Metagenomic analysis of viral communities in (hado)pelagic sediments.
No full textIn this study, we analyzed viral metagenomes (viromes) in the sedimentary habitats of three geographically and geologically distinct (hado)pelagic environments in the northwest Pacific; the Izu-Ogasawara Trench (water depth = 9,760 m) (OG), the Challenger Deep in the Mariana Trench (10,325 m) (MA), and the forearc basin off the Shimokita Peninsula (1,181 m) (SH). Virus abundance ranged from 10(6) to 10(11) viruses/cm(3) of sediments (down to 30 cm below the seafloor [cmbsf]). We recovered viral DNA assemblages (viromes) from the (hado)pelagic sediment samples and obtained a total of 37,458, 39,882, and 70,882 sequence reads by 454 GS FLX Titanium pyrosequencing from the virome libraries of the OG, MA, and SH (hado)pelagic sediments, respectively. Only 24-30% of the sequence reads from each virome library exhibited significant similarities to the sequences deposited in the public nr protein database (E-value <10(-3) in BLAST). Among the sequences identified as potential viral genes based on the BLAST search, 95-99% of the sequence reads in each library were related to genes from single-stranded DNA (ssDNA) viral families, including Microviridae, Circoviridae, and Geminiviridae. A relatively high abundance of sequences related to the genetic markers (major capsid protein [VP1] and replication protein [Rep]) of two ssDNA viral groups were also detected in these libraries, thereby revealing a high genotypic diversity of their viruses (833 genotypes for VP1 and 2,551 genotypes for Rep). A majority of the viral genes predicted from each library were classified into three ssDNA viral protein categories: Rep, VP1, and minor capsid protein. The deep-sea sedimentary viromes were distinct from the viromes obtained from the oceanic and fresh waters and marine eukaryotes, and thus, deep-sea sediments harbor novel viromes, including previously unidentified ssDNA viruses

Full text linkHado Boirudo/Hado Rakku adalah sebuah novel karya Yoshimoto Banana yang terbit pada tahun 1999. Novel ini terdiri dari dua judul yang tidak ada keterkaitan cerita antara satu dengan lainnya. Cerita pertama berjudul Hado Boirudo, mengisahkan tentang tokoh Aku yang sedang mendaki gunung tepat di hari satu tahun setelah kematian pasangan lesbiannya yang bernama Chizuru. Dalam perjalan mendaki gunung tersebut ia mengalami malam yang panjang dimana ia dihantui oleh seorang hantu perempuan, serta mimpi – mimpi buruk tentang Chizuru. Dalam perjalanannya pula ia menghayati kisah masa lalunya tentang pengkhianatan Ibu Tirinya serta hubungannya yang tak berakhir baik dengan Chizuru. Sedangkan pada cerita Hado Rakku, mengisahkan tentang seorang perempuan sebagai tokoh Aku yang tengah dalam kondisi berkabung atas kejadian tragis yang menimpa kakak perempuannya bernama Kuni. Kuni mengalami koma akibat pendarahan otak karena terlalu sibuk menyelesaikan pekerjaan kantornya sebelum mengundurkan diri untuk mempersiapkan pernikahannya. Selama Kuni koma tokoh aku menjadi dekat dengan kakak dari tunangan Kuni. Namun hubungan mereka tidak dapat dilanjutkan karena rasa empati tokoh Aku terhadap Kuni.
Hado Boirudo/ Hado Rakku merupakan novel dengan tema dan plot yang ringan. Tokohnya yang sebagian besar perempuan menjadikan novel ini padat akan informasi keperempuan. Oleh karena itu, dalam analisis yang bertujuan untuk mengetahui relasi dan citra perempuan akan digunakan teori kritik sastra feminis. Melalui perantara teori kritik sastra feminis akan dijelaskan lebih dalam mengenai hubungan – hubungan yang dimiliki oleh tokoh – tokoh perempuan serta citra perempuan pada masing – masing tokoh perempuan.
Hasil analisis terhadap relasi pada tokoh – tokoh perempuan menunjukkan bahwa hubungan apapun yang dimiliki oleh perempuan dengan perempuan memiliki ikatan yang lebih kuat dibandingkan hubungan antara perempuan dengan laki – laki. Kemudian analisis terhadap citra perempuan menunjukkan bahwa relasi yang terjalin dalam yang dimiliki oleh perempuan akan menguras emosi perempuan dan mempengaruhi citra diri perempuan. Kemudian didapati pula bahwa kehilangan orang terdekat pada perempuan akan meningkatkan citra feminim pada diri perempuan
BODE-Index vs HADO-Score in Chronic Obstructive Pulmonary Disease: Which one to use in general practice?
No full textAbstract Background Forced expiratory volume in one second (FEV1) is used to diagnose and establish a prognosis in chronic obstructive pulmonary disease (COPD). Using multi-dimensional scores improves this predictive capacity.Two instruments, the BODE-index (Body mass index, Obstruction, Dyspnea, Exercise capacity) and the HADO-score (Health, Activity, Dyspnea, Obstruction), were compared in the prediction of mortality among COPD patients. Methods This is a prospective longitudinal study. During one year (2003 to 2004), 543 consecutively COPD patients were recruited in five outpatient clinics and followed for three years. The endpoints were all-causes and respiratory mortality. Results In the multivariate analysis of patients with FEV1 1 ≥ 50%, statistically significant differences were observed across HADO categories for all-cause and respiratory mortality, while differences were observed across BODE categories only in all-cause mortality. Conclusions HADO-score and BODE-index were good predictors of all-cause and respiratory mortality in the entire cohort. In patients with severe COPD (FEV1 1 ≥ 50%), the HADO-score was as good a predictor of respiratory mortality as the BODE-index. These differences suggest that the HADO-score and BODE-index could be used for different patient populations and at different healthcare levels, but can be used complementarily.</p
