    RAGE and HMGB1 expressions in fetal membranes of premature rupture of membranes patients

    BACKGROUND Premature rupture of membranes (PROM) often occurs in pregnancy. The fetal membrane weakening is caused by inflammation involving receptor activation for advanced glycation end-products (RAGE) and high mobility group box protein 1 (HMGB1). The associations between RAGE and HMGB1 with PROM are rarely studied. Hence, this study aimed to determine those associations in fetal membranes with PROM occurrence. METHODS This case-control study was conducted at Dr. Ramelan Central Naval Hospital, Surabaya, Indonesia, from August to November 2019. The subjects, determined using a non-probability sampling method (a saturated sample), were divided into PROM and normal pregnancy with intact fetal membranes (control) groups. Fetal membrane specimens were collected during vaginal and cesarean section deliveries. The expressions of RAGE and HMGB1 were determined using the immunohistochemical method and further analyzed using the Mann–Whitney U test. RESULTS The expression of RAGE in fetal membranes with PROM was significantly higher than the control (52.74% versus 14.9% expression/mm2, p<0.001), as well as the expression of HMGB1 (45.9% versus 8.5% expression/mm2, p<0.001). CONCLUSIONS The higher expressions of RAGE and HMGB1 in fetal membranes were associated with PROM

    Alcohol is used as beverage with various purposes. Drinking alcohol is supposed to be able to distress central nerve system. Saliva production is controlled by both symphatic and parasymphatic nerves. This design of study was a cross sectional. This study aimed to evaluate the effect of alcoholic drink on salivary acidity (pH) and volume. The subjects were 30 males, aged 15-35 years old, with the approval of ethical clearance from Ethic Comittee Medical Faculty of Yogyakarta Muhammadiyah University, Yogyakarta. The subjects were divided into 2 groups (alcoholic and non-alcoholic drink as control) with 15 persons, respectively. Saliva sample was collected at night. In alcoholic group, saliva was collected for 60 minutes after drinking alcohol. The subjects were instructed to gargle mineral water for 1 minute, then they were asked to stand and let the saliva flow for 3 minutes into saliva-collecting pot. Data analysis of salivary volume and pH using unpaired t-test (p<0,05). The results showed that there were significant differences in salivary volume and acidity (pH) between alcoholic and non alcoholic drink.     Paracetamol has analgesic properties comparable to NSAIDs, but paracetamol have minimal side effects. Paracetamol is metabolized via sulfation and glucuronidation conjugation which is then excreted in the urine. A small part of the paracetamol has been changed to NAPQI. NAPQI will be detoxified by gluthathione. In high doses, there in an increase in NAPQI and a decrease in glutathione levels that results in oxidative stress and liver cell necrosis. Curcumin is often used as a traditional medicine to treat liver disease where it contains phenolic groups capable to scavenge free radicals. Curcumin extract can improve cellular responses to oxidative stress such as increasing the expression of Nrf2, SOD, and gluthathione. The purpose of this research was to know the effect of curcumin on the improvement of liver function in white rats (Rattus novergicus) induced by high dose  paracetamol. The design of this research was a descriptive research using literature studies from at least 15 international journals indexed by Scimago or national journals indexed by Sinta published in 2015-2020. Based on the journals used in this research, giving curcumin at a dose of 200 mg/kg BW/day for 2 weeks was effective in significantly increasing gluthathione levels in rats receiving high dose paracetamol. Giving curcumin at a dose of 100 mg/kg BW/day for 7 days can reduce AST and ALT activity in rats receiving high dose paracetamol, but the dose of curcumin that was more effective in reducing AST and ALT activity was 200 mg/kg BW/day for 2 weeks. This is because of curcumin which functions as a hepatoprotector that bind directly to the toxic metabolite of paracetamol, thereby reducing the use of glutathione and quench free radicals, so that oxidative stress in the liver decreased and gluthathione levels increased, AST and ALT activity decreased

    Peran Ekstrak Sargassum Duplicatum terhadap Penurunan Edema Sendi Pergelangan Kaki Tikus dengan Artritis Ajuvan yang Terpajan Stresor Dingin

