    Effect of Sphingosine-1-phosphate on trophectoderm differentiation in bovine embryos

    O desenvolvimento inicial do embrião é marcado pelo primeiro evento de diferenciação celular que define massa celular interna (MCI) e trofectoderma (TE). Em camundongos, a glicose (GLC) é essencial para esse processo e a tensão de oxigênio (O2) também interfere na formação de TE e ainda, há evidências em embriões e em células cultivadas de que a esfingosina-1-fosfato (S1P) pode regular as vias envolvidas neste evento. Nossa hipótese é que a glicose é necessária, a maior tensão de O2 é prejudicial para a diferenciação do TE e que o S1P induz a diferenciação do TE em embriões bovinos produzidos in vitro. Este estudo teve dois objetivos relatados em dois capítulos: 1. Avaliar o efeito da presença (+GLC) ou ausência (-GLC) de GLC em diferentes níveis de O2 (5% ou 20%) na formação do TE em embriões bovinos; 2. Estudar o papel da estimulação S1P na diferenciação de TE adicionando S1P ou o inibidor do receptor S1P, JTE-013. Inicialmente, os embriões foram submetidos aleatoriamente aos tratamentos no dia 1, em delineamento fatorial 2x2, considerando GLC (presente/ausente) e O2 (baixa/alta tensão). No segundo capítulo, usou-se apenas o modelo sem GLC em dois experimentos diferentes. O primeiro experimento consistiu no tratamento com S1P, enquanto o segundo visou a inibição da via com tratamento com JTE-013. As taxas de formação de blastocistos e os blastocistos foram obtidos 180 horas pós-inseminação. Realizou-se para todos os experimentos contagens de células totais, TE (GATA3-positivas) e MCI (GATA3-negativas), bem como análise de qRT-PCR para GATA3, YAP1, SOX2, CDX2, TFAP2C e OCT4. As taxas e contagens de células foram analisadas por ANOVA, enquanto para qRT-PCR foi considerado um modelo linear misto. No primeiro estudo, houve efeito do O2 nas taxas de clivagem, mas não na taxa de blastocistos. Houve interação entre GLC e O2 na contagem de células da MCI, expressão de CDX2 e expressão de SOX2. No segundo capítulo, houve um aumento na taxa de desenvolvimento causada por S1P em comparação ao controle e NaOH, embora não houve alterações induzidas por JTE-013. Células totais e células da MCI estavam aumentadas no grupo S1P em relação ao controle e NaOH. A expressão de CDX2 aumentou nos grupos S1P e NaOH, enquanto TFAP2C diminuiu no grupo S1P. A adição do JTE-013 não interferiu na expressão gênica ou contagem celular. Em conclusão, a diferenciação de TE ocorreu em ausência de glicose e, curiosamente, esta condição aumentou a expressão do gene relacionado ao TE CDX2 quando em maior tensão de O2. Ainda, S1P não modulou positivamente a diferenciação do TE, apesar de aumentar CDX2, mas induziu aumento no número total de células e no número de células MCI, sugerindo uma ação diferente da S1P em embriões bovinos.The early development of the embryo is marked by the first cellular differentiation that defines the inner cell mass (ICM) and the trophectoderm (TE). In mice, glucose (GLC) is essential for this process and oxygen tension (O2) also interferes with the formation of TE and there is evidence in embryos and in cultured cells that sphingosine-1-phosphate (S1P) can regulate the pathways involved in this event. We hypothesize that glucose is necessary and higher O2 tension is detrimental to TE differentiation, and that S1P induces TE differentiation in bovine embryos produced in vitro. This study had two objectives reported in two chapters: 1. To evaluate the effect of the presence (+GLC) or the absence (-GLC) of GLC at different O2 levels (5% or 20%) on the formation of the TE in bovine embryos; 2. to study the role of S1P stimulation in TE differentiation by adding S1P or the S1P receptor inhibitor, JTE-013. Initially, the embryos were randomly subjected to treatments on day 1, in a 2x2 factorial design, considering GLC (present/absent) and O2 (low/high voltage). In the second chapter, only the model without GLC was used in two different experiments. The first experiment consisted of treatment with S1P, while the second aimed to inhibit the pathway with JTE-013 treatment. Blastocyst formation rates and blastocysts were obtained 180 hours post-insemination. Total cell counts, TE (GATA3-positive) and MCI (GATA3-negative) were performed for all experiments, as well as qRT-PCR analysis for GATA3, YAP1, SOX2, CDX2, TFAP2C and OCT4. Rates and cell counts were analyzed by ANOVA, while for qRT-PCR a linear mixed model was considered. In the first study, there was an effect of O2 on cleavage rates, but not on blastocyst rates. There was an interaction between GLC and O2 on ICM cell count, CDX2 expression and SOX2 expression. In the second chapter, there was an increase in the rate of development for S1P compared to control and NaOH, although there were no changes to JTE- 013. Total cells and ICM cells were increased in the S1P group compared to control and NaOH. CDX2 expression increased in the S1P and NaOH groups, while TFAP2C decreased in the S1P group. The addition of JTE-013 did not interfere with gene expression or cell counts. In conclusion, TE differentiation occurred in the absence of glucose and, interestingly, this condition increased the expression of the TE-related gene CDX2 when exposed to greater O2 tension. Furthermore, S1P did not positively modulate TE differentiation, despite increasing CDX2, but induced an increase in the total number of cells and in the number of ICM cells, suggesting a different action of S1P in bovine embryos

