Ektrak Daging Buah Mahkota Dewa Meregenerasi Sel Pulau Langerhans Pada Tikus Putih Diabetes
In Indonesia, the increasing prevalence of diabetes mellitus is accompanied by the increasing financial burden to the patients. It is necessary to find alternative herbal treatment for diabetes, which is effective, cheap, and easy to find: the mahkota dewa. This research aims to find out the histological regeneration of Langerhans islet cells after given by flesh extract of mahkota dewa.This experimental research used complete randomized design (CRD). There were 5 treatments given to the twenty five mice; the negative control group (the normal one), the positive control (male white mice (Rattus norvegicus which were made diabetic by administering injection of 70 mg/kg of body mass of diabetogenic alloxan), the group of diabetic male white mice given 3, 6 and 9 grams/day/200 bodyweights), then were measured glucose in blood at 0, 1st and 4th week and counted Langerhans islet cells every 5 overviews.Statistically results showed that flesh extracts of mahkota dewa can decreased glucose in diabetic mice (r=0,75) with contribute 56,89% in decreasing, regenerated Langerhans islet cells of pankreas {Fcount(17,558)>Ftab(2,870)} with the highest increasing is 3,84 amount and started with 3grams/day/200 bodyweights doses, and the best effectiveness in regenerating was 11,81% consentration or 0,1181grams/day (6.56 grams/day in human)
Kafein Meningkatkan Ketebalan Tunika Muskularis Eksterna Rectum Embrio Gallus gallus Strain ISA Brown
Kafein dipergunakan secara luas dalam konsumsi sehari-hari dan di bidang pengobatan. Kafein meningkatkan kadar katekolamin, merangsang pertumbuhan dan proliferasi otot polos dan dapat merangsang sistem saraf simpatis. Tujuan penelitian adalah mengetahui pengaruh pemberian kafein terhadap ketebalan tunika muskulartis eksterna rectum embrio Gallus-gallus strain ISA brown. Metode penelitian adlah eksperimental dengan menggunakan tigapuluh butir telur Gallus-gallus strain ISA brown fertil dengan berat 60-70 gram dikelompokkan dalam 2 kelompok, 15 butir dalam perlakuan “caff” yaitu diberi kafein 0,1 mg/ml setiap hari sejak hari ke-4 masa inkubasi sampai hari ke-8 inkubasi dan kelompok kontrol mendapat aquabidestilasi dalamn waktu yang sama. Hari ke-19 inkubasi, hewan coba dibedah dan diambil rectum, diukur ketebalan tunika musculartis eksterna (stratum sirkuler dan stratum longitudinal) dibawah mikroskop. Hasil penelitian menunjukkan rerata ketebalan tunika muskularis rektum adalah 54,5313 µm (stratum longitudinal,“caff”) dibanding 42,0833 µm (stratum longitudinal, “kontrol”) dan 74,2969 µm (stratum sirkular “caff”) dibanding 72,7083 µm (stratum sirkular, “kontrol”) tetapi tidak ada perbedaan yang signifikan antara kelompok perlakuan dengan ketebalan tunika muskularis rektum stratum longitudinal (t=0,442) dan stratum sirkular (t=0,135) secara statistik. Kesimpulan terdapat peningkatan ketebalan tunika muskularis eksterna rektum embrio Gallus-gallus strain ISA brown setelah pemberian kafein walaupun secara statistik tidak signifikan
The time administration and cost burden of intravenous immunoglobulin (IVIG) therapy in a 4-year-old girl with Kawasaki disease: A case report
Kawasaki disease (KD) is an acute systemic vasculitis frequently affecting children under five years old. KD can cause severe complications. It can lead to coronary aneurysms in 15-25% of untreated cases. Intravenous immunoglobulin (IVIG) treatment within ten days of initial onset can reduce the rate of coronary artery aneurysms. However, IVIG administration in Indonesia is currently limited due to its high cost. The case reported a 4-year-old girl with a chief complaint of fever. She complained about a high fever seven days before hospitalization. She also complained about vomiting, cough, joint pain, diarrhea, skin peeling, and rash on her body, palms, and soles. On examination, she looked irritable and sluggish. The temperature was 39.9o C. There were red eyes without discharge, strawberry