A multilayered microfluidic blood vessel-like structure
There is an immense need for tissue engineered blood vessels. However, current tissue engineering approaches still lack the ability to build native blood vessel-like perfusable structures with multi-layered vascular walls. This paper demonstrated a new method to fabricate tri-layer biomimetic blood vessel-like structures on a microfluidic platform using photocrosslinkable gelatin hydrogel. The presented method enables fabrication of physiological blood vessel-like structures with mono-, bi- or tri-layer vascular walls. The diameter of the vessels, the total thickness of the vessel wall and the thickness of each individual layer of the wall were independently controlled. The developed fabrication process is a simple and rapid method, allowing the physical fabrication of the vascular structure in minutes, and the formation of a vascular endothelial cell layer inside the vessels in 3–5 days. The fabricated vascular constructs can potentially be used in numerous applications including drug screening, development of in vitro models for cardiovascular diseases and/or cancer metastasis, and study of vascular biology and mechanobiology.American University of Beirut (startup grant and University Research Board grant)National Council for Scientific Research (Lebanon)National Science Foundation (U.S.) (EFRI-1240443)Immodgel (602694)National Institutes of Health (U.S.) (EB012597, AR057837, DE021468, HL099073, AI105024, AR063745)National Institute of General Medical Sciences (U.S.) ( Award Number P20GM103638-04)King Abdulaziz City for Science and Technology (Grant No. 12-MED3096-3
Reduced Graphene Oxide Incorporated GELMA Hydrogel Promotes Angiogenesis for Wound Healing Applications
Non-healing or slow healing of chronic wounds is among the serious
complications of diabetes. The decrease in blood supply due to lack of angiogenesis
leads to reduced supply of oxygen, nutrients and growth factors resulting in decreased
proliferation and migration of endothelial cells involved in formation of blood vessels.
The absence of vascular network also decreases the number of fibroblasts and
keratinocyte cells in wounded area and therefore results in low collagen deposition in
diabetic wounds. In this study, we report the development of rGO nanoparticles
impregnated GelMa hydrogels where rGO nanoparticles were used for enhancing
angiogenesis and GelMa hydrogel was applied for promoting the growth of tissue
forming cells for wound healing applications. Gelatin methacrylate (GelMA) based
hydrogels loaded with different concentration of rGO nanoparticles were synthesized
by UV crosslinking. Characterization of rGO incorporated GelMA hydrogel retained
excellent porous structure and hydrophilic properties (porosity, degradation, and
swelling). In vitro cytotoxicity studies (Live/Dead and MTT assays), using three
different cell lines, confirmed that the hydrogels are biocompatible. The GelMA
hydrogel containing 2% rGO enhanced the proliferation of the cells (fibroblasts,
Endothelial cells, Keratinocytes) and significant wound healing was observed in wound
healing scratch assay. In vivo using the chick chorioallantoic membrane model showed
that the presence of rGO in the GelMA hydrogel significantly enhanced angiogenesis

Cell Microenvironment Engineering and Monitoring for Tissue Engineering and Regenerative Medicine: The Recent Advances
In tissue engineering and regenerative medicine, the conditions in the immediate vicinity of the cells have a direct effect on cells' behaviour and subsequently on clinical outcomes. Physical, chemical, and biological control of cell microenvironment are of crucial importance for the ability to direct and control cell behaviour in 3-dimensional tissue engineering scaffolds spatially and temporally. In this review, we will focus on the different aspects of cell microenvironment such as surface micro-, nanotopography, extracellular matrix composition and distribution, controlled release of soluble factors, and mechanical stress/strain conditions and how these aspects and their interactions can be used to achieve a higher degree of control over cellular activities. The effect of these parameters on the cellular behaviour within tissue engineering context is discussed and how these parameters are used to develop engineered tissues is elaborated. Also, recent techniques developed for the monitoring of the cell microenvironment in vitro and in vivo are reviewed, together with recent tissue engineering applications where the control of cell microenvironment has been exploited. Cell microenvironment engineering and monitoring are crucial parts of tissue engineering efforts and systems which utilize different components of the cell microenvironment simultaneously can provide more functional engineered tissues in the near future.Version of Recor
