion body, which encapsulates the virions and protects them against physical and chemical decay. The gene swap assay was applied to investigate the specificity of the polyhedrin gene in the generation of occlusion bodies (Chapter 7). The BusuNPV polyhedrin gene expressed by AcMNPV resulted in the formation of normal occlusion bodies containing multiple-capsid virions. The occlusion bodies were as infective as normal AcMNPV suggesting that the occlusion process is not SNPV or MNPV specific. However, the recombinant AcMNPV expressing the polyhedrin gene of Spodoptera exigua (Se) MNPV, had an altered polyhedron morphology, pirmidal in shape, with few occluded virions. The infectivity was also lower than that of wild-type AcMNPV. These data suggest that the occlusion process is fine-tuned and may be dependent on one or more factors in addition to polyhedrin, that determine the size, shape and occlusion of virions.The genetic analysis compiled in this thesis shows that BusuNPV is a distinct baculovirus species. Alignment of various BusuNPV ORFs with baculovirus homologues and phylogenetic analyses indicated that SNPVs do not form a monophyletic group within the phylogenetic trees. The gene content of BusuNPV is very similar to that of AcMNPV, BmNPV and OpMNPV, suggesting that baculoviruses may have most of their genes in common. In addition to gene homology genome organization may also reflect the evolutionary history of baculoviruses. The molecular genetic studies of BusuNPV and their impact to future engineering of the virus for improved insecticidal activity are discussed (Chapter 8).</p

Agelosus auricomus is the only species of the genus in Taiwan. It can be distinguished from most Staphylinina in Taiwan by the mandibles each with a subdental indentation. However, this character convergently evolved in the genus Nelmanwaslus Smetana, 2006. So far, only N. ornatus Smetana, 2006 has been found in Taiwan and is newly reported in this paper. Agelosus auricomus can be easily distinguished from N. ornatus by the patches of golden-yellow tomentose pubescence on the elytra (Fig. 3). Bionomics. Based on an import of locality name into Google Earth from collection data (Smetana & Hu 2019), Ag. auricomus occurs from 20– 800 m. Apparently, the species only occurs in the lowlands or low mountains. The specimen from Guishan was collected from the foot of a mountain near Nanshi River (ĦBữ). Smetana & Hu (2019) reported a specimen that was collected from under a stone in a valley in mixed wood forest. Distribution. Agelosus auricomus is at present known from Taiwan, including Nantou County, New Taipei City, Taipei City and Yilan County (Smetana & Hu 2019 and this study). The additional specimen from Nantou indicates that this species is perhaps widespread in Taiwan. Apecholinus fraternus Fairmaire, 1891 (Fig. 4) Material examined. TAIWAN: Hsinchu County: 1 spec., Talutrail (±ẘIJǎ), 01.VIII.2015, leg. Y.-L. Lin (TARI). Nantou County: 1 male, Meifeng (đø), Renai To., 05-VI-2019, leg. W. Z. Wang (FSHc). Diagnosis. Apecholinus fraternus can be distinguished from all other species of Apecholinus in Taiwan by the lack of golden-yellow or grey tomentose pubescence on the body. Bionomics. Based on an import of locality name into Google Earth from collection data (Hayashi 1978, Smetana 2018, Smetana & Hu 2019), Ap. fraternus occurs from 1400–2600 m in Taiwan. Some specimens of this species were collected by pitfall traps, but nothing known about the habitat the traps were set in (Smetana 2018, Smetana & Hu 2019). Smetana & Hu (2019) reported a specimen collected from under a rock near a creek. Distribution. Apecholinus fraternus is widespread in mainland China including Guangxi, Guizhou, Henan, Hubei, Hunan, Shaanxi, Sichuan and Yunnan. The species also widespread in Taiwan including Chiayi County, Hsinchu County, Hualien County, Nantou County and Taichung City (Smetana 2018, Smetana & Hu 2019). The specimen from Hsinchu represents the first specimen from northern Taiwan. Remarks. O endomarketing como técnica interveniente no processo de acreditação do HU-UFSC e seus reflexos à cultura organizacional

