SAN FRANCESCO E LA DEVOZIONE PER L’ARCANGELO MICHELE NELLE TRADIZIONI AGIOGRAFICHE: DALLA VERNA AL GARGANO

    No full text

Rescue of a neutralizing anti-viral antibody fragment from an intracellular polyclonal repertoire expressed in mammalian cells

AbstractThe intracellular expression of recombinant antibodies in mammalian cells is an experimental strategy to inhibit in vivo the function of selected molecules. However, one limitation of this technology is represented by the unpredictable behaviour of antibodies, under conditions of intracellular expression. For this reason, it would be desirable to exploit intracellular expression of antibodies to select or rescue more efficient ones from a polyclonal mixture. In this work we have successfully explored this possibility by rescuing a neutralizing anti-viral antibody fragment from an intracellularly expressed anti-reverse transcriptase polyclonal repertoire

Inhibition of murine leukaemia virus retrotranscription by the intracellular expression of a phage-derived anti-reverse transcriptase antibody fragment

The intracellular targeting of recombinant antibodies is an experimental strategy to interfere with the function of selected molecules that is being utilized in a variety of different systems for research and medical applications. Since recombinant antibodies are increasingly being derived from phage display libraries, we have exploited phage technology to isolate, from a large combinatorial library, human antibody fragments directed against human immunodeficiency virus type 1 reverse transcriptase (HIV-1 RT). We describe in this paper the in vitro and in vivo properties of a neutralizing anti-RT antibody fragment. We demonstrate that the heavy chain domain (VH-CH1) of the phage-derived antibody is able to inhibit the retroviral enzyme, in that it neutralizes the RNA-dependent DNA polymerase activity of HIV-1 RT. The VH-CH1 antibody fragment also neutralizes the activity of RT of drug-resistant HIV-1 mutants as well as that of murine retrovirus RT. To confirm the broad reactivity of the synthetic antibody fragment, we have assessed the ability of the intracellularly expressed VH-CH1 protein to interfere with murine retroviral infection. To this end, we developed an in vivo selection procedure based on the antibody-mediated resistance to a cytotoxic retrovirus and used this selection procedure to rescue, from a heterogeneous population, cells expressing the VH-CH1 antibody fragment. We finally demonstrate that the intracellular expression of the recombinant heavy chain antibody fragment leads to an efficient inhibition of viral retrotranscription by murine-based retrovirus

Ahlswede R, Gargano L, Haroutunian HS, Khachatrian LH. Fault-tolerant minimum broadcast networks. NETWORKS. 1996;27(4):293-307.Broadcasting is the task of transmitting a message originated at one processor of a communication network to all other processors in the network. A minimal k-fault-tolerant broadcast network is a communication network on n vertices in which any processor can broadcast in spite of up to k line failures in optimal time T-n(k). In this paper, we study B-k(n), the minimum number of communication lines of any minimal k-fault-tolerant broadcast network on n processors. We give the value of B-k(n) for several values of n and k and, in case k < [log n], give almost-minimum k-fault-tolerant broadcast networks. (C) 1996 John Wiley & Sons, Inc

Technical note: Applicability of infrared thermography as a non invasive measurements of stress in rabbit

[EN] Among the main physiological stress indicators, the temperature evaluation is very important and innovative because it may be monitored without directly interacting with the animal. The use of a thermographic system, which is based on the detection of infrared radiation emitted by a subject, is a suitable method in order to measure temperature without any contact. In this research, a thermographic system was employed in order to single out the rabbit skin's zones most suitable for the temperature monitoring during stress challenges. Six hybrid rabbits were observed during induced stress; the areas selected as reference were: the ocular area (globe and periocular area), the internal auricle pavilion, and a shaved area of the head. The results of this pilot study show that the thermographic technique is a suitable method for the evaluation of temperature on rabbit. The best areas singled out were the eye bulb, the periocular area and the ear skin. The results concerning the effect of stress on cutaneous temperature showed that during stress condition a decrease in temperature occurs with respect to the basal condition (AT~1°C) and this trend is more evident for the auricle pavillion. In fact, this reaction is more evidenced in the ear skin, where a vasoconstriction process occurs. Moreover, corticosterone levels slightly increase (P=0.08) following the stressor's challenge due to tonic immobility test. In this research, both temperature and the change in corticosterone level show that the stress reaction induced by tonic immobility test is stronger than the one due to the other stressors applied to rabbits. The authors wish to thank for the kind collaboration: Dr. Elena Cavallone, Dr. Marino Campagnol, Dr. Eugenio Heinzl, Dr. Daniela Zucca and Dr. Alessandra Cocilovo (Veterinary Medicine Faculty – University of Milan). The structure of the N = 50 isotones Cd-98, Ag-97 and Pd-96 is studied in terms of a shell model employing a realistic effective interaction derived from the Bonn-A nucleon-nucleon potential. The single-hole energies are fixed by resorting to an analysis of the low-energy spectra of the isotones with A greater than or equal to 89; Comparison shows that our results are in very satisfactory agreement with the available experimental data. This supports confidence in the predictions of our calculations
