Kupffer cells (KC) play major roles in immunity and tissue injury or repair. Because recapitulation of KC biology and function within liver will allow superior insights into their functional repertoire, we studied the efficacy of the cell transplantation approach for this purpose. Mouse KC were isolated from donor livers, characterized, and transplanted into syngeneic recipients. To promote cell engraftment through impairments in native KC, recipients were preconditioned with gadolinium chloride. The targeting, fate, and functionality of transplanted cells were evaluated. The findings indicated that transplanted KC engrafted and survived in recipient livers throughout the study period of 3 months. Transplanted KC expressed macrophage functions, including phagocytosis and cytokine expression, with or without genetic modifications using lentiviral vectors. This permitted studies of whether transplanted KC could affect outcomes in the context of acetaminophen hepatotoxicity or hepatic ischemia-reperfusion injury. Transplanted KC exerted beneficial effects in these injury settings. The benefits resulted from cytoprotective factors including vascular endothelial growth factor. In conclusion, transplanted adult KC were successfully targeted and engrafted in the liver with retention of innate immune and tissue repair functions over the long term. This will provide excellent opportunities to address critical aspects in the biogenesis, fate, and function of KC within their native liver microenvironment and to develop the cell and gene therapy potential of KC transplantation

    No full text
This work aims at developing a better understanding towards the process of Video Content Analysis. Video analysis is basically how the computer interprets any video. This process of analyzing is completed in 4 steps namely, feature extraction, Structural Analysis, Clustering and Indexing, Browsing and Retrieval. In this paper, we have included various algorithms to understand how each of the above steps is applied in analyzing of any of the sports video. For example, in cricket we first extract the various features by attacking on the most often occurring color, i.e. ground color. Then, we divide the video according to various high points e.g., sixes and fours. Finally we cluster these divisions and arrange them in a table of contents for users to browse and retrieve the video part they require

The objective of this paper is to classify Brain Computer Interfacing (BCI) competition III, dataset IIIA with low computational complexity and high classification accuracy rate. This study presents the investigation of self-filtering (SF) as a pre-processing technique and Wigner trispectrum as feature extraction technique followed by the proposed [Formula: see text]-means clustering SVM (KC-SVM) classifier. KC-SVM is an improved form of classifier, which comprises KC-SVM with 10-fold cross validation technique. To analyze each stage of classification, different evaluation indexes of respective methods are considered. Substantial improvement on the performance of proposed classification method is scrutinized. Performance measurement accuracy 98.1%, Kappa coefficient 0.94, mutual information of feature vectors 0.14 and Fisher score 6.5 are achieved for considered dataset. In summary, this paper demonstrates that the classification accuracy of proposed classifier together with SF and Wigner–Ville Trispectrum (WT) satisfies the required criteria of classification. </jats:p