Programa de Pós-Graduação em Administração UniversitáriaA gestão da qualidade nas últimas décadas ultrapassou as fronteiras da indústria, chegando às organizações comerciais e às prestadoras de serviços, dentre elas as organizações prestadoras de serviços de saúde. Dentro deste contexto, o modelo de Acreditação hospitalar emergiu como a ferramenta de gestão da qualidade, preconizando a padronização da assistência ao paciente ou cliente por meio da normatização e classificação dos hospitais em três níveis distintos, quais sejam: Nível 1; Nível 2; e Nível 3. Desta forma, em 1999, foi editado o primeiro Manual Brasileiro da Organização Nacional de Acreditação (ONA, 2010). Razões pelas quais, elaborou-se a presente dissertação no Programa de Mestrado Profissional em Administração Universitária, um estudo de caso realizado no Hospital Universitário Polidoro Ernany de São Thiago da Universidade Federal de Santa Catarina (HU-UFSC) sobre esta problemática. As variáveis analisadas foram: Acreditação Hospitalar, cultura organizacional e endomarketing. A amostra obedeceu a dois momentos distintos: inciamente foi realizada uma entrevista semiestruturada, intensiva e intencional, com o grupo de 14 dirigentes de primeiro escalão do HU-UFSC. Na sequência, aplicou-se um questionário estruturado e tratado de forma probabilística, do Tipo Likert, a 268 servidores que trabalham neste hospital. O período das coletas de dados foi de janeiro a março de 2012. As técnicas de tratamento dos dados foram quanti-qualitativas. Os dados quantitativos foram tabulados em planilha Excel e processados pelo programa SPSS®. Agregaram-se fontes secundárias que valorizassem a pesquisa. Conclui-se que o HU-UFSC é considerado pelos participantes da pesquisa um hospital de boa a muito boa qualidade de atendimento e assistência à saúde, mas requer algumas providências que enderecem à Acreditação Hospitalar Nível 1, tais como: reforçar a comunicação geral e, especialmente, o estreitamento das relações profissionais entre os dirigentes e os servidores. A ferramenta de comunicação mais sinalizada pelos participantes para contribuir no processo de sensibilização à Acreditação hospitalar foi a promoção de palestras sobre o referido programa e reuniões constantes a propósito deste tema. O estudo permitiu concluir ainda que o ambiente para a implantação da Acreditação Hospitalar, Nível 1, parece ser favorável para se iniciar este processo, desde que as técnicas e ferramentas de endomarketing sejam corretamente aplicadas, com o fito de remodelar de forma positiva a cultura organizacional endereçada ao Nível 1, do Manual Brasileiro de Acreditação Hospitalar (ONA, 2010).Quality management in the recent decades has exceeded the industry boundaries, reaching the commercial organizations and service providers, among them, organizations that provide health care services. Within this context, the hospital accreditation model has emerged as a tool for quality management, aiming at the standardization of patient care or client through the standardization and classification of hospitals into three distinct levels, wich are: Level 1, Level 2 and Level 3.Thus in 1999, it was published the first manual of the Brazilian National Accreditation Organizations (ONA, 2010). On these issues this master's dissertation was elaborated to the University of Professional Master of Business Administration, a study case conducted at the University Hospital Polidoro Ernany de São Thiago - Universidade Federal de Santa Catarina (HU-UFSC). The variables analyzed were: Hospital Accreditation, Organizational Culture and Internal Marketing. The sample followed two distinct moments: initially a semi-structured interview was conducted, intensive and intentional, with a group of 14 leaders first step at HU-UFSC. In the sequency, we applied a structured questionnaire and treated in a probabilistic manner, the Ukert Type, to 268 servers working in this hospital. The period of data collection was from January through March, 2012. The treatment techniques of quantitative and qualitative data were: Quantitative data formulated in an Excel spreadsheet and processed by SPSS®. And added to secondary sources that value the search. It is concluded that the UFSC-HU hospital is considered to have a good to a very good quality of health care and assistance by the research participants and requires some steps that addresses the Hospital Accreditation Level 1, such as strengthening the general communication and especially the strengthening of professional relationships between managers and servers, the communication tool most marked by the participants to contribute in the process of sensltl7atlon to Hospital Accreditation. Also it was mentioned the promotion of lectures on the program and constant meetings on the topic. It is thought that the environment for the implementation of the Hospital Accreditation, Level 1, seems to be favorable to stan this process, since the internal marketing techniques and tools are correctly applied, aiming on reshaping a positive culture addressed to an organizational level, the Brazilian Manual of Hospital Accreditation (ONA, 2010)