tongue, oral thrush, and red, dry, cracked lips, swollen neck lymph nodes, and skin rash on her body, palms, and soles. Laboratory testing showed microcytic hypochromic anemia with Hb 7.9 g/dL and leukocytosis 24.230/mm3. Chest X-ray showed perihilar and paracardial infiltrates. Electrocardiogram revealed sinus tachycardia. An echocardiogram showed left ventricle dilatation with trivial mitral regurgitation and no coronary abnormalities were found. She was diagnosed with Kawasaki Disease. She was treated by IVIG 30 gr single dose during 12 hours on day 7 of initial onset, methylprednisolone injection 10 mg/8 hours, paracetamol 150 mg/8 hours, and aspirin 400 mg/8 hours orally and discharged from the hospital with improvement. There weren’t any coronary artery abnormalities found. IVIG administration within ten days of initial onset in KD patients can reduce the risk of coronary artery complications. IVIG administration after day 10 of initial onset can achieve resolution of inflammation but can be insufficient for preventing coronary artery lesions (CALs)
Purwoceng (Pimpinella pruatjan Molk.) nanosuspension repairs spatial white albino wistar strains’ spatial memory degeneration after sleep deprivation
Paradoxical Sleep Deprivation (PSD) induces oxidative stress and interferes permanent memory consolidation in hippocampus. Purwoceng (Pimpinella prutjan Molken) is herbal medicine with antioxidant effect. These effects will be more effective with nanosuspension technology. The research purposes is to find effect of PSD to rats’ spatial memory reduction and to find purwoceng nanosuspension extract effect to improve rats’ spatial memory after PSD stress induction. The research method is randomized controlled trial with posttest only with control group approach to 36 white male albino rats Wistar strain which is induced by 96 hours PSD stress model. Experiment groups is regular extract 25 mg/300g BW/day, purwoceng nanosuspension extract 25 mg/300 gBW/day, 50 mg/300 gBW/day, and 75 mg/300 gBW/day. Statistical analysis found that there is no significant effect of purwoceng extract to memory traveling time (p=0,414) and memory track length (p=0,316) between pre-PSD and post-PSD group. There is significant effect of PSD stress after purwoceng administration to memory track length and memory travelling time (p=0,005). The conclusion is that there is significant correlation between the difference of memory latency and memory traveling time of MWM test between the various purwoceng nanoemulsion dose. The 25-50 mg/300 gBW/day is effective dose of purwoceng nanoemulsion extract to repairs spatial memory degeneration after PSD stress.</jats:p
Differences in body weight post-induction sleep deprivation and sleep recovery in white male rats (Rattus norvegicus)
Sleep Deprivation (SD) reduced leptin levels, increased ghrelin levels, and it caused were multifactorial, so research was conducted on experimental animals to prove whether SD was the single factor causing changes in body weight (BW). The study's objectives were to know the difference in BW after induction of paradoxical and total SD and to observe the improvement in sleep recovery (SR). This study was true experimental with posttest only, and a control group design used 25 male albino rats randomly shared into five groups; control, PSD, TSD, PSD+SR, dan TSD+SR on August – September 2021. The weight is measured by OHAUS® balance. Statistical analysis used by One-way ANOVA and paired t-test denoted no significant difference after SD (p=0,277) and SR (p=0,297), a significant difference in the TSD+SR and TSD between before (p=0,014), after SD (p=0,008), and after SR (p=0,034). Sleep deprivation increases BW through raised ghrelin, and SR reverses the effects by increasing the antioxidant. Results must be confirmed by measuring ghrelin levels and leptin orexin type 1 and 2 receptors. In conclusion, that was a significant difference in the TSD+SR and TSD between pre and post-sleep deprivation and the TSD+SR between pre and post-SR.