    Replacement of fetal bovine serum in oocyte maturation medium transfer of improved freezing of in vitro produced bovine embryos

    A congelação de embriões, aliada a produção in vitro de embriões (PIVE), é uma ferramenta importante para disseminação de genética de alto valor na bovinocultura. Entretanto, esta ferramenta poderia ser ainda mais disseminada com a melhoria dos resultados de prenhez com uso do método de transferência direta (DT do inglês direct transfer). Os resultados inconsistentes da DT podem ter influência da adição de soro fetal bovino (SFB) durante a maturação oocitária, visto que influencia a composição lipídica dos blastocistos gerados na PIVE. Portanto, o objetivo do presente estudo foi avaliar e comparar os benefícios da substituição do SFB por outra fonte de lipídeos durante a maturação in vitro (MIV). A hipótese do trabalho foi que a adição de fase apolar ou polar do SFB em substituição ao SFB integral durante a MIV melhoraria a sobrevivência embrionária pós-criopreservação. O delineamento experimental foi realizado com 3 grupos experimentais: complexos cúmulos-oócito (COCs) grau 1 foram maturados in vitro com suplementação de 10% de SFB como grupo controle, 10% da adição da fração apolar ou 10% da fração polar. Os COCs foram fertilizados in vitro e os possíveis zigotos foram cultivados em meio livre da adição de SFB por sete dias. As taxas de clivagem e blastocistos foram avaliadas após 96 e 168 horas de cultivo, respectivamente e observou-se que não houve diferença estatística entre os grupos com relação a ambas as taxas. Alguns blastocistos foram fixados para contagem de células da massa celular interna (MCI) e do trofectoderma (TE). Os demais blastocistos foram então submetidos ao congelamento lento com etilenoglicol. Após o descongelamento e cultivo por 24h, não houve diferença significativa entre os grupos na sobrevivência embrionária avaliada pela re-expansão da blastocele. Foi realizado o teste de integridade de membrana celular através da coloração diferencial, que apresentou uma diminuição no número total de células íntegras dos grupos apolar e polar em relação ao controle, entretanto não houve diferença quanto ao número de células lesadas nem quanto à proporção de células lesadas sobre as íntegras. O teste de integridade da membrana epitelial do trofectoderma indicou que não houve diferença entre os grupos. Ainda à incidência de apoptose foi avaliada por quantificação de caspase ativa, que apresentou-se reduzida no grupo Apolar em relação aos outros dois grupos. Embriões descongelados foram também fixados e tiveram as células da MCI e TE contadas por imunofluorescência e não houve diferenças estatísticas entre os grupos, antes ou depois do congelamento. Em conclusão, a adição das fases polar ou apolar na MIV não melhorou a sobrevivência embrionária pós- congelação, entretanto o teste da caspase ativa nos indica que são necessários mais estudos para investigar se a relação de menor apoptose tem influência significativa na sobrevida do embrião após sua transferência na receptora.The cryopreservation of embryos, combined with in vitro embryo production (IVP), is an important tool for disseminating high-value genetics in cattle farming. However, this tool could be even more widespread with improved pregnancy outcomes using the direct transfer (DT) method. The inconsistent results of DT may be influenced by the addition of fetal bovine serum (FBS) during oocyte maturation, as it affects the lipid composition of IVP blastocysts. Therefore, the objective of the present study was to evaluate and compare the benefits of replacing FBS with another lipid source during in vitro maturation (IVM). The study′s hypothesis was that the addition of the apolar or polar phase of FBS, replacing whole FBS during IVM, would improve post-cryopreservation embryonic survival. The experimental design consisted of 3 experimental groups: grade 1 cumulus-oocyte complexes (COCs) were matured in vitro with supplementation of 10% FBS (control group), 10% apolar fraction, or 10% polar fraction. COCs were fertilized in vitro, and the possible zygotes were cultured in FBS-free medium for seven days. Cleavage and blastocyst rates were evaluated after 96 and 168 hours of culture, respectively, and