Gomory-Hu trees
Klasični Ford-Fulkersonov rezultat zlepi problema maksimalnega u-v pretoka in minimalnega u-v prereza med izbranima vozliščema u in v v omrežju - uteženem grafu. V diplomski nalogi se ukvarjamo s problemom Gomory-Hu drevesa, ki v eni sami drevesni strukturi hrani informacijo o vseh minimalnih prerezih v grafu. Natančneje, iskanje minimalnega prereza med poljubnima vozliščema u in v v omrežju lahko predstavimo z iskanjem drevesne povezave z najmanjšo vrednostjo prepustnosti na edini poti med istima vozliščema v Gomory-Hu drevesu. V delu implementiramo Gusfieldov algoritem za izračun Gomory-Hu drevesa in ga časovno ovrednotimo.
Zaradi velike časovne zahtevnosti izračuna Gomory-Hu drevesa implementiramo algoritem za dinamičen izračun novega drevesa iz obstoječega drevesa pri spremembi prepustnosti posamezne povezave v grafu G. Izkaže se, da je algoritem za izračun drevesa pri povečanju prepustnosti povezave v grafu G hitrejši v primerjavi z osnovnim algoritmom. V primeru zmanjšanja prepustnosti povezave ne pride do kvalitativnih razlik.The classical Ford-Fulkerson algorithm computes a maximum u-v flow and a minimum u-v cut between two selected nodes u, v from flow network - weighted graph. In this thesis we study Gomory-Hu trees which in one tree structure include information about all minimum in the graph. More precisely computing a minimum cut between a pair of nodes u and v nodes in flow network can be reduced to searching for an edge with smallest capacity in the unique u-v path in the Gomory-Hu tree. We implement and evaluate Gusfield algorithm for computing Gomory-Hu tree.
The presented algorithm for computing Gomory-Hu trees has relatively high time complexity, so we also implement an algorithm for dinamically computing Gomory-Hu trees following a capacity change in the graph. It turns out that in the case of increasing capacity the dynamic approach outperforms the basic algorithm. However, we measure no substantial improvement in the case of reducing capacity of an edge
Water-soluble mercury induced by organic amendments affected microbial community assemblage in mercury-polluted paddy soil
Mercury (Hg) pollution or organic amendments (OA) may individually induce changes in the microbial community of paddy soils. However, little is known regarding the interaction of Hg and OA and the effect of different OA applications on the microbial community assemblage in Hg-polluted paddy soil. A soil incubation experiment was performed by applying three organic amendments (OA), namely a food waste compost (FC), and its HA and FA, into an Hg-polluted paddy soil to examine the changes in the microbial community and merA/merB gene abundance. The results showed that the OA treatments promoted total (SOC) and dissolved organic carbon (DOC) in soils, which may harbor copiotrophic bacteria. The HA and FA treatments decreased microbial diversity and richness along with an increase of water-soluble Hg (WHg) through the complexation of DOC to Hg, which may be mainly attributed to the enhanced Hg biotoxicity to soil microbiome induced by the increased WHg under these two treatments. Additionally, the WHg enhancement also contributed to the increase of Hg-resistant bacteria and merA/merB gene abundance, and consequently, induced changes in the microbial community. These results indicated the interaction of Hg and different OA induced the variation of WHg fraction in paddy soil, which played a fundamental role in the distinct responses of the microbial community assemblage. Collectively, the application of FA and HA to Hg-polluted soil should be limited considering Hg risk to microbiome, and FC can be an alternative. (C) 2019 Elsevier Ltd. All rights reserved
