    Motion of James Neff to Quash Subpoenas or Protective Order

    This motion was filed by investigative journalist and author James Neff to quash a subpoena by the State and the Sheppard Estate. Neff began to investigate the murder of Marilyn Sheppard in 1989 with plans to publish his findings in a book titled, “Chasing the Fugitive”, with Random House Publishing. He argued that compelling him to testify, and make public the results of his investigation and research, prior to publication, would interfere, and likely injure, his livelihood as a self-employed investigative journalist. Neff asks the court not to compel him to testify, or in the alternative to compensate him for his testimony

    Pheochromocytomas (PCCs) are tumors arising from neural crest-derived chromaffin cells. There are currently few animal models of PCC that recapitulate the key features of human tumors. Since such models may be useful for investigations of molecular pathomechanisms and development of novel therapeutic interventions, we characterized a spontaneous animal model (MENX rats) that develops endogenous PCCs with complete penetrance. Urine was longitudinally collected from wild-type (wt) and MENX-affected (mutant) rats and outputs of catecholamines and their O-methylated metabolites determined by mass spectrometry. Adrenal catecholamine contents, cellular ultrastructure and expression of phenylethanolamine N-methyltransferase (PNMT), which converts norepinephrine to epinephrine, were also determined in wt and mutant rats. Blood pressure was longitudinally measured and end-organ pathology assessed. Compared to wt rats, mutant animals showed age-dependent increases in urinary outputs of norepinephrine (P=0.0079) and normetanephrine (P=0.0014) that correlated in time with development of tumor nodules, increases in blood pressure and development of hypertension-related end-organ pathology. Development of tumor nodules, which lacked expression of PNMT, occurred on a background of adrenal medullary morphological and biochemical changes occurring as early as 1 month of age and involving increased adrenal medullary concentrations of dense cored vesicles, tissue contents of both norepinephrine and epinephrine and urinary outputs of metanephrine, the metabolite of epinephrine. Taken together, MENX-affected rats share several biochemical and pathophysiological features with PCC patients. This model thus provides a suitable platform to study the pathogenesis of PCC for preclinical translational studies aimed at development of novel therapies for aggressive forms of human tumors

    Das Themengebiet der vorliegenden kumulativen Dissertation sind Erkrankungen und Funktionsstörungen des Kiefergelenkes mit Analysen zum evidenzbasierten Vorgehen bei der idiopathischen Kondylusresorption, der Kiefergelenkluxation und den inflammatorischen Erkrankungen des Kiefergelenkes im Sinne der juvenilen idiopathischen Arthritis und rheumatoiden Arthritis des Kiefergelenkes. Ziel der vorliegenden kumulativen Arbeit war es, offene Fragen bezüglich evidenz- und konsensusbasierter Diagnostik und Therapie zu identifizieren und die im Rahmen der Erstellung der jeweiligen S3-Leitlinien aufgetretenen Kontroversen und/oder Desiderate zu analysieren. Die Publikation der Ergebnisse dieser Analysen soll zur Ausrichtung künftiger Forschung beitragen und die Etablierung von Standards auf diesen Themengebieten ermöglichen. Die Grundlage der vorliegenden Originalarbeiten bildete jeweils die umfassende Aktualisierung bzw. Erarbeitung der entsprechenden Leitlinien „Idiopathische Kondylusresorption“, Registernummer 007/066; „Kiefergelenkluxation“, Registernummer 007/063 (jeweils als Mitglied der Lenkungsgruppe Neff & Riechmann) und „Inflammatorische Erkrankungen des Kiefergelenks: Juvenile Idiopathische Arthritis (JIA) und Rheumatoide Arthritis (RA) des Kiefergelenks“, Registernummer 007/061 (Letztere als Koautorin). In eigener Arbeit fanden hierbei die Literaturrecherche inklusive Handsuche und Aktualisierungen, die anschließende Evidenzbewertung und Bewertung der methodischen Qualität der eingeschlossenen Publikationen, das Verfassen der Langfassung inklusive aktualisierten und neuen Empfehlungen und Statements sowie bei den Leitlinien „Idiopathische Kondylusresorption“, Registernummer 007/066 und „Kiefergelenkluxation“, Registernummer 007/063 die Initialisierung und Auswertung der Abstimmungsergebnisse der multiplen Delphi-Runden durch die Verfasserin der vorliegenden Arbeit als unabhängigen Monitor (nicht stimmberechtigt) statt. Anschließend erfolgte in eigener Arbeit das Verfassen der Kurzfassung und des Leitlinienreports. Die gesamte Leitlinienarbeit fand unter Anleitung und Korrektur von Herrn Prof. Dr. Dr. Neff statt. Nach Abschluss der Arbeit ließ sich zusammenfassend feststellen, dass auf allen drei Themengebieten trotz zum Teil erstmaliger Festlegung neuer Standards und erheblichen Errungenschaften, z.B. in Form einer erstmals international konsentierten Nomenklatur der Kiefergelenkluxation, weiterhin großer Forschungsbedarf besteht. Aufgrund der geringen Patientenzahl bei epidemiologisch schlechter Datenlage bleiben evidenzbasierte Aussagen in vielen Bereichen weiterhin nur unter Vorbehalt möglich.The subject area of this cumulative dissertation is diseases and functional disorders of the temporo-mandibular joint with analyses of the evidence-based approach to idiopathic condylar resorption, temporomandibular joint dislocation and inflammatory diseases of the temporomandibular joint as in juvenile idiopathic arthritis and rheumatoid arthritis of the temporomandibular joint. The aim of this cumulative work was to identify open questions regarding evidence- and consensus-based diagnostics and therapy and to analyse the controversies that arose during the preparation of the respective S3 guidelines. The publication of the results of these analyses should contribute to the orientation of future research and enable the establishment of standards in these subject areas. The foundation of these original works was the comprehensive updating and development of the corresponding guidelines „Idiopathische Kondylusresorption“, registry number 007/066; „Kieferge-lenkluxation“, registry number 007/063 (in each case as a member of the Neff & Riechmann steering group) and „Inflammatorische Erkrankungen des Kiefergelenks: Juvenile Idiopathische Arthritis (JIA) und Rheumatoide Arthritis (RA) des Kiefergelenks“, registry number 007/061 (the latter as co-author). The literature search including hand search and updates, the subsequent evidence assess-ment and evaluation of the methodological quality of the included publications, the writing of the long version including updated and new recommendations and statements as well as the initialisati-on and evaluation of the voting results of the multiple Delphi rounds for the guidelines „Idiopathische Kondylusresorption“, registry number 007/066 and „Kiefergelenkluxation“, registry number 007/063 were carried out by the author of the present work as an independent monitor (not entitled to vote). Subsequently, the abridged version and the guideline report were written in own work. The entire guideline work took place under the guidance and correction of Prof. Dr. Dr. Neff. At the end of the work, it could be summarised that there is still a great need for research in all three subject areas, despite the establishment of new standards for the first time in some cases and considerable achievements, e.g. in the form of the first internationally agreed nomenclature for tem-poromandibular joint dislocation.     Crew Resource Management for Large Unmanned Aircraft Systems Operations