no statistical differences were observed among groups in both rates. Some blastocysts were fixed for cell counting of the inner cell mass (ICM) and trophectoderm (TE). The remaining blastocysts were then subjected to slow freezing with ethylene glycol. After thawing and 24-hour culture, no significant differences were found among groups considering embryonic survival, assessed by blastocoel re-expansion. A cell membrane integrity test was conducted using differential staining, which showed a decrease in the total number of intact cells in the apolar and polar groups compared to the control; however, no differences were found regarding the number of damaged cells or the proportion of damaged to intact cells. The trophectoderm epithelial membrane integrity test indicated no differences among groups. The incidence of apoptosis was also evaluated by quantifying active caspase, which was reduced in the Apolar group compared to the other two groups. Thawed embryos were also fixed, ICM and TE cells were counted using immunofluorescence, and no statistical differences between groups were observed either before or after freezing. In conclusion, the addition of polar or apolar phases did not improve post-cryopreservation embryonic survival; however, the active caspase test suggests that further studies are needed to investigate whether the lower apoptosis rate significantly affects embryo survival after transfer to the recipient

    Influence of HIPPO signaling pathway on segregation between internal cell mass and trophectoderm in bovine embryos

    O primeiro evento de diferenciação celular em mamíferos consiste na separação entre a massa celular interna (MCI) e o trofectoderma (TE). Em camundongos, a via de sinalização HIPPO atua no controle da expressão de genes que definem essa diferenciação. Em bovinos, há indícios que os mesmos genes não participam da mesma maneira, porém a via HIPPO pode estar implicada nesse evento biológico. No presente estudo buscou-se testar a hipótese de que a atividade da proteína quinase LATS2 é necessária para a diferenciação da MCI em embriões bovinos. Para isso, realizou-se edição gênica via sistema CRISPR/Cas9 com intuito de inibir a função quinase de LATS2. Zigotos bovinos produzidos in vitro foram microinjetados com RNAs guia para o gene LATS2 acrescidos da enzima de restrição Cas9. Os zigotos foram separados em 3 grupos experimentais, sendo: grupo controle (não microinjetado), grupo Cas9 (microinjetado apenas com a proteína Cas9) e grupo gRNA (microinjetado com dois RNAs guias para o gene LATS2 e proteína Cas9). Os embriões foram cultivados para avaliação da taxa de formação de blastocistos, distribuição celular e fixados em paraformol 3,8% para posterior realização da imunofluorescência para YAP, seguido de avaliação de genótipo. Os resultados obtidos sugeriram que a clivagem não foi afetada no grupo editado, sendo as taxas 58%, 36% e 50% para grupo controle, Cas9 e gRNA respectivamente; porém, as taxas de formação de blastocisto sugeriram uma interferência direta no grupo editado, sendo 30%, 10% e 6% respectivamente, para os grupos controle, Cas9 e gRNA. Observou-se no âmbito morfológico que os embriões do grupo gRNA aparentavam estruturas assemelhadas a mórulas, sugestivo de morte embrionária ou bloqueio da diferenciação nessa fase do desenvolvimento. Houve redução numérica no número de células nas estruturas do grupo gRNA. As imagens obtidas via microscopia confocal, após a realização de imunofluorescência para YAP, demonstram a marcação de células do TE nos embriões microinjetados apenas com Cas9 e controle, porém, essa marcação não ocorreu nas no grupo gRNA em embriões que não formaram blastocele. Após a extração individual de DNA dos embriões testados na imunofluorescência, a PCR da região alvo de LATS2 resultou em uma banda de 478pb, além da banda original de 650pb, em uma amostra do grupo gRNA. Após sequenciamento dos produtos de PCR pelo método Sanger, constatou-se a alteração genética oriunda da deleção de trecho do gene LATS2 em 2 embriões, demonstrando o sucesso da edição gênica e corroborando com os demais achados. Isso sugere que a via HIPPO possui papel significativo na diferenciação de MCI e TE em embriões bovinos e de que o gene LATS2 está ligado à formação de blastocistos na espécie bovina.The first cellular differentiation event in mammals consists of the separation between the internal cellular mass (ICM) and the trophectoderm (TE). In mice, the HIPPO signaling pathway acts to control the expression of genes that define this differentiation. In cattle, there are indications that the same genes do not participate in the same way, but the HIPPO pathway may be involved in this biological event. In the present study we aimed to test the hypothesis that the activity of LATS2 kinase is necessary for the differentiation of ICM in bovine embryos. In order