Een verkenning ten aanzien van de waterkwaliteit van het toekomstige Markiezaatsmeer
In 1983 is het in het kader van het WABASIM-project ontwikkelde eutrofiëringsmodel BLOOM II/CHARON overgedragen aan DDMI. Om nu ervaring met de werking. van het model op te doen, is besloten enige berekeningen voor een zoetwaterbekken uit te voeren. Om de hierboven vermelde reden is het nog niet bestaande nieuw te vormen Markiezaatsmeer gekozen. In de jaren 1977 - 1979 zijn er t.a.v. het Markiezaatsmeer reeds een aantal waterkwaliteitsvoorspellingen gedaan, die mede bepalend zijn geweest voor de inrichting van het gebied. Door de overdracht van het model BLOOM II/CHARON is er een nieuwe methodiek beschikbaar die alsnog toegepast wordt om de kwaliteit van het Markiezaatsmeer te voorspellen. Bovendien zijn ook twee andere voorspellingsmethodieken (Vollenweider en CUWVO) uitgewerkt.Wabasi
Veerse Meer aan de Oosterschelde: Toestand ecosysteem Veerse Meer vóór ingebruikname doorlaatmiddel
Als het water vanaf mei 2004 wordt ververst, zal er een grote verandering optreden in met name het watersysteem. De waterkwaliteit zal naar verwachting merkbaar verbeteren en die verbetering zal effect hebben op het ecologisch functioneren van het hele gebied. Door de ontwikkelingen in het watersysteem goed in de gaten te houden (te monitoren) kan worden getoetst of het verversen met Oosterscheldewater voldoende is, of dat er nog aanvullende maatregelen nodig zijn om het gewenste streefbeeld te bereiken. De opgetreden verbetering is alleen te kwantificeren als de monitoringsresultaten afgezet kunnen worden tegen de uitgangssituatie (ook wel T0-situatie genoemd) van vóór het doorlaatmiddel. De uitgangssituatie wordt in dit rapport beschreven aan de hand van water- en bodemparameters en aspecten van planten en dieren die al geruime tijd gemeten (gemonitord) worden. Ook over aspecten die heel direct op de waterkwaliteit en het waterpeil reageren of daarvan afhangen, wordt de uitgangssituatie beschreven. Een en ander blijft beperkt tot het waterlichaam van het Veerse Meer, en de oeverdelen die onder directe invloed van het water staan. De verwachte effecten van het doorlaatmiddel op de visserij, recreatie, bewoning en beleving komen in dit rapport niet aan de orde. Aan de functie-eisen van het RBPN (Regionaal Beheerplan Nat) moet nu of op termijn worden voldaan (Anonymus 2002). Het beschrijven van de uitgangssituatie, op basis van onder andere de resultaten van het huidige monitoringsprogramma, was een goede gelegenheid om te toetsen of aan de functie-eisen wordt voldaan en een hypothese op te stellen voor de haalbaarheid in de toekomst. In het water van het Veerse Meer komen de voedingsstoffen stikstof (N) en fosfor (P) en ook enige verontreinigingen nu nog in concentraties voor die hoger zijn dan de MTR (Maximaal Toelaatbaar Risiconiveau) of de GW (Grenswaarde), gesteld in de Vierde Nota Waterhuishouding (NW4). Uit de toetsing volgde bovendien dat diverse funstie-eisen aangepast moeten worden aan de nieuwe situatie. Het verversen van water als nieuwe beheersmaatregel zal grote veranderingen van de waterkwaliteit, en daarmee het ecosysteem, tot gevolg hebben. Het monitoringsprogramma dient afgestemd te zijn op de toekomstige ontwikkelingen en veranderende functie-eisen. In dit rapport wordt de uitgangssituatie beschreven en een voorstel gedaan voor de monitoring van de effecten van het doorlaatmiddel op het waterlichaam. In de tabel op de volgende pagina is samengevat op grond van welke kenmerken de ontwikkelingen kunnen worden getoetst, aan welk streefbeeld dit is gerelateerd en in welk monitoringskader dit kan worden ondergebracht
Measurements in an incompressible three-dimensional turbulent boundary layer, under infinite swept-wing conditions, and comparison with theory
