Highly efficient small interfering RNA delivery to primary mammalian neurons induces MicroRNA-like effects before mRNA degradation
The study of protein function in neurons has been hindered by the lack of highly efficient, nontoxic methods of inducing RNA interference in such cells. Here we show that application of synthetic small interfering RNA( siRNA) linked to the vector peptide Penetratin1 results in rapid, highly efficient uptake of siRNA by entire populations of cultured primary mammalian hippocampal and sympathetic neurons. This treatment leads to specific knock-down of targeted proteins within hours without the toxicity associated with transfection. In contrast to current methods, our technique permits study of protein function across entire populations with minimal disturbance of complex cellular networks. розкопок експедиції М. Корфмана в Трої, доходить висновку, що варто відійти від традиційного уявлення про сім
міст, які після своєї загибелі стали культурними шарами
й утворюють стратиграфічну колонку пагорба Гісарлик.
Натомість, це швидше образи міст, що виступають у
свідомості дослідників під час читання Іліади та сприйнятті археологічних матеріалів. Відтворенню згаданих
образів міст Троя, за якими проступають риси культурних та етнічних спільнот, присвячено дослідження. Мешканці міст Троя стали свідками й учасниками міграційних
рухів, що призвели до індоєвропеїзації Анатолії.В статье автор, обобщив выводы своих исследований 1980—1990-х гг. и учтя последние результаты раскопок экспедиции М. Корфмана в Трое, приходит к выводу, что традиционное представление о семи городах, ставших
после своей гибели культурными слоями холма Гисарлик, требует пересмотра. Это скорее образы городов, возникающих в сознании исследователей при чтении «Илиады» и восприятии археологических материалов. Воссозданию упомянутых образов городов Троя, за коими просматриваются черты археологических культур и этнических
общностей, посвящено исследование. Обитатели городов Троя стали свидетелями и участниками миграционных

Gq Protein-Coupled Membrane-Initiated Estrogen Signaling Rapidly Excites Corticotropin-Releasing Hormone Neurons in the Hypothalamic Paraventricular Nucleus in Female Mice
CRH neurons in the hypothalamic paraventricular nucleus (PVN) play a central role in regulating the hypothalamus-pituitary-adrenal (HPA) axis and are directly influenced by 17β-estradiol (E2). Although compelling evidence has suggested the existence of membrane-associated estrogen receptors (mERs) in hypothalamic and other central nervous system neurons, it remains unknown whether E2 impacts CRH neuronal excitability through this mechanism. The purpose of the current study is to examine the existence and function of mER signaling in PVN CRH neurons. Whole-cell recordings were made from CRH neurons identified by Alexa Fluor 594 labeling and post hoc immunostaining in ovariectomized female mice. E2 (100nM) rapidly suppressed the M-current (a voltage-dependent K(+) current) and potentiated glutamatergic excitatory postsynaptic currents. The putative Gq-coupled mER (Gq-mER) characterized in hypothalamic proopiomelanocortin neurons initiates a phospholipase C-protein kinase C-protein kinase A pathway; therefore, we examined the involvement of this pathway using selective inhibitors. Indeed, the ER antagonist ICI 182780 and inhibitors of Gq-phospholipase C-protein kinase C-protein kinase A blocked E2's actions, suggesting dependence on the Gq-mER. Furthermore, STX, a selective ligand for the Gq-mER, mimicked E2's actions. Finally, to examine the in vivo effect of Gq-mER activation, E2 or STX injection increased c-fos expression in CRH neurons in the PVN, suggesting CRH neuronal activation. This corresponded to an increase in plasma corticosterone. We conclude that the Gq-mER plays a critical role in the rapid regulation of CRH neuronal activity and the HPA axis. Our findings provide a potential underlying mechanism for E2's involvement in the pathophysiology of HPA-associated mood disorders.Peer reviewe

Previous issue date: 2020-05-14Low-level cloud cover over the Southern Ocean (SO) has a significant influence on global radiative balance. Climate models have had difficulty in the past reproducing shortwave radiation reflected by these clouds because of the inability to represent low-level cloud top properties, more specifically, they underrepresent cloud top supercooled liquid water concentrations. Past in-situ and satellite studies over the region have emphasized the abundance and importance of supercooled liquid water in cold sector SO clouds, especially at or near cloud top. SO supercooled clouds contribute between 27% and 38% of the total reflected solar radiation between 40ºS and 70ºS and represent a significant source of uncertainty in global climate models. Understanding these cloud’s properties and processes is critical to accurately simulating them in the climate system. Past global climate model studies have found that replacing all mixed-phase clouds with supercooled clouds at temperatures between -35ºC and 0ºC would result in 17 W m-2 more radiation reflected back to space. Ice crystals are typically thinner for the same liquid water content present as a pure supercooled liquid cloud.
This study uses collocated airborne radar, lidar, and thermodynamic data from twelve high-altitude flight legs during the SO Clouds, Radiation, Aerosol Transport Experimental Study (SOCRATES) field campaign to characterize SO cold sector cloud top phase (top 96 m) and cloud precipitation properties as a function of cloud top temperature. Training datasets were developed to create probabilistic phase classifications based on High Spectral Resolution Lidar data and HIAPER Cloud Radar data. These classifications were used to identify cloud top phase. Results indicate that nearly 88.7% of the cloud tops contained liquid, 1.1% contained ice, 0.8% could not be identified based on the training datasets, and 9.4% were unknown (meaning they fell outside of the training datasets). 75.6% of cloud cover had cloud top temperatures less than 0ºC. Considering only these clouds we found that 89.9% had supercooled liquid water at cloud top, 0.4% contained ice, 0.1% could not be identified based on the training datasets and 9.6% were unknown (meaning they fell outside the training datasets). Case studies are also presented illustrating examples of low-level cold sector cloud top phase identification.
Ice production in clouds with warm cloud top temperatures (greater than -5ºC) occurred in several locations during 8 research flights. A transient analysis of a shunt thyristor bridge excitation system for synchronous machines

The transient performance characteristics of the excitation system are important because they relate to: 1. Speed of response of the excitation system and magnitude of the generator field transient forcing which is one of the important aspects of a good excitation system. 2. Maximum possible transient overvoltages, affecting the insulation design of all excitation system components. These points are investigated for a particular case of transient operation of the excitation system. Construction and testing of components for the 6.5 m MMT adaptive optics system
In recent years astronomers have been pushing to build larger ground-based telescopes with apertures greater than 5 m in order to see deeper into space and resolve smaller objects. Realistically, while a larger telescope aperture allows more light to be collected, atmospheric turbulence caused by thermal gradients in the atmosphere limits the achievable resolution to a level comparable with apertures on the order of half a meter or less. Adaptive optics (AO) can be used to counteract the degrading effects of the atmosphere in real time and recover diffraction-limited resolution. With the help of AO, better science can be done, and as more large ground-based telescopes are built, the need for reliable AO systems grows. The 6.5 m upgrade to the Multiple Mirror Telescope (MMT) on Mt. Hopkins is an example of a large telescope project. An infrared adaptive optics system for this telescope is currently under construction at Steward Observatory in the Center for Astronomical Adaptive Optics. This dissertation reports on the design, construction, and testing of various components of this AO system with which the author was involved.