    Stresor dingin meningkatkan proses peradangan karena peningkatan produksi reactive oxygen species (ROS). Sargassum duplicatum mengandung antioksidan seperti polifenol (flavonoid dan florotanin) dan fukosantin. Tujuan penelitian ini mengetahui peran ekstrak S.duplicatum (ESD) dalam mengurangi edema dan menghambat ekpresi MMP-3 di sendi pergelangan kaki tikus dengan artritis ajuvan (AA) yang terpajan stresor dingin. Penelitian dilakukan pada bulan Januari - Februari tahun 2017 di Laboratorium Biokimia unit hewan coba Fakultas Matematika dan Ilmu Pengetahuan Alam Universitas Brawijaya, Malang. Digunakan 30 tikus AA jantan usia 10-12 minggu yang dibagi menjadi tiga kelompok, yaitu kelompok yang tidak mendapat perlakuan, kelompok yang dipajankan stresor dingin, dan kelompok yang diberi ekstrak ESD 400 mg/kg BB/hari per oral selama 21 hari serta dipajankan stresor dingin. Tebal sendi pergelangan kaki tikus diukur dengan kaliper dan ekspresi MMP-3 dengan metode imunohistokimia. Hasil uji anova menunjukkan kelompok tikus yang mendapatkan stresor dingin mengalami ketebalan sendi yang berbeda bermakna dengan kelompok tikus tanpa perlakuan (p=0,042) dan pemberian S. duplicatum menurunkan edema kaki tikus (p=0,001) secara bermakna jika dibandingkan kelompok yang hanya mendapat stresor dingin. S. duplicatum menurunkan secara bermakna jumlah sel yang mengekspresi MMP-3 di sendi pergelangan kaki tikus yang mendapatkan stresor dingin (p=0,001). Stresor dingin dapat menimbulkan edema di pergelangan kaki tikus model AA dan pemberian ekstrak S. duplicatum menurunkan edema secara bermakna serta menghambat ekspresi MMP-3 pada tikus model A

    Indomethacin is a nonsteroidal anti-inflammatory drugs (NSAIDs) that act to inhibit COX-1. The inhibition of COX-1 leads to inhibition of prostaglandin production. Prostaglandin is a regulator of gastric acid buffer secretion. Inhibition of prostaglandin decrease gastric and duodenum pH and damage the gastric and duodenum. Sea grapes (Caulerpa racemose var. cylindracea) are marcoalgae that contain flavonoids as antioxidants and anti-inflammatory. The aim of this study was to determine the effect of sea grapes on gastric and duodenum pH in rattus norvegicus induced by indomethacin. 32 male Rattus norvegicus were divided into 4 groups. Group K (-) without treatment. Group K (+) was induced by 30 mg/Kg BW indomethacin for 7 days. Group P1 was induced by 30mg/Kg BW indomethacin for 7 days followed by administration of 1g/100g BW sea grape extract for 14 days. Group P2 was induced by 30mg/Kg BW indomethacin for 7 days followed by administration of 2g/100g BW sea grape extract for 14 days. On the 29th day, rats were terminated, gastric and duodenal were isolated then the fluid pH was measured. One-way Anova test obtained p = 0,023. Post hoc test pH Gaster significantly different between group K (-) and K (+) (p= 0,005) and between group K (-) with P2 (p= 0,020). While in group K (+) with P1 and P2 there was no significant difference in pH.  Post hoc test of pH duodenum showed no differences in all group. The administration of sea grape extract did not show a differences of the gastric and duodenal pH significantly between rats induced by indomethacin