to achieve this, we employed CRISPR/Cas9 gene editing system to inhibit the kinase function of LATS2. In vitro produced bovine zygotes were microinjected with RNAs guide aiming the LATS2 gene plus Cas9 restriction enzyme. The zygotes were sorted in 3 experimental groups: control group (not microinjected), Cas9 group (microinjected only with Cas9 protein) and gRNA group (microinjected with guide RNAs aiming the LATS2 gene and Cas9 protein). The embryos were cultured to evaluate the rate of blastocysts formation, cellular distribution, then fixed in 3.8% paraformol for subsequent immunofluorescence for YAP, followed by genotyping. The results suggested that the cleavage was not affected in the edited group, as rates were 58%, 36% and 50% for control , Cas9 and gRNA groups respectively; however, the blastocyst formation rates suggested a direct interference in the edited group, being 30%, 10% and 6% respectively, for the control groups, Cas9 and gRNA. Morphologically, it was observed that the embryos of the gRNA group appeared as morulas, suggestive of embryonic death or inhibition of differentation in this phase of development. There was a numeric reduction in total cell count in the structures of the gRNA group. The images obtained via confocal microscopy, after performing immunofluorescence for YAP demonstrate the marking of cells of the TE and MCI in the microinjected embryos only with Cas9 and control, however, this marking did not occur in the microinjected structures that did not form a blastocoel. After individual DNA extraction from the embryos tested in immunofluorescence, PCR of the LATS2 target region resulted in a band of 478pb, in addition to the original 650pb band, in one sample of the gRNA group. After Sanger sequencing of PCR products, genetic alteration derived from deletion of part of the LATS2 gene was observed in 2 embryos, demonstrating gene editing success and corroborating the findings. This suggests that HIPPO pathway has a significant role in the differentiation of MCI and TE in bovine embryos and that the LATS2 gene is linked to the formation of blastocysts in bovine species

    Insertion of fluorescent reporter gene into SOX2 gene region to study cell differentiation in early bovine embryo development

    A separação entre massa interna celular (MCI) e trofectoderma (TE) é o primeiro evento de diferenciação celular que ocorre no embrião mamífero, seguida pela diferenciação na MCI do epiblasto (EPI) e endoderma primitivo (PE). Falhas nestes processos de desenvolvimento inicial podem ocasionar perdas embrionárias e consequentemente econômicas. A influência do sistema de produção embrionária nos primeiros eventos de diferenciação celular precisa ser mais elucidada, refletindo em índices que ainda podem ser melhorados na produção de embriões bovinos in vitro. O soro fetal bovino (SFB), apesar de seus efeitos deletérios sobre fatores epigenéticos, ainda é usado em sistemas de produção in vitro e pode influenciar esta diferenciação. Ainda, as técnicas para estudo dos embriões normalmente envolvem a destruição dos mesmos e não permitem acompanhar em tempo real a dinâmica de expressão dos genes relacionados à diferenciação em MCI e TE. Novas técnicas como CRISPR/Cas9 permitem alteração direcionada da sequência de DNA genômico, assim, a introdução direcionada de proteínas fluorescentes repórteres permitiria a visualização em tempo real da expressão de genes de interesse nos embriões. Diante disso, nossos objetivos foram: testar a influência do soro fetal bovino nas primeiras diferenciações e, realizar a inserção dirigida de uma proteína repórter fluorescente na região do gene SOX2 por recombinação homóloga. Ainda, comparamos a eficiência da inserção gênica com uso do sistema CRISPR em diferentes momentos do desenvolvimento embrionário, com diferentes horários pós inseminação e com diferentes concentrações de material injetado. No primeiro experimento observamos que a retirada do SFB do cultivo embrionário não causa alterações na alocação de células na MCI ou TE, quando se alia esta remoção à renovação de uma parte do meio de cultivo às 90 horas pós inseminação (hpi). A ausência de suplementação de SFB ou de meio de cultivo levou à diminuição das células do TE, entretanto não alterou o número de células do PE. No segundo experimento, nenhum embrião produzido apresentou fluorescência, portanto todos os blastocistos tiveram seu DNA extraído para genotipagem. Foi realizada uma amostragem, e de 34 embriões verificados, um apresentou banda correspondente ao local do inserto e também banda correspondente ao embrião controle (wild type), sugerindo mosaicismo. Ao final, observamos que as melhores probabilidades de inserção gênica nos embriões bovinos aconteceram quando a injeção dos componentes do sistema CRISPR foi realizada após 8 horas de FIV e com maiores concentrações. Em conclusão, pudemos