Cytogenetic and DNA-fingerprint characterization of choriocarcinoma cell lines and a trophoblast/choriocarcinoma cell hybrid
We report the successful fusion of human choriocarcinoma cells with normal human trophoblast cells to a choriocarcinoma/trophoblast hybrid. The hybrid cells ACH1P were derived from fusion of primary male trophoblast cells with the HGPRT-defective choriocarcinoma cell line ACI-I. The karyotypes of the parental choriocarcinoma cell line JEG-3, its HGPRT-defective mutant clones AC1-1, ACZ-5, and AC1-9, and the choriocarcinoma/trophoblast hybrid ACH1P are presented, together with a detailed characterization of the AC1-specific chromosomal marker add(X)(q26) using conventional cytogenetic banding techniques and multiplex-fluorescence in situ hybridization (M-FISH). To our knowledge, this is the first report of a stably proliferating human cell hybrid of trophoblastic origin, providing a unique cell culture model to study trophoblast-related invasion and ifs underlying genetic mechanisms. Topological features of lithium disilicate glass-ceramics uncovered through materials informatics

Dental Materials , (2025); https://doi.org/10.1016/j.dental.2025.09.004.Objective: The aim of this study was to inversely predict the topological features underlying SEM images from arbitrary biaxial flexural strengths of glass-ceramics by Materials Informatics (MI) approach. Methods: The scanning electron microscopic (SEM) image and in vitro biaxial flexural strength of 10 commercially available/experimental glass-ceramics were collected. The total of 200 SEM images were prepared as input data. Topological features underlying the SEM images were extracted using persistent homology analysis and compressed using principal component analysis. Gaussian mixture regression was employed to develop a machine learning model for predicting biaxial flexural strength based on the topological features. Arbitrary biaxial flexural strengths (390, 411, 442, 478, 515, 564, 597, 610, and 640 MPa) were defined, and an inverse analysis was conducted with the constructed machine learning model to overlay topological features onto SEM images. Results: The topological features were compressed into 18 principal components. The machine learning model was selected and optimized based on the Bayesian Information Criterion. Using the constructed machine learning model, the biaxial flexural strengths were predicted with a test score of 72 % (Root Mean Squared Error: 53.5, Mean Absolute Error: 40.3). From the arbitrary biaxial flexural strengths, topological features were inversely predicted and overlaid onto SEM images. Conclusion: The inverse analysis established in this study successfully predicted the topological features on SEM images of glass-ceramics from the biaxial flexural strengths. Cognitive-behavioral therapy modifies the naturalistic course of social anxiety disorder: findings from an ABA design study in routine clinical practices
AIMS: While randomized evidence appears to have established efficacy of cognitive-behavioral therapy (CBT) and some pharmacotherapy for social anxiety disorder (SAD), their real-world effectiveness has been called into question by long-term naturalistic cohort studies of patients with SAD as they show very low probability of recovery and sustained social dysfunctions despite some drug and psychological therapies. METHOD: The present study examines the effectiveness of group CBT for SAD in real-world settings (n=62) by examining the course of patients' symptomatology and social functions through approximately 6 months on the waiting list, through 6 months receiving the manualized group CBT intervention consisting of 16 2-h sessions, and for 12 months after the treatment. RESULTS: We found: (i) that the patients with SAD changed little or possibly worsened through the 6 months on the waiting list, although two in three of them were on antidepressants, benzodiazepines or both; (ii) that both their symptomatology and social function improved significantly and substantively through the group CBT; and (iii) that this improvement was maintained through the 3- and 12-month follow ups. CONCLUSIONS: We can implement and must disseminate evidence-based, effective CBT for more patients with SAD to lessen their suffering and stop the perpetuation of their symptoms.Toshi A. Interpretations of "Thusness" (zhenru) : Sanskrit tathatā and Chinese benwu, ru, ruru and zhenru
