Implementation and verification of HPV PCR method on cobas® 6800 for cervical cancer screening at Bordeaux University Hospital
No full textDepuis 2019, pour les femmes de plus de 30 ans, le dépistage du cancer du col de l’utérus (CCU) en France repose en première intention sur le test HPV, qui correspond à la détection des acides nucléiques des HPV à haut-risque (HPV-HR) par biologie moléculaire sur frottis cervico-utérin (FCU). Ce travail décrit la mise en place et la vérification de méthode en portée A de la PCR multiplex en temps réel cobas® HPV, marquée CE-IVD (Conformité Européenne In Vitro Diagnostic), pour la détection des HPV-HR dans le cadre du dépistage du CCU sur l’automate cobas® 6800 system (Roche Diagnostics, Meylan, France) au laboratoire de Virologie du CHU de Bordeaux. Une évaluation des performances analytiques a été réalisée afin de répondre aux exigences de la norme ISO 15189, ainsi qu’une comparaison avec la méthode précédemment en place, Xpert® HPV sur l’automate Cepheid GeneXpert® (Cepheid Europe SAS, Maurens-Scopont, France). Pour les résultats discordants, une troisième technique, le génotypage Anyplex II HPV28 Detection (Seegene, Eschborn, Germany), a été utilisée. La répétabilité a été évaluée sur des milieux de transport vierges, sur des échantillons négatifs et positifs et a présenté des résultats satisfaisants. Les résultats de la fidélité intermédiaire, appréciés sur un pool d’échantillons positifs servant de contrôle de qualité interne, sont conformes. Le laboratoire a obtenu la note maximale sur le programme d’évaluation externe de la qualité et aucune contamination n’a été mise en évidence dans l’essai effectué. Les résultats des deux méthodes pour 45 FCU de patientes ont été confrontés : 38 prélèvements présentaient des résultats concordants, 4 HPV-HR n’ont pas été détectés par cobas® HPV alors qu’ils étaient détectés avec Xpert® HPV (dont 2 prélèvements positifs faibles), 3 échantillons ont été rendus positifs sur cobas® 6800 alors qu’interprétés non détectables par le Cepheid GenXpert® (cycle d’amplification trop tardif). Les aspects pratiques de la mise en place de la méthode, la réorganisation du laboratoire et du circuit des prélèvements ont également été discutés. La technique cobas® HPV est mise en production depuis janvier 2021 au CHU de Bordeaux.Since 2019, for women over 30 years old, cervical cancer screening in France has been primarily-based on HPV testing, using detection of nucleic acids of high-risk HPV (HR-HPV) by molecular biology on smear test. This work describes implementation and verification of the method (type A) of multiplex real-time qualitative PCR cobas® HPV on cobas® 6800 Roche system (Roche Diagnostics, Meylan, France) for detection of HR-HPV in Virology laboratory of Bordeaux University Hospital. Evaluation of analytical performances was carried out in order to meet the requirements of the International Standard Organization (ISO) 15189. Comparison with the previous routine method, Xpert® HPV on Cepheid GeneXpert® (Cepheid Europe SAS, Maurens-Scopont, France), was performed. Discrepant results were investigated by HPV genotyping with Anyplex II HPV28 Detection assay (Seegene, Eschborn, Germany). Repeatability, evaluated on non-used transports medias, negatives and positives samples was satisfying, as well as intermediate precision, assessed on a pool of positive samples also used as an internal quality control. No contamination was found in the specific carried out assay. Highest score was obtained for international external quality challenges. Both HPV tests were compared in 45 cervical samples: 38 samples presented concordant results, 4 samples were HR-HPV negative with cobas® HPV although they were positive with Xpert® HPV (including 2 weak positive results), and 3 samples were positive with cobas® 6800 while non-detectable with Cepheid GenXpert® (late amplification cycle). Practical aspects of cobas® HPV assay set up, laboratory and samples routing reorganization were also discussed. Cobas® HPV test has been deployed since January 2021 at Bordeaux University Hospital
Going Beyond Counting First Authors in Author Co-citation Analysis

counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Get PDF“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
Get PDFWe provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