remover o SFB do nosso sistema de produção de embriões e surpreendentemente o SFB não influenciou a diferenciação em PE. Ainda, são necessários maiores estudos a fim de se otimizar a ferramenta para a obtenção de knock-in em embriões bovinos, principalmente para que se obtenha taxas de maior sucesso e redução da probabilidade de ocorrência de mosaicismo nestes embriões.The separation between inner cell mass (ICM) and trophectoderm (TE) is the first cell differentiation event that occurs in the mammalian embryo, followed by differentiation into the epiblast MCI (EPI) and primitive endoderm (PE). Failures in these early development processes can lead to embryonic and subsequent economic losses. The influence of the embryo production system on the first cell differentiation events needs to be further elucidated, reflecting in rates that can still be improved in the production of in vitro bovine embryos. Fetal bovine serum (FBS), despite its deleterious effects on epigenetic factors, is still used in in vitro production systems and may influence this differentiation. Furthermore, the techniques for studying embryos usually involve their destruction and do not allow real-time monitoring of the expression dynamics of genes related to differentiation in ICM and TE. New techniques such as CRISPR/Cas9 allow targeted alteration of genomic DNA sequence, thus targeted introduction of fluorescent reporter proteins would allow real-time visualization of the expression of genes of interest in embryos. Therefore, our objectives were: to test the influence of fetal bovine serum in the first differentiations and to carry out the targeted insertion of a fluorescent reporter protein in the region of the SOX2 gene by homologous recombination. Furthermore, we compared the efficiency of gene insertion using the CRISPR system at different moments of embryonic development, with different times after insemination and with different concentrations of injected material. In the first experiment, we observed that FBS removal from the embryonic culture does not cause changes in cell allocation in the ICM or TE, when this removal is combined with the renewal of a part of the culture medium at 90 hours after insemination (hpi). The absence of FBS or culture medium supplementation led to a decrease in TE cells, however it did not change the number of PE cells. In the second experiment, no embryo showed a fluorescent signal, so all blastocysts had their DNA extracted for genotyping. We sampled 34 embryos and one presented a band corresponding to the insertion site and also a band corresponding to the control embryo (wild type), suggesting mosaicism. We observed that best chances of gene insertion occurred when the injection of components of the CRISPR system was performed after 8 hours of IVF and with higher concentrations. In conclusion, we were able to remove SFB from our embryo production system and surprisingly SFB did not influence differentiation into PE. Still, further studies are needed in order to optimize the tool for obtaining knock-in in bovine embryos, mainly to obtain higher success rates and reduce the probability of occurrence of mosaicism in these embryos

    Functional evaluation of NANOG gene in segregation of cell lineages in bovine embryos

    O desenvolvimento embrionário pré-implantacional é marcado pelos primeiros eventos de diferenciação celular em mamíferos. O primeiro evento de segregação celular consiste na separação entre a massa celular interna (MCI) e o trofectoderma (TE), enquanto a segunda diferenciação é o momento da separação entre epiblasto (EPI) e endoderma primitivo (PrE) dentro da MCI. Os mecanismos moleculares que controlam essa diferenciação envolvem a expressão temporal de fatores de transcrição específicos. Dados recentes indicam que o estudo do desenvolvimento embrionário inicial bovino seria um bom modelo para humanos. Sabe-se que, em camundongos, o fator de transcrição NANOG atua opostamente a outro fator de transcrição, GATA6 e as análises de expressão gênica agrupam populações celulares de forma que NANOG e GATA6 tornam-se mutuamente excludentes em EPI e PrE. O presente projeto procurou testar a hipótese de que NANOG é necessário para a especificação de EPI e repressão de PrE em embriões bovinos. Para tal, almejou-se realizar a deleção gênica de NANOG mediada por CRISPR/Cas9 em zigotos produzidos in vitro. Os grupos experimentais consistiram em zigotos microinjetados 8 ou 16 horas após a FIV com os RNA guias visando a deleção do gene NANOG em duas concentrações, 12,5 ou 80 ng/µl, e Cas9, sendo denominados Cas9-12,5 e Cas9-80, respectivamente, enquanto o grupo controle não foi microinjetado. Posteriormente os embriões foram cultivados para avaliação das taxas de clivagem, formação de blastocistos e desenvolvimento, análise da expressão proteica por imunofluorescência, análise de expressão