    Thiol–disulfide balance and trace element levels in patients with seasonal allergic rhinitis

    Background: The prevalence of allergic diseases is gradually increasing worldwide. The most common such allergic disease is allergic rhinitis (AR). Objective: The present study investigated the possible relationship between seasonal AR and the thiol-disulfide balance and zinc and copper levels in adult individuals. Study Design and Methods: 130 male and female adults were included in the study. The participants’ serum thiol-disulfide balance and zinc and copper levels were measured spectrophotometrically using commercial kits. Statistical significance was accepted as p < 0.05 between the groups. Results: The serum copper (p = 0.001), native thiol (p = 0.006), reduced thiol (p < 0.001), and thiol oxidation reduction ratio (p < 0.001) levels were significantly lower in the seasonal AR group than in the control group. Conclusion: In AR patients, the low level of copper, which is an important trace element, the deterioration of the thiol-disulfide balance, which represents a unique indicator of the oxidant-antioxidant balance, the increased disulfide level caused by oxidative stress, and the decreased native thiol level can all serve as important biochemical markers.     Comparison of Growth Factor Levels in Injectable Platelet-Rich Fibrin Obtained From Healthy Individuals and Patients With Chronic Periodontitis: A Pilot Study

    Background This study aimed to assess and compare the concentrations of growth factors, white blood cells (WBCs), and platelets in injectable platelet-rich fibrin (i-PRF) derived from people with healthy periodontal conditions and those with chronic periodontitis.Methods Venous blood samples were obtained from 30 patients diagnosed with chronic periodontitis (test group) and 30 participants with healthy periodontal conditions (control group). The i-PRF was then acquired from centrifuged blood. The growth factors (VEGF, IGF-1, TGF-beta 1, PDGF-BB and EGF) released from the i-PRF samples were compared between groups with ELISA testing. The amounts of WBCs and platelets were also compared.Results No significant differences in the concentrations of growth factors were found between the groups (the mean values for the control and test groups were, respectively: IGF: 38.82, 42.46; PDGF: 414.25, 466.28; VEGF: 375.69, 412.18; TGF-beta 1: 21.50, 26.21; EGF: 138.62, 154.82). The test group exhibited a significantly higher WBC count than the control group (8.80 vs. 6.60, respectively). However, the platelet count did not show a statistically significant difference between the groups (control group 242.0 vs. test group 262.50). No significant correlation was observed between WBC count and growth factor level in either group.Conclusions The growth factor levels in i-PRFs did not exhibit significant difference between the two groups. This suggests that the levels of these growth factors may be unaffected by the periodontal disease.ALKU BAPNot applicable

    Evaluation of thiol/disulfide homeostasis and ischemia modified albumin as potential markers for periodontitis

    Abstract Background The current study aimed to assess the impact of periodontitis on oxidative stress parameters by examining serum total antioxidant capacity (TAS), total oxidant status (TOS), oxidative stress index (OSI), thiol/ disulfide homeostasis and ischemia modified albumin (IMA). Methods The study had 90 participants, categorized into 3 groups: Group 1: Periodontally healthy; Group 2: Stage II Grade B periodontitis; Group 3: Stage III and IV Grade B periodontitis. Demographic and periodontal variables were assessed. The levels of serum TAS, TOS, OSI, IMA, and thiol/disulfide were assessed. Results No significant differences in sex and age were detected among the groups (p > 0.05). When compared to Group 1, all clinical measurements were statistically significantly greater in Group 3 (p  0.05). The highest serum IMA value was observed in Group 3 (p = 0.037), whereas native thiol (p = 0.00), total thiol (p = 0.00) and disulfide values (p = 0.023) were highest in Group 1. Conclusions These findings indicate that thiol/disulfide homeostasis and IMA could hold promise as a potential biomarker of inflammation in periodontitis

    Improved catalytic activity by catalase immobilization using γ‐cyclodextrin and electrospun PCL nanofibers

There is limited research available on the change in enzyme activity following interaction with cyclodextrin. In this study, catalase enzyme was immobilized into nanofibrous structures by various techniques, with and without -CD addition, and the enzymatic activity of catalase was evaluated. In addition, catalase--CD complex containing PEO polymer solution was electrospun in between PCL nanofibrous layers as a newly developed technique. The enzyme immobilized nanofibrous structures were characterized by SEM, XRD, and FT-IR analysis methods. Among all the activity tests, best enzyme activity was recorded with catalase--CD physical mixture encapsulated PCL nanofibrous layers.     Can Inflammatory Indices Predict Myringosclerosis

    Objectives: To examine the relationships between the neutrophil-to-lymphocyte ratio (NLR), the platelet-to-lymphocyte ratio (PLR), and the systemic immune-inflammation index (SII) in myringosclerosis after ventilation tube insertion (VTI). Methods: We conducted this descriptive case-control study between March 2022 and September 2024. A total of 75 children aged 6 to 12 years with a history of VTI were enrolled. Myringosclerosis+ (n=43) included children with myringosclerosis, while Myringosclerosis-(n=32) included children without myringosclerosis. Preoperative values for NLR, PLR, and SII were collected by hematological testing and calculations. Demographic factors, prognostic risk factors and discrimination power of parameters were determined by statistical methods. Results: Compared to the myringosclerosis-, the myringosclerosis+ had significantly higher NLR, PLR, and SII values (p=0.00, p=0.04, and p=0.01, respectively). Receiver operating characteristic (ROC) curve area of NLR is 0,76 (95% CI: 0.65-0.87, p=0.00). Area under curve of SII is 0,62 (95%CI: 0.57-0.81, p=0.01). Neutrophil-to-lymphocyte ratio affects myringosclerosis development at 1.34 with sensitivity=0.63, specificity=0.78. Neutrophilto-lymphocyte ratio(NLR) and systemic immune inflammation index(SII) show myringosclerosis development at different powers of predictivity. The ROC area of NLR is highest of all parameters but is moderately predictive (<0.80). It is slight for SII. The NLR may be a marker of prediction of myringosclerosis OR=0.182 (95%CI:0.067-0.496). Conclusion: Neutrophil-to-lymphocyte ratio and SII are systemic inflammatory prognostic markers in myringosclerosis, and elevated NLR values are a predictor of myringosclerosis with modest affect. Other parameters should be sought in myringosclerosis development

    Açıklanamayan İnfertilitede Tiol-Disülfit Homeostazının Klinik Önemi

    Amaç: Açıklanamayan infertilite (UI), belirsiz bir nedene sahip çok yönlü bir üreme bozukluğudur. Oksidatif stres, patofizyolojisinde olası bir faktör olarak ortaya çıkmıştır. Bu çalışma, UI'li kadınlarda oksidatif ve antioksidan seviyeleri ile tiol ve disülfit seviyeleri arasındaki dengenin önemini değerlendirmeyi amaçlamıştır. Gereç ve Yöntem: UI'li 23 kadından ve 27 sağlıklı doğurgan kontrolden alınan serum örnekleri, toplam antioksidan durumu (TAS), toplam oksidan durumu (TOS), oksidatif stres indeksi (OSI), paraoksonaz-1 (PON), arilesteraz (ARES), tiol-disülfit homeostazı (TDH) ve iskemi modifiye albümin (IMA) açısından analiz edildi. Hematolojik ve rutin biyokimyasal parametreler de değerlendirildi. Seçili biyobelirteçlerin tanı performansını değerlendirmek için Receiver Operating Characteristic (ROC) eğrisi analizi yapıldı. Bulgular: UI grubunda TAS ve doğal tiol seviyeleri istatistiksel olarak anlamlı şekilde düşük; TOS, OSI, disülfit, oksitlenmiş tiol oranı ve IMA seviyeleri ise anlamlı şekilde yüksek bulundu. (p<0.001). ARES aktivitesi yükseldi, ancak PON aktivitesi ve toplam tiol konsantrasyonları önemli ölçüde farklılık göstermedi. TAS (AUC = 0,848) ve azaltılmış tiol oranı (AUC = 0,799), test edilen biyobelirteçler arasında en güçlü tanısal faydayı gösterdi. Sonuç: Oksidatif dengesizliğin UI patofizyolojisinde önemli bir rol oynadığı görülmektedir. TAS ve TDH ile ilişkili endeksler gibi biyobelirteçler değerli tanısal içgörüler sunabilir ve infertilite yönetiminde terapötik müdahale için potansiyel hedefler olarak hizmet edebilir