Abstract B13: Synthetic vulnerabilities in MLL3 deficient pancreatic tumors

Abstract Myeloid/lymphoid or mixed-lineage leukemia 3 (MLL3) is a histone 3-lysine 4 methyltransferase, frequently mutated in a variety of solid tumors including pancreatic ductal adenocarcinoma (PDAC), a nearly lethal disease due to our current inability to therapeutically target major oncogenic proteins driving this cancer. Like other tumor suppressor genes, direct targeting of MLL3 is not feasible, and thus our objective was to find biological pathways which are synthetic lethal targets in MLL3-deficient PDAC tumors. Identification of drug targets in such a specific genetic context could potentially dictate stratification of patients based on the mutational profile of their tumor. To achieve this, we generated a novel genetically engineered mouse model (GEMM) of pancreatic cancer, in which targeted mutation of Kras (G12D) and loss of functional MLL3 was driven by Cre-recombination from early pancreatic lineage-specific transcription factor, Pdx1. Our unpublished data suggests that loss of MLL3 function in exocrine pancreas cooperates with mutant Kras to accelerate the progression of invasive pancreatic neoplasia. Using cell lines generated from spontaneous tumors arising in KC (KrasG12D alone) and KMC (KrasG12D; MLL3δ/δ) mice, we performed functional genomic analyses and found several key oncogenic pathways upregulated in MLL3-deficient cells. We selected two well-known oncogenic pathways and through a series of cellular and biochemical assays, further validated their selectively activation in KMC cells, in contrast with KC cells. Next, we picked multiple pharmacological inhibitors of these pathways, which are already approved at various phases of clinical trials, and tested for their efficacy on growth of KC and KMC cells in both two- and three-dimensional growth assays. In agreement with our data showing preferential activation in KMC cells, we found KMC cells to be significantly more sensitive than KC cells to the effect of these inhibitors. Thus, identification of synthetic lethal hits in context of MLL3 functional loss could potentially enable us to design strategies to limit metastasis in patients with surgically-resectable tumors. ABSTRACT Housing is a basic need besides food and clothing. The role of banking greatly affects the economic activities of a country. Providing housing financing is one of the most basic bank businesses, one of which is providing housing financing by BRK Syariah. Before providing financing the bank needs to assess customers who apply for financing. Because, not a few customers fail to fulfill their obligations in house payments which results in problematic financing. In resolving problematic financing, settlement is not always carried out through court channels, but also efforts to resolve problems carried out outside the court provided by BRK Syariah. The formulation of the problem in this research is how to review the law regarding the resolution of problematic home ownership financing at BRK Syariah KC Arifin Ahmad Pekanbaru and what factors cause problematic home ownership financing at BRK Syariah KC Arifin Ahmad Pekanbaru. The type of research used is included in the empirical group and the sample for this research is one part of the BRK Syariah bank KC Arifin Ahmad Pekanbaru, namely the Management Director and several customers who have been declared in default or failed to carry out their obligations. From the results of research that has been conducted by the author, that the settlement of problematic financing of home ownership in BRK Syariah KC Arifin Ahmad Pekanbaru is litigation and non-litigation, such as rescheduling, reconditioning and restructuring and there are 2 (two) factors causing the financing of problematic home ownership in BRK Syariah KC Arifin Ahmad Pekanbaru, namely internal factors originating from the bank that are not careful both from analyzing prospective customers, as well as in the calculation of home financing and ext from the bank that is not careful both from analyzing prospective customers, as well as in the calculation of rumaa financing and external factors from the customer, where there are some customers who do not make payments because there is an element of intentionality and an element of customer unintentionality. Transcriptome profile in Drosophila Kc and S2 embryonic cell lines

    No full text
Drosophila melanogaster cell lines are an important resource for a range of studies spanning genomics, molecular genetics, and cell biology. Amongst these valuable lines are Kc167 (Kc) and Schneider 2 (S2) cells, which were originally isolated in the late 1960s from embryonic sources and have been used extensively to investigate a broad spectrum of biological activities including cell-cell signaling and immune system function. Whole-genome tiling microarray analysis of total RNA from these two cell types was performed as part of the modENCODE project over a decade ago and revealed that they share a number of gene expression features. Here, we expand on these earlier studies by using deep-coverage RNA-sequencing approaches to investigate the transcriptional profile in Kc and S2 cells in detail. Comparison of the transcriptomes reveals that ∼75% of the 13,919 annotated genes are expressed at a detectable level in at least one of the cell lines, with the majority of these genes expressed at high levels in both cell lines. Despite the overall similarity of the transcriptional landscape in the two cell types, 2,588 differentially expressed genes are identified. Many of the genes with the largest fold change are known only by their CG designations, indicating that the molecular control of Kc and S2 cell identity may be regulated in part by a cohort of relatively uncharacterized genes. Our data also indicate that both cell lines have distinct hemocyte-like identities, but share active signaling pathways and express a number of genes in the network responsible for dorsal-ventral patterning of the early embryo. © The Author(s) 2023.