gênica e genotipagem. Foi avaliada a expressão proteica de SOX17, um marcador de PrE por imunofluorescência, entretanto, nenhum dos blastocistos do grupo Cas9-12,5 apresentou a deleção esperada no gene NANOG. Foi observada uma diminuição importante da expressão de NANOG no grupo experimental Cas9-80 quando comparado ao grupo controle (p=0.0360), indicando que houve uma perturbação da expressão gênica, porém sem indícios de deleção total de NANOG nestas condições. As condições de tratamento do grupo Cas9-80, entretanto, não afetaram a expressão gênica de GATA6. Portanto, foi possível reduzir a expressão de NANOG com uso do sistema CRISPR em bovinos, sem alteração na formação de blastocistos e na expressão de GATA6, o qual tem relação com a diferenciação em PrE.The early cell differentiation events mark the preimplantation embryonic development in mammals. The first cell differentiation event consists of the segregation between the trophectoderm (TE) and the inner cell mass (ICM), while the second event occurs within the ICM, with the segregation of epiblast (EPI) and primitive endoderm (PrE) lineages. The molecular mechanisms in the control of these events include time- related expression of specific transcription factors. Recent data indicate that the study of bovine embryos would be a good model for early human development. In mice, the transcription factor NANOG works opposite to another transcription factor, GATA6, and gene expression leads to cell population clusters in such a way that NANOG and GATA6 become mutually exclusive in EPI and PrE. This study sought to test the hypothesis that NANOG is necessary for EPI specification and PrE repression in bovine embryos. For this purpose, this study targeted CRISPR/Cas9-mediated NANOG deletion of in vitro produced zygotes. The experimental groups consisted of microinjected embryos with guide RNAs in two concentrations, 12.5 or 80 ng/µl, and Cas9 protein (Cas9-12.5 and Cas9-80) aiming to delete the NANOG gene, while the control group was not microinjected. Subsequently, embryos were cultured for evaluation of cleavage rates, blastocyst formation, and development rates, protein expression analysis by immunofluorescence, gene expression analysis, and genotyping. The protein expression of SOX17, a marker of PrE, was evaluated by immunofluorescence, however, none of the Cas9-12.5 embryos presented the expected gene deletion of NANOG. A significant decrease in NANOG expression (p=0.0360) was observed in the Cas9-80 experimental group when compared to the control group, suggesting a disturbance of gene expression, but with no evidence of NANOG full deletion under these conditions. The Cas9-80 treatment conditions, however, did not affect GATA6 gene expression. Therefore, it was possible to reduce the expression of NANOG using the CRISPR system in cattle embryos, without changing the blastocysts formation and the expression of PrE differentiation-related gene GATA6

    Assisted reproductive technologies (ARTs) in domestic animals are used to increase the efficiency of cattle production, especially by reducing the generation interval in breeding programs. Somatic cell nuclear transfer (SCNT) allows for the generation of identical individuals of high genetic merit and it is also used to generate transgenic animals by modification of the donor's genome. However, SCNT efficiency is still very low when compared to other ARTs such as in vitro fertilization. The use of embryonic stem cells (ESCs) as donor nucleus could enhance the outcome of SCNT as well as allowing targeted gene modification. Nevertheless, true ESCs were not derived so far in the bovine species. Thus, we focused on SCNT as an ART that can accomplish not only the horizontal propagation of a given genome but also introduce genome modifications. During SCNT, a somatic cell must be transformed into a pluripotent cell first - in a matter of hours - and then reinitiate embryonic differentiation. The first differentiation event the pre-implantation embryo is lineage specification, in which a fraction of the cells in the embryo will form the trophectoderm and the rest will form the fetus itself. This process is poorly understood in the bovine, not only in the context of SCNT but in fertilized embryos as well. We suggest that studying the mechanisms of lineage specification in bovine could help understand some of the inefficiencies observed in bovine SCNT and it could also help us develop novel strategies to obtain true bovine ESC. We focused in three genes, OCT4, CDX2 and SOX2 that are key regulatory factors in most pre-implantation mammalian embryos. First we tested the hypothesis that expression of OCT4 in donor cells would improve the efficiency of SCNT. Subsequently we tested the hypothesis that CDX2 is not required for trophectoderm establishment, but important in maintaining its integrity in bovine embryos. To further comprehend lineage specification in bovine, we tested the hypothesis that SOX2 is required for inner cell mass formation. We found that indeed preconditioning the somatic cells with OCT4 expression prior to SCNT has a distinct effect on the phenotype of the cloned blastocysts. We also found, contrary to our expectations, that CDX2 and SOX2 are not required for the first cell differentiation event in the bovine fertilized embryo. Overall, the data presented here has direct implications in the understanding of bovine embryology. It could help improve SCNT outcome and further understand lineage specification in the pre-implantation embryo as well as subsequent embryonic processes.Thesis (Ph.     