    Hiperkolesterolemia merupakan faktor resiko terjadinya gangguan kardiovaskuler pada diabetes mellitus. Pada diabetes mellitus kekurangan insulin meningkatkan lipolisis sehingga asam lemak bebas meningkat sehingga meningkatan kadar kolesterol total dan kolesterol LDL. Tomat mengandung likopen yang dapat menurunkan kadar kolesterol LDL karena mampu menghambat HMG-KoA reduktase. Penelitian ini untuk mengetahui pengaruh pemberian jus tomat (Solanum lycopersicum L.) terhadap kadar kolesterol total dan LDL tikus putih (Rattus norvegicus) jantan galur wistar yang diinduksi aloksan.  24 ekor yang dibagi menjadi 3 kelompok: 1). Kelompok kontrol negatif tanpa perlakuan 2). Kelompok kontrol positif hewan coba yang diinduksi aloksan 3). Kelompok perlakuan yang diinduksi aloksan dan diberi jus tomat dengan dosis 11 gram/KgBB selama 14 hari. Uji One-Way Annova menunjukkan peningkatan kadar kolesterol LDL yang signifikan antara kelompok kontrol positif dan kelompok hewan kontrol negatifnamun tidak pada kadar total kolesterol. Rerata kadar total kolesterol dan LDL pada kelompok perlakuan menunjukkan penurunan yang tidak signifikan di bandingkan kelompok hewan coba yang diinduksi aloksan. Pemberian aloksan dengan dosis 150 mg/Kg BB dapat meningkatkan kadar kolesterol LDL secarabermakna namun tidak pada kadar total kolesterol. Pemberian jus tomat dengan dosis 11 gram/KgBB cenderung menurunkan rerata kadar total kolesterol dan LDL

    A high-fat diet can increase lipoprotein levels, total cholesterol, (Low Density Lipoprotein) LDL, and triglycerides. Starfruit has saponin and flavonoid compounds which are expected to reducing LDL cholesterol levels. The aim of the study was to determine the effect of starfruit juice on lowering the blood cholesterol LDL of Wistar rats fed with high-fat diet. Experimental study with post-test only control group design. As many as 24 male white rats from the Wistar strain were divided into 3 groups: negative control groups (K-) that were given standard feed for 28 days; positive control group (K+) who were given a high-fat diet for 28 days; the treatment group (KP) was given a high-fat diet for 28 days and on the 15th day 28th they were given a starfruit juice with a dose of 4ml / 200grBB / day. Day 29 measured LDL cholesterol. The statistic test showed a significant increase in LDL cholesterol levels (p=0.001) in the positive control group (x=12.125±2.642 mg/dL) compared to the negative control group (x=7.625±1.506 mg/dL). There was no significant different the mean cholesterol level of the treatment group (x=11±1.927) compared to the mean LDL cholesterol level of the positive control group (x=12.125±2.642). A high-fat diet significantly increases the mean LDL cholesterol level. Starfruit juice did not significantly reduce LDL cholesterol level

    IMPLEMENTASI MODEL PEMBELAJARAN KOOPERATIF PADA MATA PELAJARAN KONTINENTAL

    Pembelajaran kooperatif ialah sistem pembelajaran yang memberikan peluang pada siswa agar saling bekerja sama dalam beberapa tugas terstruktur serta pendidik hanya bertugas sebagai fasilitator sehingga dapat diterapkan pada mata pelajaran kontinental. Tujuan artikel ini membahas tentang 1) konsep pembelajaran kooperatif, 2) mata pelajaran kontinental, dan 3) implementasi pembelajaran kooperatif pada hasil belajar pada mata pelajaran kontinental. Penulisan ini sebagai jenis penulisan empiris dikarenakan terdapat masalah yang mempunyai sifat sementara sehingga akan terdapat perkembangan sesudah berada di lapangan. Dalam metode penulisan artikel ini, dilakukan dengan menentukan data dan informasi pendukung penulisan. Kemudian, diolah pada tahap pengumpulan data dan yang terakhir dilakukan proses analisis yang merupakan evaluasi dari data yang telah diolah. Konsep pembelajaran kooperatif yaitu sebagai bentuk model pembelajaran melalui cara yaitu siswa bekerja dan belajar di berbagai kelompok kecil dengan berkolaboratif yang mana anggotanya meliputi 4-6 orang. Dengan struktur kelompok yang sifatnya sama. Mata pelajaran kontinental yaitu mata pelajaran yang menerapkan pengetahuan dan keterampilan siswa dalam mengolah beberapa hidangan gaya kontinental. Implementasi pembelajaran kooperatif pada mata pelajaran kontinental hanya dibatasi pada tipe STAD, Co-op Co-op, Make A Match, Snowball Throwing, dan TAI. Pengaplikasian model pembelajaran kooperatif terbukti bisa memaksimalkan hasil pembelajaran serta prestasi yang diraih siswa dalam mata pelajaran kontinental yang berkisar antara 77,78% sampai 100% sehingga model pembelajaran kooperatif efektif untuk dipergunakan pada mata pelajaran kontinental