    Abstract The author explored the application of crew resource management (CRM) principles to commercial flight operations conducted by large unmanned aircraft systems (UAS). Currently, there is no regulatory structure governing the operation of large UAS in commercial flight operations. However, the National Aeronautics and Space Administration (NASA) forecasted the future by demonstrating the feasibility of large remotely piloted UAS flight in June 2018 by flying Ikhana, a modified MQ-9 Predator B aircraft, over a 415-mile route in Class A and in Class E airspace over southern California (Kooser, 2018). Ikhana is a 10,500-pound aircraft with a wingspan of 66 feet capable of climbing to 40,000 feet. In order to establish the viability of CRM as an element of a safety management system to mitigate human factors errors, the research used a cross-case analysis by Neff (2016) of four large UAS accidents to derive a ranked list of the eight most prevalent hard-skill and soft-skill human errors identified as causal to the accidents. An additional study of five Indian Air Force UAS accidents by Sharma and Chakravarti (2005) was referenced and listed eight primarily soft-skill human factors contributing factors. The author applied a CRM teamwork centered template using communications and workload management inputs and situational awareness and threat and error management outputs to analyze and propose mitigations to the contributing human factors errors. The author concluded that a safety management system incorporating CRM principles and supported by appropriate leadership philosophy, policies, procedures, and best practices is an essential element to combat human factors errors and facilitate safe integration of large commercial UAS flight operations into joint use national airspace

    Resumé v anglickém jazyce Summary This thesis is an attempt to show two authors and mainly two of their major works created in two different cultures, concluded by comparison of these two key works. Vladimír Neff, through his five-part cycle of novels Sňatky z rozumu (and following) dealing to the broad extent with the lives of members of Born and Nedobyl families, has proved his outstanding narrative skills which, together with a fair dose of irony and perfect knowledge of historical facts, make this pentalogy Neff's life work that in a remarkable manner made its way into the history of Czech literature. The author draws very accurate picture of Prague changing over a hundred of years and portrays the glamour of the age that is to be noted for the pursuit of economic and technological progress. Thanks to psychologically very thoroughly and accurately depicted key figures of the founders of abovementioned families, some of which were given the typically bourgeois features by Neff, the readers become close witnesses of their rise and fall. Jaroslaw Iwaszkiewicz in his relatively extensive work Čest a sláva presents the life of two generations on the historical and social background depicted in great detail. It captures the hasty and sometimes breakneck changes in the modern history and the fate of the Polish..

    Progesterone receptor targets play critical roles in maintaining pregnancy and disease prevention

    The ovarian hormone progesterone influences many facets of uterine physiology through the transcriptional activity of progesterone receptor (PR). This body of work investigates the role of progesterone receptor targets heart and neural crest derivatives expressed 2 (HAND2) and insulin receptor substrate 2 (IRS2) in regulating uterine functions. Our lab has previously shown that the PR target HAND2 is a negative regulator of uterine epithelial proliferation and is hypermethylated and silenced in uteri from women diagnosed with endometrial hyperplasia and cancer. The study presented in chapter 2 demonstrates that exposure to the environmental toxicant bisphenol-A (BPA) causes increased methylation at the Hand2 promoter and reduced HAND2 expression. BPA also stimulated expression of fibroblast growth factors (FGF), stromal derived factors that drive uterine epithelial proliferation. Increased expression of the pro-proliferative FGFs coupled with decreased expression of the anti-proliferative HAND2 led to aberrant epithelial proliferation. This is notable in the uterine glands, the site of origin for endometrial hyperplasia and cancer, suggesting that environmental BPA exposure poses as a risk factor for these uterine conditions. We recently identified IRS2 as a direct target of PR in human endometrial stromal cells. IRS2 serves as an adaptor molecule for insulin and IGF1 receptors (IR and IGF1R, respectively). The study presented in chapter 3 explores the function of IRS2 during stromal differentiation. We find that IRS2, acting downstream of IR, is critical for inducing the expression of decidual markers and the changes in cell morphology characteristic of stromal differentiation. We also show that IRS2 promotes expression and membrane localization of glucose transporters (GLUT), allowing for increased glucose movement into the cell to meet the metabolic demands of differentiation. This work provides insight into the metabolic shifts in differentiating stromal cells and the hormone regulated factors that mediate it.