Development and validation of a equine genotyping multiplex kit based on microsatellite marker analysis for individual identification and parentage testing

    O desenvolvimento da indústria equina tem aumentado a demanda por testes de verificação de parentesco para fins de registro na raça, os quais são realizados pela análise de regiões microssatélites de repetições curtas (STR). O Ministério da Agricultura, Pecuária e Abastecimento exige, atualmente, a análise de 12 marcadores para o painel principal (sendo 9 obrigatórios e 3 a escolher entre outros 8) e 12 marcadores para um painel adicional (a escolher entre possíveis 15). No Brasil há apenas um kit comercial disponível para um painel principal, o qual possui elevado custo, e nenhum kit comercial disponível para o painel adicional. Desta forma, muitos laboratórios são levados a desenvolver seus próprios testes, os quais são realizados utilizando-se várias reações de PCR. Este trabalho teve como objetivo o desenvolvimento de um sistema de reações múltiplas (multiplex) para cada um dos painéis com custo baixo e alta eficiência. Foi realizada análise in silico dos primers recomendados pela International Society for Animal Genetics (ISAG), organização responsável pela padronização desses testes, para 17 marcadores do painel principal (AHT4, AHT5, ASB2, ASB17, ASB23, CA425, HMS1, HMS2, HMS3, HMS6, HMS7, HTG4, HTG6, HTG7, HTG10, LEX3 e VHL20) e para 15 marcadores do painel adicional (TKY279, TKY287, TKY294, TKY297, TKY301, TKY312, TKY321, TKY325, TKY333, TKY337, TKY341, TKY343, TKY344, TKY374 e TKY394), os quais foram testados, quanto a sua eficiência, em formato monoplex e depois em formato multiplex. Foram redesenhados primers para 11 marcadores STR visando a melhor eficiência do sistema multiplex para cada painel. As concentrações de cada primer foram ajustadas para obtenção de uma amplificação equilibrada para todos os loci. As amostras foram sequenciadas no equipamento ABI 3130XL® (Applied Biosystems) e posteriormente analisadas no software Genemapper® v 5.0 (Applied Biosystems). Foi desenvolvido parcialmente um sistema multiplex com 14 marcadores para o painel principal de genotipagem de equinos (AHT4, AHT5, ASB2, ASB17, HMS1, HMS2, HMS3, HMS6, HMS7, HTG4, HTG6, HTG7, LEX3 e VHL20), uma vez que o marcador obrigatório HTG10 obteve resultado satisfatório apenas no formato monoplex, precisando ser testado em uma reação de PCR a parte. Os resultados das 50 amostras analisadas pelo sistema desenvolvido neste estudo, para o painel principal, foram consistentes quando comparados aos do kit comercial Equine Genotypes® (Thermo Fisher Scientific). Para o painel adicional (série TKY) foi possível o desenvolvimento composto de dois sistemas multiplex, um contemplando 8 marcadores (TKY 279, TKY297, TKY301, TKY312, TKY325, TKY337, TKY374 and TKY394) e outro contemplando 5 marcadores (TKY287, TKY321, TKY333, TKY343 and TKY344). Ambos os sistemas reproduziram os resultados das amostras referências provenientes da ISAG e testadas neste estudo. Pode ser concluído que ambos os painéis apresentaram bons resultados para análises, sendo possível desenvolver dois sistemas multiplex, atendendo o objetivo de desenvolver um método de baixo custo a alta eficiência, embora em duas etapas. Mais estudos deverão ser desenvolvidos com a finalidade de reunir em um único sistema multiplex todos os marcadores necessários.The development of the modern horse industry has increased the demand for equine parentage tests for pedigree verification, which are based in short tandem repeats (STR) analysis. These tests use 12 STR markers (of which 9 are compulsory and 3 chosen from another 8) for a main panel and other 12 STR (chosen from possible 15) for an additional panel and are regulated by the Ministry of Agriculture, Cattle and Supply (MAPA). There is only one imported commercial kit available in Brazil for the main panel, which it is costly and there are not any commercial kits available for the secondary panel. Thus, many laboratories must customize their own tests, which are performed by several PCR reactions. This work aimed to develop a novel multiplex STR typing system for each panel with low cost and high efficiency. Seventeen primers recommended by ISAG for the main panel (AHT4, AHT5, ASB2, ASB17, ASB23, CA425, HMS1, HMS2, HMS3, HMS6, HMS7, HTG4, HTG6, HTG7, HTG10, LEX3 and VHL20) and fifteen recommended for the additional panel (TKY279, TKY287, TKY294, TKY297, TKY301, TKY312, TKY321, TKY325, TKY333, TKY337, TKY341, TKY343, TKY344, TKY374 and TKY394) were submitted to in silico analysis. Then primers were validated individually and then jointly to test the efficiency of the multiplex system. Eleven primers were redesigned to establish a novel multiplex PCR system for each panel. Primer concentrations were adjusted in order to achieve a well-balanced set of amplicons for all markers. Samples were loaded into ABI 3130XL® Analyzer (Applied Biosystems) and then analyzed in Genemapper® v5.0 software (Applied Biosystems). A single multiplex STR genotyping system was partially developed for the main equine panel containing 14 loci (AHT4, AHT5, ASB2, ASB17, HMS1, HMS2, HMS3, HMS6, HMS7, HTG4, HTG6, HTG7, LEX3 and VHL20) because HTG10 which is compulsory - only amplified well in a separately PCR reaction. Results from 50 tested samples were concordant between Equine Genotypes® kit (Thermo Fisher Scientific) and the system developed in this study for the main panel. Two multiplex PCR systems were developed for the additional panel, one containing 8 markers (TKY279, TKY297, TKY301, TKY312, TKY325, TKY337, TKY374 and TKY394) and other containing 5 markers (TKY287, TKY321, TKY333, TKY343 and TKY344). The developed multiplex systems were able to detect all alleles from ISAG reference samples tested in this work. Results indicate that multiplex systems worked well and are cost- effective while performed in 2 PCR reactions for each panel. Further studies are needed to include all markers in a single multiplex PCR reaction

    O estabelecimento de cultivo de células-tronco embrionárias (ESC) ainda não foi realizado com sucesso. Verificação de marcadores de pluripotência e diferenciação nos três folhetos germinativos são necessárias para validação de uma linhagem celular pluripotente. O objetivo deste estudo foi estabelecer e caracterizar o cultivo de ESC suínas usando Matrigel e comparar a expressão dos marcadores de pluripotência Oct-4, CD9 e α6-integrina em embriões. Blastocistos in vitro ou in vivo foram submetidos à imunocirurgia para cultura da massa celular interna, fixados para imunocitoquímica ou extração de RNA total para RT-PCR. Nenhuma colônia de ESC foi obtida usando co-cultivo em fibroblastos embrionários murinos (MEF) ou em Matrigel. Expressão de Oct-4, CD9 e α6-integrina foi detectada por PCR. Os produtos de PCR de CD9 e α6-integrina tiveram suas sequências nucleotídicas determinadas e comparadas com bases de dados públicas. O produto de CD9 foi idêntico à seqüência do CD9 suíno e o produto de α66integrina foi similar à humana e eqüina. Reação de Imunocitoquímica revelou a presença de Oct-4 no citoplasma de células da massa celular interna e do trofoblasto. CD9 e α6-integrina foram observados preferencialmente em células do trofoblasto. Não foi possível comparar a expressão dos marcadores de pluripotência entre ESC e embriões em suínos. Porém, este estudo descreve pela primeira vez a expressão de CD9 e α6-integrina em blastocistos suínos, os quais podem não estar relacionados com células pluripotentes embrionárias suínas.Establishment of embryonic stem cell (ESC) culture in pigs has not been achieved. Verification of pluripotency markers and differentiation in the three embryonic layers are necessary for validation of a pluripotent cell line. The objective of this study was to establish and characterize porcine ESC culture using Matrigel and compare the expression of pluripotency markers Oct-4, CD9 and α6-integrin with embryos. In vitro or in vivo porcine blastocysts were submitted to immunosurgery for culture of inner cell mass, fixation for immunocytochemistry or total RNA extraction for RT-PCR. No ESC colonies were obtained using co-culture on mouse embryonic fibroblasts (MEF) or on Matrigel. Expression of Oct-4, CD9 and α6-integrin was detected by PCR. CD9 and α6-integrin PCR products had their nucleotide sequence assessed and compared with public nucleotide database. CD9 product was identical to CD9 porcine sequences and α6-integrin product was similar to human and equine α6-integrin. Immunocytochemistry revealed Oct-4 expression in cytoplasm of the inner cell mass and trophoblast cells. CD9 and α6-integrin were observed preferentially on trophoblast cells. It was not possible to compare expression of pluripotency markers between porcine ESC and embryos. However, this study describes for the first time expression of CD9 and α6-integrin in porcine blastocysts, which may not be related to pluripotent porcine embryonic cells

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.