Development and characterization of nickel phosphorus based nanocomposite coatings for corrosion protection of steel
Corrosion is the major challenge faced by many industries like marine, automobile, oil and gas industry, etc. Nickel Phosphorus (Ni-P) based coatings are
extensively studied to mitigate corrosion due to their improved corrosion resistance. However, these coatings lack mechanical strength limiting their applications. In the present study, novel Ni-P-X (X=TiC (titanium carbide) and ZrC (zirconium carbide)) were developed through the electrodeposition process. Various amounts of titanium carbide TiC nanoparticles (0, 0.5, 1.0, 1.5, and 2.0 g/L) and ZrC nanoparticles (0, 0.75 and 1.5 g/L) were co electrodeposited in the Ni-P matrix under optimized conditions and then characterized by employing various techniques. It is noticed that the concentration of reinforcing ceramic particles has a significant on the structural, mechanical, tribological, and electrochemical properties of Ni-P nanocomposite coatings. The structural analysis of both types of prepared nanocomposite coatings indicates uniform, compact, and nodular structured coatings without any noticeable defects. Vickers microhardness and nanoindentation results of Ni-P-TiC nanocomposite coatings demonstrate the increase in the hardness with an increasing amount of TiC nanoparticles attaining its terminal value (5.98 GPa) at the concentration Corrosion is the major challenge faced by many industries like marine, automobile, oil and gas industry, etc. Nickel Phosphorus (Ni-P) based coatings are extensively studied to mitigate corrosion due to their improved corrosion resistance. Recent Advances in Application of Biosensors in Tissue Engineering
Biosensors research is a fast growing field in which tens of thousands of papers have been published over the years, and the industry is now worth billions of dollars. The biosensor products have found their applications in numerous industries including food and beverages, agricultural, environmental, medical diagnostics, and pharmaceutical industries and many more. Even though numerous biosensors have been developed for detection of proteins, peptides, enzymes, and numerous other biomolecules for diverse applications, their applications in tissue engineering have remained limited. In recent years, there has been a growing interest in application of novel biosensors in cell culture and tissue engineering, for example, real-time detection of small molecules such as glucose, lactose, and H2O2 as well as serum proteins of large molecular size, such as albumin and alpha-fetoprotein, and inflammatory cytokines, such as IFN-g and TNF-α. In this review, we provide an overview of the recent advancements in biosensors for tissue engineering applications.Version of Recor
AN ANGIOGENIC AND ANTIBACTERIAL GELMA MICRONEEDLE ARRAY PATCH FOR DIABETIC WOUND HEALING APPLICATIONS
Diabetic wounds represent a significant healthcare challenge due to their protracted healing processes and susceptibility to infection. Nitric oxide (NO) and oxygen have emerged as promising therapeutic agents for enhancing wound healing in diabetic patients due to their angiogenic and antibacterial properties respectively. In this study, we developed a GelMA (gelatin methacryloyl) microneedle array patch capable of delivering nitric oxide and oxygen to diabetic wounds simultaneously. Nitric oxide was delivered via S-nitrosoglutathione (GSNO) to promote angiogenesis, while calcium peroxide served as an oxygen-releasing agent to exert antibacterial effects. The developed microneedle (MNs) showed sufficient mechanical strength under compression test indicating a potential candidate for transdermal administration of drugs. In addition, the microneedle array patch demonstrated sustained release of nitric oxide and oxygen over a period of 180 minutes. In vitro Live/Dead assay demonstrated no significant cytotoxicity of blank and dual release GelMA MNs in human dermal fibroblast cell lines. Cell proliferation assay using MTT dye showed enhanced proliferation of cells when treated with dual release MNs samples. Finally, scratch assay performed on same cell lines exhibited enhanced migration when treated with dual release samples compared to control and blank samples. Overall, our findings suggest that the Nitric oxide and oxygen-releasing GelMA microneedle array patch holds great promise as an advanced therapeutic approach for diabetic wound healing, offering a synergistic combination of angiogenic and antibacterial effects to expedite the healing process and mitigate complications associated with diabetic wounds