First a three-dimensional turbulent boundary-layer experiment is described. This has been carried out with the specific aim of providing a test-case for calculation methods. Much attention has been paid to the design of the test set-up. An infinite swept-wing flow has been simulated with good accuracy. The initially two-dimensional boundary layer on the test plate was subjected to an adverse pressure gradient, which led to three-dimensional separation near the trailing edge of the plate. Next, a calculation method for three-dimensional turbulent boundary layers is discussed. This solves the boundary-layer equations numerically by finite differences. The turbulent shear stress is obtained from a generalized version of Bradshaw's two-dimensional turbulent shear stress equation. The results of the calculations are compared with those of the experiment. Agreement is good over a considerable distance; but large discrepancies are apparent near the separation line
Epiretinal Membrane Formation Associated With Idiopathic Macular Telangiectasia: Case Report
A 46-year-old woman complained of blurred and distorted vision in both eyes. Ophthalmic examination showed that visual acuity was 20/200 for the right eye and counting fingers left eye. Fundoscopy revealed perimacular hemorrhages, aneurismal dilatation of the vessels in the posterior pole, and a white and elevated lesion adjacent to vascular changes. We report a case of idiopathic macular telangiectasia and epiretinal membrane that occurs concomitantly. Clinical Trials In Brazilian Journals Of Ophthalmology: Where We Are
Purpose: To compare clinical trials published in Brazilian journals of ophthalmology and in foreign journals of ophthalmology with respect to the number of citationsand the quality of reporting [by applying the Consolidated Standards for ReportingTrials (CONSORT) statement writing standards]. Methods: The sample of this systematic review comprised the two Brazilianjournals of ophthalmology indexed at Science Citation Index Expanded and sixof the foreign journals of ophthalmology with highest Impact Factor® accordingISI. All clinical trials (CTs) published from January 2009 to December 2010 at theBrazilians journals and a 1:1 randomized sample of the foreign journals wereincluded. The primary outcome was the number of citations through the end of2011. Subgroup analysis included language. The secondary outcome includedlikelihood of citation (cited at least once versus no citation), and presence orabsence of CONSORT statement indicators. Results: The citation counts were statistically significantly higher (P < 0.001) in theForeign Group (10.50) compared with the Brazilian Group (0.45). The likelihoodcitationwas statistically significantly higher (P < 0.001) in the Foreign Group (20/20-100%) compared with the Brazilian Group (8/20-40%). The subgroup analysis ofthe language influence in Brazilian articles showed that the citation counts werestatistically significantly higher in the papers published in English (P <0.04). Scaling of shoot and root respiration ofwoody and herbaceous plants

Published by the Royal Society under the terms of a Creative Commons Attribution License By .Woody and herbaceous plants are the main components of global terrestrial ecosystems, and their growth, adaptation and survival depend largely on the metabolism of shoots and roots. Therefore, understanding size-scaling of metabolic rates in woody and herbaceous plants, and in shoots and roots, is a fundamental issue in ecology. However, few empirical studies have examined metabolic scaling exponents across a wide range of plant sizes. Using whole-plant chamber systems, we measured respiration rates of entire root systems and shoots of 96 woody species (n = 1243) and 33 herbaceous species (n = 463) from various terrestrial biomes, with plant masses spanning nine orders of magnitude. Scaling exponents for relationships between respiration rates and fresh mass were greater in shoots than in roots, and both were greater in herbaceous plants than in woody plants. Furthermore, scaling of whole-plant respiration, including various species, converged separately for woody and herbaceous plants. These findings suggest some general physico-chemical constraints on energy use by shoots and roots of individual plants in various terrestrial biomes, including forests and grasslands. You will be contacted as soon as possible. Abstract Adrenal corticosteroid hormones act via mineralocorticoid (MR) and glucocorticoid receptors (GR) in the brain, influencing learning and memory. MRs have been implicated in the initial behavioral response in novel situations, which includes behavioral strategies in learning tasks. Different strategies can be used to solve navigational tasks, for example hippocampus-dependent spatial or striatum-dependent stimulus-response strategies. Previous studies suggested that MRs are involved in spatial learning and induce a shift between learning strategies when animals are allowed a choice between both strategies. In the present study, we further explored the role of MRs in spatial and stimulus-response learning in two separate circular holeboard tasks using female mice with forebrain-specific MR deficiency and MR overexpression and their wildtype control littermates. In addition, we studied sex-specific effects using male and female MR-deficient mice. First, we found that MR-deficient compared to control littermates and MR-overexpressing mice display altered exploratory and searching behavior indicative of impaired acquisition of novel information. Second, female (but not male) MR-deficient mice were impaired in the spatial task, while MR-overexpressing female mice showed improved performance in the spatial task. Third, MR-deficient mice were also impaired in the stimulus-response task compared to controls and (in the case of females) MR-overexpressing mice. We conclude that MRs are important for coordinating the processing of information relevant for spatial as well as stimulus-response learning
Produção de biomassa do fungo Agaricus subrufescens por processos fermentativos sólido e submerso para obtenção de polissacarídeos bioativos
Tese (doutorado) - Universidade Federal de Santa Catarina, Centro de Ciências Biológicas, Programa de Pós-graduação em Biotecnologia, Florianópolis, 2010Agaricus subrufescens (=A. brasiliensis) é um fungo comestível amplamente comercializado no Brasil e em outros países, devido as suas propriedades nutricionais e medicinais relacionadas principalmente aos polissacarídeos presentes em sua parede celular, tais como glucanas e glucomananas. Certos processos biotecnológicos têm sido utilizados para aumentar a escala produtiva do micélio e, consequentemente, aumentar a obtenção dos polissacarídeos. Previamente ao desenvolvimento desses processos de produção é importante estabelecer métodos de preservação do fungo in vitro e a recuperação do micélio após estocagem. Para a produção de biomassa foi utilizada a fermentação no estado sólido (FES) em grãos de trigo pré-tratados e a fermentação submersa (FSm) em hidrolisados de resíduos agroindustriais, como farelo de trigo (FT) e resíduo de cervejaria (RC). Polissacarídeos extraídos da frutificação (controle), do micélio em FES e do micélio isolado da FSm foram separados de acordo com suas massas molares por meio de diferentes membranas. Essas frações de polissacarídeos foram testadas na ativação de macrófagos in vitro. Entre os métodos de preservação, melhores resultados foram obtidos quando o meio de cultura foi suplementado com carvão ativo, tanto para a preservação quanto para a recuperação de A. subrufescens, sem alterações significativas na morfologia e genética do fungo durante um período de até 12 meses de estocagem. Para a FES, 21 min de cozimento dos grãos de trigo seguido de 24 min de molho após o cozimento demonstrou ser mais adequado para a produção de polissacarídeos (glucomananas) pelo fungo, inclusive quando o substrato não foi suplementado com cálcio, ou com adição máxima de 0,25 % CaCO3 (pH 6,6). A quantidade inicial de inoculante para a melhor produção de glucomananas (6,89 mg.mL-1) foi aproximadamente 10,3 %, numa temperatura ótima de incubação de 27,2 °C. As maiores biomassas de A. subrufescens para a FSm em hidrolisados de RC e FT foram obtidas na concentração de HCl 0,45 % e 20 min de hidrólise, apresentando 9,65 g.L-1 e 22,1 g.L-1, respectivamente. Porém, melhores resultados de bioconversão foram obtidos em hidrolisados utilizando até 0,3 % de HCl e 30 min de hidrólise, com adição de 2 g.L-1 de carvão ativo durante a hidrólise. Também foram obtidas altas taxas de biomassa de A. subrufescens no cultivo em hidrolisado com pH 5,3 e temperatura de incubação de 30,2 ºC. Na separação com membranas houve aumento da retenção dos polissacarídeos de todos os extratos à medida que o tamanho dos poros da membrana diminuiu. Os polissacarídeos separados pelas membranas de microfiltração (MF) e ultrafiltração (UF1) favoreceram a retenção de polissacarídeos de maior massa molar da frutificação (627 g.mol-1) e do micélio (310 g.mol-1), obtido tanto em FES quanto em FSm. Todos os polissacarídeos extraídos e retentados pelas membranas ativaram macrófagos através da expressão do Fator de Necrose Tumoral-a (TNF-a). Porém, somente os extratos brutos da frutificação e do micélio isolado da FSm, e seus polissacarídeos de maior massa molar retidos pela membrana MF ativaram macrófagos através da expressão de Óxido Nítrico Sintase Induzível (iNOS). Resultados importantes também foram obtidos para os polissacarídeos das frutificações de A. subrufescens, apresentando efeito antigenotóxico e atividade antiherpética quando esses foram sulfatados. Baseado nesses resultados, pesquisas complementares estão sendo desenvolvidas para a obtenção de produtos biotecnológicos com finalidade nutricêutica e/ou farmacêutica.Agaricus subrufescens (=A. brasiliensis) is an edible fungus widely commercialized in Brazil and other countries, mainly due to its nutritional and medicinal properties related principally to the polysaccharides present in the fungus cell wall, such as glucans and glucomannans. Biotechnological processes may in turn be used to scale up the production of mycelium and consequently the polysaccharides. Previous to the development of the production process it is important to establish techniques for the in vitro preservation and recovery of the mycelium after storage. Solid state fermentation (SSF) on pre-treated wheat grains and submerse fermentation (SmF) on hydrolyzed residues from the agro industries, such as wheat bran (FT) and brewery residues (RC) were used for biomass production. Polysaccharides extracted from fruiting bodies (control), mycelium on SSF and isolated mycelium from SmF were separated by molecular weight through different membranes. These polysaccharide fractions were tested on macrophage activity in vitro. Among the preservation techniques, better results were obtained when the media was supplemented with activated charcoal both for preservation and further recovery of A. subrufescens, without significant morphological and genetic changes within the 12 month storage period. For SSF, 21 min of wheat grain cooking followed by a 24 min resting time has shown to be the optimal condition for the production of polysaccharides (glucomannans) by the fungus, either with no supplement added, or when up to 0.25 % CaCO3 (pH 6.6) has been added to the substrate. The initial inoculum amount for the best polysaccharide production levels (6.89 %) was around 10.3 % with an optimal temperature of 27.2 °C. The highest biomass of A. subrufescens for the SmF in hydrolyzed RC and FT using HCL 0.45% and 20 min hydrolysis was 9.65 g.L-1 and 22.1 g.L-1, respectively. On the other hand, better results of bioconversion were obtained in hydrolyzed residues using up to 0.30 % HCl and 30 min hydrolysis, with the addition of 2 g.L-1 of activated charcoal throughout the process. High biomass levels were also obtained when the cultivation was carried out at a pH of 5.3 and with incubation temperature of 30.2 ºC. The results showed an increase in retention of all polysaccharide extracts as the membro ane porosity decreased. Microfiltration (MF) and ultrafiltration (UF1) membranes retained the highest polysaccharides from fruiting body (627 g.mol-1) and mycelium (310 g.mol-1) obtained on both SSF and SFm. All polysaccharide extracts and fractions separated by the membranes showed biological activity for TNF-a cytokines. On the other hand, only extracts obtained from fruiting bodies and isolated mycelium of FSm, and its high molecular weight polysaccharides retained by the MF membrane activated the iNOS. Important results were also obtained for the polysaccharides extracted from the fruiting bodies, showing antigenotoxic and antiherpetic activities after sulphatation. Based on these findings, additional research is currently in progress to purpose biotechnological products that can be used in both the nutraceutical and/or pharmaceutical areas