With special attention to the term "thusness (zhenru), " the present article attempts to explore the Chinese way of thinking which is firmly based on Classical Chinese and Chinese culture. Tracing the transition of Chinese Buddhist translations of Sanskrit tathatā (suchness), namely "benwu (the original nothingness), " "ru-ru (the true state of reality)" and "zhenru (thusness or suchness), " the author emphasizes the significance of the two-Sinograph fundamental terms of Chinese Buddhism such as zhenru (lit. "true and certain"), ru-shi ("thus"/"in this way" ; Skt. evam), shisu ("worldly"/"conventional" ; Skt. samvrti), rulai ("Thus Come [One]" ; Skt. tathāgata). All these two-character words can be divided into each Sinograph as possessing the synonymous, yet notably differentiated connotation ; as a result, when bound into one term, they broaden the original meanings as applicable to wide contexts. The Tang Buddhist commentators Kuiji (632-82), Fazang (643-712), Chengguan (738-839) and Zongmi (780-841) and so forth explained zhenru in several ways, by dividing the single term zhenru into zhen and ru and reuniting them. This type of Chinese Buddhist interpretation was obtained by newly accepting the Indian Buddhist exegetics called nirukti or nirvacana (folk etymology in Sanskrit), on the one hand, and adopting the preceding Chinese orthodox scholarship as found in Ruist commentaries and historiographies as well as exegetical dictionaries such as the Shiming "Explaining Terms" composed by Liu Xi in the Later Han. Lack of Transcription Triggers H3K27me3 Accumulation in the Gene Body
SummaryTrimethylated H3K27 (H3K27me3) is associated with transcriptional repression, and its abundance in chromatin is frequently altered in cancer. However, it has remained unclear how genomic regions modified by H3K27me3 are specified and formed. We previously showed that downregulation of transcription by oncogenic Ras signaling precedes upregulation of H3K27me3 level. Here, we show that lack of transcription as a result of deletion of the transcription start site of a gene is sufficient to increase H3K27me3 content in the gene body. We further found that histone deacetylation mediates Ras-induced gene silencing and subsequent H3K27me3 accumulation. The H3K27me3 level increased gradually after Ras activation, requiring at least 35 days to achieve saturation. Such maximal accumulation of H3K27me3 was reversed by forced induction of transcription with the dCas9-activator system. Thus, our results indicate that changes in H3K27me3 level, especially in the body of a subset of genes, are triggered by changes in transcriptional activity itself

Reconstruction of low hairline microtia of Treacher Collins syndrome with a hinged mastoid fascial flap
Treacher Collins syndrome (TCS) is a rare genetic disorder leading to congenital craniofacial malfoiniations. Although this syndrome presents with various symptoms, corrective surgery for bilateral microtia with low hairline is one of the most challenging operations given the complex contours of the external ear. In this technical note, a novel, simple procedure for dealing with the low hairline by using a hinged mastoid fascial flap simultaneously with costal cartilage grafting is described. Several techniques for the reconstruction of low hairline microtia have been reported previously, such as skin graft, skin flap, and tissue expander, but the high number of repeat operations and residual scars remain problematic. As a simultaneous procedure with framework grafting, the use of a temporoparietal flap with skin grafting is popular; however, its drawbacks include the operative scar, decreased hair growth, and hair thinning Patients with TCS show anatomical variations of the superficial temporal vessels supplying the temporoparietal flap. In contrast, due to the high vascularity of the mastoid fascia, the mastoid fascial flap can be elevated safely and easily as an anteriorly, posteriorly, superiorly, or inferiorly based flap
Transarterial Embolization and Transmucosal Sclerotherapy That Led to Successful Deliveries in a Patient With Symptomatic Arteriovenous Malformation of the Tongue
Patients with arteriovenous malformations (AVMs) are at risk of significant hemorrhage and AVMs are especially difficult to manage in those desiring future pregnancy. Few patients with successful deliveries have been previously reported. The authors report an unusual case of AVM of the tongue in a pregnant woman who presented with massive pulsatile bleeding from a ruptured artery in the tongue in late gestation, this was thought to be caused by the changes in hormonal balance and the increase in cardiac output. The bleeding was controlled with transarterial embolization and transmucosal absolute ethanol sclerotherapy. Furthermore, her second and third deliveries were successfully managed. The authors managed symptomatic tongue AVM by combining transarterial embolization and transmucosal sclerotherapy, which was followed by successful deliveries. This patient supports the utility of transmucosal absolute ethanol sclerotherapy for tongue AVM and multidisciplinary medical care for a successful delivery