Paradoxical sleep deprivation decreases serum testosterone and Leydig cells in male rats
BACKGROUND Chronic stress increases glucocorticoid levels and accelerates reduction in Leydig cells functions and numbers. Chronic stress models in the working place comprise sleep deprivation, sedentary stress, and physical stress. The aim of this study was to evaluate the effect of various work stress models, such as stress from paradoxical sleep deprivation (PSD), immobilization, and footshock, on serum testosterone levels and number of Leydig cells in male albino rats. METHODS This study was of experimental randomized post-test only with control group design using 24 male Wistar albino rats (Rattus norvegicus). The sample was divided into 4 groups: K1 (control), K2 (PSD), K3 (immobilization) and K4 (footshock), receiving treatment for 25 days. Measured parameters were serum testosterone level and Leydig cell number. Analysis of variance (ANOVA) was used for statistical analysis, followed by post hoc LSD. RESULTS Mean serum testosterone levels (0.07 ± 0.08 ng/ml) and Leydig cell numbers (4.22 ± l0.96) were lowest in the PSD stress model. Serum testosterone levels differed significantly between controls and PSD group (p=0.014), while there was a significant difference in numbers of Leydig cells between footshock stress and PSD (p=0.011) and between the three stress groups and controls (p=0.006). CONCLUSION This study demonstrated that PSD, immobilization and footshock stress significantly decreased serum testosterone levels and number of Leydig cells in male albino rats (Rattus norvegicus). The mechanism by which PSD affects serum testosterone is still unclear

VITAMIN D PADA PASIEN ACNE VULGARIS DAN KORELASI DENGAN LUARAN KLINIS: REVIEW SISTEMATIK
Background: Vitamin D is thought to have a role in the pathogenesis of acne vulgaris through anti-inflammatory effects. Research regarding the relationship between vitamin D and the severity of acne vulgaris still shows mixed results. Objective: To determine the correlation between vitamin D levels and clinical severity in acne vulgaris patients. Method: This systematic review uses Pubmed, Google Scholar, and Science Direct databases. Of the 362 articles, 13 articles that met the inclusion criteria were analyzed narratively. Results: Of 13 studies, 4 showed significant differences in serum vitamin D levels between degrees of acne severity. Meanwhile, the other 9 did not show any significant differences. One study showed significant differences in tissue vitamin D levels with severity. Conclusion: Overall, results are still inconsistent. There is evidence of a higher risk of vitamin D deficiency and decreased tissue vitamin D levels in people with acne vulgaris as severity increases. It is suspected that this is related to the role of vitamin D in suppressing inflammation. Further studies considering confounding factors are needed to clarify this association.Latar Belakang: Vitamin D diduga memiliki peran dalam patogenesis acne vulgaris melalui efek antiinflamasi. Penelitian terkait hubungan vitamin D dengan derajat keparahan acne vulgaris masih menunjukkan hasil yang beragam. Tujuan: Mengetahui korelasi antara kadar vitamin D dengan derajat keparahan klinis pada pasien acne vulgaris. Metode: Review sistematik ini menggunakan basis data Pubmed, Google Scholar, dan Science Direct. Dari 362 artikel, 13 artikel yang memenuhi kriteria inklusi dianalisis secara naratif. Hasil: Dari 13 studi, 4 menunjukkan perbedaan bermakna kadar vitamin D serum antar derajat keparahan akne. Sementara 9 lainnya tidak menunjukkan perbedaan bermakna. Satu studi menunjukkan perbedaan bermakna kadar vitamin D jaringan dengan derajat keparahan. Kesimpulan: Secara keseluruhan, hasil masih inkonsisten. Terdapat bukti risiko defisiensi vitamin D lebih tinggi dan penurunan kadar vitamin D jaringan pada penderita akne vulgaris seiring peningkatan derajat keparahan. Diduga hal ini berkaitan dengan peran vitamin D dalam menekan inflamasi. Diperlukan studi lebih lanjut dengan mempertimbangkan faktor perancu untuk memperjelas asosiasi ini
