Hydroxychloroquine for treatment of rheumatoid arthritis: multifocal electroretinogram and laser flare-cell photometry study
Stefano Sebastiani, Michela Fresina, Mauro Cellini, Emilio C Campos Department of Experimental, Diagnostic, and Specialty Medicine, Ophthalmology Service, University of Bologna, Bologna, Italy Purpose: To evaluate early changes in multifocal electroretinogram (mfERG) and subclinical aqueous humor flare and cellularity in patients receiving hydroxychloroquine (HCQ) as treatment for rheumatoid arthritis. Methods: Ten patients receiving treatment with HCQ and no ophthalmic symptoms were enrolled. After complete ocular examination, mfERG and laser flare-cell photometry were performed. Patients were also divided into two subgroups with HCQ cumulative dose (CD) higher or lower than 500 g. Results obtained were compared with a control group of ten healthy subjects and statistical analysis was performed. Results: In patients receiving HCQ treatment, mfERG P1-wave in ring 2 showed a significant reduction in amplitude and a significant increase in latency compared to healthy control subjects, respectively resulting in 1.143 µV vs 1.316 µV (P=0.040) and 38.611 ms vs 36.334 ms (P=0.024). These changes are highly related to CD. Furthermore, when using the laser flare-cell photometry, a significant increase in aqueous humor flare and cellularity was shown in patients with CD higher than 500 g, resulting in a mean value of 14.4 ph/ms compared to 8.1 ph/ms in patients with CD lower than 500 g (P=0.0029). These reports appear highly related to CD (P=0.001). Receiver operating characteristic curve analysis showed mfERG P1-wave amplitude in ring 2 as the most sensitive value in detecting early HCQ-related retinopathy. Conclusion: MfERG was shown to be a very sensitive test in detecting early retinal toxicity and should be used for the screening of patients receiving HCQ treatment. Although less sensitive, laser flare-cell photometry can provide further information to evaluate early toxic retinal cell damage. Superconducting and microstructural studies on sputtered niobium thin films for accelerating cavity applications
The aim of the present research activity was to verify the influence of the applied bias voltage on the microstructural and functional properties of magnetron sputtering physical vapour deposition (MS-PVD) niobium thin films for use in superconducting resonant cavities for particle accelerators. Four different sets of samples were produced, by varying both the applied bias voltage and the nature of the substrate (copper or quartz). The morphological, microstructural, and mechanical properties of the coatings were experimentally determined by focused ion beam scanning electron microscopy (FIB-SEM), atomic force microscopy (AFM), transmission electron microscopy (TEM), and nanoindentation techniques, and then correlated to the applied bias and nature of the substrate. The superconducting properties (critical temperature T(c) and residual resistivity) were determined by a calibrated four-contact probe and a cryogenic apparatus. The microstructures and surface properties of biased films grown on copper and quartz were compared. The observed differences are likely connected to the low conductivity of quartz that induces a re-sputtering effect and a consequent modification of the superconducting performances

Near UV-Vis and NMR spectroscopic methods for rapid screening of antioxidant molecules in extra-virgin olive oil
Several spectroscopic techniques have been optimized to check extra-virgin olive oil quality and authenticity, as well as to detect eventual adulterations. These methods are usually complementary and can give information about different olive oil chemical components with bioactive and antioxidant properties. In the present work, a well-characterized set of extra-virgin olive oil (cultivar Frantoio) samples from a specific area of Tuscany (Italy) were investigated by combining near UV-Vis absorption spectroscopy, 1H and 13C nuclear magnetic resonance (NMR) to identify and quantify different chemical components, such as pigments, secoiridoids and squalene, related to the nutritional and quality properties of olive oils. Moreover, the pigmentation index of olives, organoleptic and sensory properties, total phenolic compound contents and the lipidic fractions of olive oils were investigated. The results obtained are, finally, compared and discussed in order to correlate several properties of both olives and olive oils with specific features of the cultivation area

Morphological and Functional changes in the retino-tectal system of the pigeon during the early posthatching period
Anterograde transport of either HRP or wheat germ agglutinin‐conjugated HRP was used to study the posthatching development of the retinotectal connection in the pigeon. The functional maturation of the retinotectal system was also investigated by recording electroretinographic (ERG) and tectal evoked (TEP) responses to either flash or pattern stimuli. Two main morphological changes occurred in the retinotectal system during the first 6 days after hatching: (1) an ipsilateral retinofugal component that was present at hatching disappeared and (2) the outer tectal layers were progressively invaded by the contralateral retinofugal axons, which at hatching were limited to the stratum griseum et fibrosum superficiale of the dorsolateral tectal quadrant. During the early posthatching period, at the same developmental stage at which an ERG to unpatterned or patterned stimulation could first be recorded, a visually evoked response could be elicited in the contralateral optic tectum. Therefore, the retina and optic tectum seem to start functioning simultaneously, the limiting factor being the late maturation of photosensitive lamellae in the outer segments of the developing photoreceptors. During the first 20 days posthatching, the retinotectal system undergoes extensive development as revealed by latency and amplitude changes of the visually evoked potentials. We suggest that the pigeon visual system serves as a useful model for studies concerning visual development and plasticity
