    M-Quantile and expectile random effects regression for multilevel data

    The analysis of hierarchically structured data is usually carried out by using random effects models. Theprimary goal of random effects regression is to model the expected value of the conditional distributionof an outcome variable given a set of explanatory variables while accounting for the dependence structureof hierarchical data. The expected value, however, may not offer a complete picture of this conditionaldistribution. In this paper we propose using linear M-quantile regression, to model other parts of theconditional distribution of the outcome variable given the covariates. The proposed random effectsregression model extends M-quantile regression and can be viewed as an alternative to the quantilerandom effects model. Inference for estimators of the fixed and random effects parameters is discussed.The performance of the proposed methods is evaluated in a series of simulation studies. Finally, wepresent a case study where M-quantile and expectile random effects regression is employed for analyzingrepeated measures data collected from a rotary pursuit tracking experiment

    m(1)A and m(6)A modifications function cooperatively to facilitate rapid mRNA degradation

    N-6-Methyladenosine (m(6)A), the most abundant internal mRNA modification, affects multiple steps in gene expression. Mechanistically, the binding of YTHDF2 to m(6)A on mRNAs elicits rapid mRNA degradation by re-cruiting several RNA degrading enzymes. Here, we show that N-1-methyladenosine (m(1)A), another type of RNA modification, accelerates rapid m(6)A RNA degradation. We identify HRSP12 as an RNA-binding protein that recognizes m(1)A. The binding of HRSP12 to m(1)A promotes efficient interaction of YTHDF2 with m(6)A, consequently facilitating endoribonucleolytic cleavage via the RNase P/MRP complex. Transcriptomewide analyses also reveal that mRNAs harboring both m(1)A and m(6)A are downregulated in an HRSP12-dependent manner compared with mRNAs harboring m(6)A only. Accordingly, a subset of endogenous circular RNAs that harbor m(6)A and associate with YTHDF2 in an HRSP12-dependent manner is also subjected to m(1)A-facilitated rapid degradation. Together, our observations provide compelling evidence for crosstalk between different RNA modifications.

    Minor Histocompatibility Antigen Ha-1 and Ha-2 Polymorphisms in Taiwan: Frequency and Application in Hematopoietic Stem Cell Transplantation

    Background: Minor histocompatibility antigens influence the occurrence of graft-vs.-host disease and graft-vs.-leukemia effects after hematopoietic stem cell transplantation (HSCT) . We determined the population frequencies of HA-1 and HA-2 alleles in Taiwan and exploited their potential applications in allogeneic HSCT. Methods: HA-1 and HA-2 were genotyped using polymerase chain reaction and restriction fragment length polymorphism in healthy controls (221 for HA-1 and 306 for HA-2) and HLA- matched donor-recipient sibling pairs with HSCT (92 for HA-1 and 38 for HA -2). The association of genetic polymorphisms with HSCT outcome was evaluated by univariate and multivariate analyses. Results: The allele frequencies in controls were 35.3% and 64.7% for HA-1(H) and HA-1(R), and 89.0% and 11.0% for HA-2(V) and HA-2(M), respectively. HA-1 disparity was denoted in 16.3% of HLA- matched donor-recipient sibling pairs, while it was not associated with HSCT outcome. HA-2 disparity was not observed in the donor-recipient pairs studied. The possibilities of using HA-1 and HA- 2 variabilities as molecular markers for hematopoietic chimerism after HSCT were 39.2% and 18.4%, respectively. Conclusions: Our data provide the information on allele and genotype frequencies of HA-1 and HA-2 in a Taiwanese population, and suggest that prospective genomic typing for HA-1 and HA-2 alleles of the donor and recipient could be a useful approach for molecular identification of hematopoietic chimerism after HSCT, rather than prognosis of clinical outcome

    Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods
