    Chemical characterization of the main constituents of the traditional chinese medicine Fang Feng Tong Sheng Wan

    Im Rahmen dieser Diplomarbeit wurden die wichtigsten chemischen Inhaltsstoffe der traditionellen chinesischen Formulierung Fang Feng Tong Sheng Wan (FFTSW), die zur oralen Applikation vorgesehen ist, untersucht. Fang Feng Tong Sheng ist eine der komplexesten traditionellen chinesischen Arzneimittel und besteht aus 17 Komponenten, welche auf 14 Pflanzendrogen und 3 Mineralien aufgeteilt sind. Ziel der Studien war die Identifizierung wichtiger FFTSW Inhaltsstoffe und überdies wurde die Qualitätskontrolle eines handelsüblichen Präparates durchgeführt. Eine geeignete Methode zur Extraktion und Fraktionierung des handelsüblichen FFTSW Präparates wurde entwickelt und optimiert. Aufgrund einer umfassenden Literaturrecherche wurde eine ultraschallbasierte methanolische Extraktion der Pillen durchgeführt. Der erhaltene Extrakt wurde mittels Middle Chromatogram Isolated (MCI) Säulenchromatographie fraktioniert, wobei der Schwerpunkt auf den unpolaren Inhaltsstoffen lag. Die erfolgreiche Extraktion und Auftrennung wurde durch die Bestimmung der Inhaltsstoffe mittels Ultra-High Performance Liquid Chromatography (UHPLC) gekoppelt mit Massenspektrometrie bestätigt. Anhand einer präzisen Fingerprint-Analyse konnten zehn Inhaltsstoffen verschiedener Substanzklassen wie Flavone (Baicalein, Wogonin, Oroxylin A), Anthrachinone (Emodin), Phthalide (Ligustilide, Senkyunolide A) und Sesquiterpenlactone (Atractylenolide I) identifiziert werden. Strukturisomere mit ähnlichem Molekulargewicht konnten mittels vergleichender Analyse von Referenzsubstanzen und anhand charakteristischer Fragmentierungsmuster identifiziert werden. Abschließend wurde eine quantitative Qualitätskontrolle des Biomarkers Baicalin mittels High Performance Liquid Chromatography (HPLC) mit Photodiodenarray Detektion gemäß dem Chinesischen Arzneibuch durchgeführt. Es konnte belegt werden, dass der Baicalingehalt des handelsüblichen Präparates mit 6.485mg den Anforderungen entspricht.This thesis focuses on the main chemical constituents of the ancient Chinese formula Fang Feng Tong Sheng Wan (FFTSW), for oral application in the form of pills (Wan). Fang Feng Tong Sheng is one of the most complex traditional Chinese medicines and consists of 17 components, including 14 herbs and 3 mineral salts. The studies were designed to detect and identify active key compounds of Fang Feng Tong Sheng Wan and furthermore to prove quality standards of a commercial FFTSW sample. First, an adequate method for extraction and fractionation of the commercial FFTSW sample was established and optimized. Based on literature, a methanol-based extraction combined with ultrasonication was applied. The obtained extract was fractionated by middle chromatogram isolated column chromatography (MCI), with a focus on non-polar constituents. In order to prove the successful extraction and separation, the process was validated by determining the main chemical constituents by Ultra-High Performance Liquid Chromatography (UHPLC) with mass spectrometry analysis. Precise fingerprint analysis of FFTSW resulted in clear identification of ten active key constituents of different substance classes such as flavones (baicalein, wogonin, oroxylin A), anthraquinones (emodin), phthalides (ligustilide, senkyunolide A) and sesquiterpene lactone (atractylenolide I).     Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Fang Feng Tong Sheng Wan ist ein traditionelles, komplexes, pflanzliches Arzneimittel, bestehend aus 14 Pflanzen und drei Mineralien. Es wird seit dem 12. Jahrhundert bei Beschwerden wie Erkältung mit Fieber und Frösteln, Asthma, Übelkeit, Kopfschmerzen und Obstipation verwendet. Aufgrund ihrer komplexen Zusammensetzung müssen chinesische Arzneimittel in ihrer Gesamtwirkung charakterisiert und auf Identität, Qualität und Reinheit geprüft werden. Im Rahmen dieser Diplomarbeit wurde zunächst eine Methode zur Extraktion und chromatographischer Auftrennung von FFTSW Tabletten mittels verschiedener Lösungsmittelgemische optimiert. Dafür wurden nicht überzogene FFTSW Tabletten mittels Ultraschall-Extraktion und einer MCI Gel Säulenchromatographie in verschiedenen Methanol-Wasser-Mischungen extrahiert und fraktioniert. Mittels Hochleistungsflüssigkeits-Chromatographie gekoppelt mit Massenspektrometrie (UHPLC-MS) wurde festgestellt, dass mit 30% und 60% methanol-haltigem Lösungsmittel die selektivste und empfindlichste Trennung der Inhaltsstoffe stattfindet. Es konnten elf Inhaltsstoffe aus den Stoffklassen der Flavonoidglykoside (Liquiritin, Luteolosid, Baicalin, Oroxindin), Flavonoidaglykone (Liquiritigenin, Luteolin, Wogonin), Phenolsäuren (Ferulasäure, Gallussäure), Alkaloide (Ephedrin) und Monoterpenglykoside (Paeoniflorin) im Vergleich mit Referenzsubstanzen detektiert und bestimmt werden. Strukturisomere wurden aufgrund ihres unterschiedlichen, charakteristischen Fragmentierungsmusters im Massenspektrum identifiziert.Außerdem wurde der aktive Inhaltsstoff Baicalin mit Hochleistungsflüssigkeits-Chromatographie gekoppelt mit Diodenarray-Detektor (HPLC-DAD) quantifiziert, der dem durch das Chinesische Arzneibuch gefordertem Mindestgehalt von 6.1 mg entsprach.Fang Feng Tong Sheng Wan is an ancient multi-component herbal patent prescription in Traditional Chinese Medicine composed of 14 herbs and three minerals. It has been used since the 12th century to treat influenza, asthma, obesity, nausea, vomiting and headache. Due to the synergistic actions of multi-component and multi-target formulas, herbal TCM prescriptions should be evaluated as a whole, therefore, a multi-component approach is recommended.The described research in this diploma thesis focuses on the extraction and fractionation of Fang Feng Tong Sheng Wan pills via ultrasound assisted extraction (UAE) and middle chromatogram isolated (MCI) gel column chromatography with different methanol/water mixtures. After the appropriate conditions for extraction and separation were determined, it was possible to unambiguously detect and identify eleven main bioactive compounds belonging to flavonoid glycosides (liquiritin, luteoloside, baicalin, oroxindin), flavonoid aglycones (liquiritigenin, luteolin, wogonin), organic acids (gallic acid, phenolic acid), alkaloids (ephedrine) and monoterpene glycosides (paeoniflorin) by using ultrahigh-performance liquid chromatographymass spectrometry (UHPLC-MS).     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods
