    Implication of CDK4 kinase in adipocyte biology

    CDK4 est une sérine/thréonine kinase qui est largement décrite pour son implication dans le contrôle du cycle cellulaire. Notre laboratoire et d'autres ont montré qu'elle jouait également un rôle majeur dans le contrôle de l'homéostasie du glucose (croissance des cellules β du pancréas et sécrétion d'insuline) et des lipides (adipogenèse). Nous avons montré au cours de cette thèse, par le biais de deux modèles de souris, invalidés pour CDK4 (Cdk4-/- ;cre/cre) ou exprimant un mutant hyperactif de la kinase (Cdk4R24C/R24C), qu'elle est un médiateur important de la voie de l'insuline et régule la lipogenèse et la lipolyse. Les souris Cdk4-/- ;cre/cre ont une diminution significative de la taille des adipocytes et du poids du WAT ou l'inverse est observé sur les souris Cdk4R24C/R24C. CDK4 est activée par l'insuline et va ainsi promouvoir le transport de glucose, la synthèse des lipides de novo et réprimer la lipolyse dans les adipocytes. De plus, nous avons démontré que dans l'adipocyte, cellule non proliférative, CDK4 et son partenaire la cycline D3 sont préférentiellement localisés dans le cytoplasme suggérant un rôle indépendant de leurs fonctions nucléaires. Nous avons identifié deux nouveaux substrats de CDK4 : IRS1 et IRS2. CDK4 phosphoryle IRS1 et IRS2 activant un rétrocontrôle positif permettant le maintien de l'action de l'insuline sur les adipocytes. Nos résultats prouvent un nouveau rôle de CDK4 sur la signalisation de l'insuline et sa fonction dans l'adipocyte. Par conséquent, la modulation de son activité pourrait avoir des conséquences majeures sur le mécanisme de résistance à l'insuline, une complication fréquente dans le développement de pathologies comme le diabète et l'obésité.CDK4 is a serine/threonine kinase mainly known by its involvement in the control of cell cycle progression. Our laboratory and other laboratories have previously shown a major role for CDK4 in the control of glucose homeostasis (pancreatic β-cell growth) and lipid homeostasis (adipogenesis). In this thesis, we showed that CDK4 is an insulin effector that controls lipogenesis and lipolysis in mature adipocytes. We used Cdk4-/- ;cre/cre mice and Cdk4R24C/R24C mice, carrying a hyperactive mutant Cdk4 allele, for this study. Cdk4-/ - ;cre/cre mice have a manifest adipose tissue phenotype with a significant decrease in body weight and WAT mass. On the other hand, Cdk4R24C/R24C mice show increased body weight and increased adiposity. Furthermore, we demonstrate that CDK4 is activated by insulin to promote glucose transport, lipogenesis and repress lipolysis in adipocytes. Interestingly, we showed that in mature quiescent adipocytes CDK4 and its partner, Cyclin D3, are preferentially localized in the cytoplasm, suggesting a role independent from their nuclear functions. We identified two novel substrates of CDK4: IRS1 and IRS2. CDK4 phosphorylates both IRS1 and IRS2 in order to sustain insulin signaling in adipocytes via a positive feed-back loop. To sum up, our results identify a new function of CDK4 on insulin signaling in adipocyte metabolism. Thus, the modulation of its activity could have consequences on insulin resistance, a common complication of obesity and diabetes

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Les régulateurs du cycle cellulaire et la fonction métabolique

    Nous avons mis en évidence l'implication de facteurs du cycle cellulaire, la cycline D3 et E2F1, dans deux processus métaboliques, l'adipogenèse et l'homéostasie du glucose. Nous avons montré que la cycline D3 favorise la différenciation adipocytaire. En effet, nous avons observé une augmentation de l'expression de la cycline D3 au cours de ce processus non prolifératif et identifié une interaction directe de cette protéine avec un facteur de transcription adipogénique majeur, le récepteur nucléaire PPARg, augmentant son activité transcriptionnelle. Comme nous l'avons mis en évidence chez les souris cycline D3 -/-, cette activation a des implications majeures dans l'obésité. Nous avons aussi montré que des souris invalidées pour le gène E2F1 ont une diminution de la taille du pancréas et une diminution de la sécrétion d'insuline. Malgré cela, ces souris sont hypersensibles à l'insuline et ne deviennent pas diabétiques.     Contrôle du métabolisme et de la prolifération par les microARN et les acteurs du cycle cellulaire

    Le contôrle métabolique implique un dialogue entre régulateurs du cycle cellulaire et facteurs métaboliques. Des stimuli externes, comme la nutrition, peuvent être traduits en signal prolifératif mais également en une réponse métabolique. Précédemment, nous avons observé que les régulateurs, E2F1, cdk4 et Rb jouent un rôle clé dans le contrôle de l'adipogenèse et la fonction pancréatique. Nous avons montré que le facteur E2F1 participait activement à la régulation de la biogenèse et la fonction mitochondriales en réprimant l'expression de PGC-1 alpha, re gulateur de l'homéostasie énergétique. De façon intéressante, les souris cdk4-/- semblent présenter une expression de gènes mitochondriaux et des activités mitochondriales, inversées à celles des souris E2fl-/-. Enfin, CDK4 semble également jouer un rôle dans le métabolisme lipidique. Les microARNs sont des petits ARN endogènes non codants qui régulent négativement l'expression des gènes et sont impliqués dans de nombreux processus cellulaires. Nous avons montré que l'expression du microARN 143 diminue durant la progression du cancer de la prostate. La protéine ERK5, cible de mir-143, a des effets prolifératifs et son expression est aberrante dans le cancer de la prostate. mir-143 pourrait être directement impliqué dans le carcinogenèse, via ERK5. Cela suggère que mir-143 joue un rôle dans le métabolisme énergétique. Nos résultats indiquent donc que les changements métaboliques et prolifératifs observés dans le développement du cancer soient étroitement liés, mais aussi régulés par les facteurs du cycle cellulaire et les microARNParticipation of cell cycle regulators in metabolic processes represents a new link between cell proliferation and cell metabolism. External stimuli, like cold and fasting conditions, could induce a proliferative response, but also a metabolic response. Previously, we have shown that the cell cycle regulators, CDK4, pRB and E2F1 play a crucial role during adipogenesis and pancreatic function. Here we show that E2F1 could actively participate in regulation of mitochondrial biogenesis and function. E2FA represses the expression of Pgc-1 alpha, which is master regulator of energy homeostasy in brown adipose tissue and muscle. Interestingly mitochondrial genes in cdk4-/- mice seems to be inversely correlated with gene expression in e2fl-/- mice. Finally, CDK4 coould [i.e.could] play a role in lipid metabolism. MicroRNAs are endogenous, non-coding and small RNAs, which negatively regulate gene expression and are implicated in many cellular processes. We showed that mir-143 expression is inversely correlated with advanced stages of prostate cancer. ERK5, a target of mir-143, is known to promote the proliferation and its expression is aberrant in prostate cancer. Mir-143 could be directly implicated in carcinogenesis througe [i.e. through] ERK5. This suggest taht [i.e. that] mir-143 could be a tumor suppressor in prostate cancer.     Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     CXCL5 drives obesity to diabetes, and further.: CXCL5 and metabolism

    International audienceWe have recently shown that the CXCL5 chemokine is secreted by adipose tissue in the obese state. We demonstrated that adipose tissue-derived CXCL5 mediates insulin resistance in muscle. We speculate in this paper that CXCL5 could also mediate other obesity, and diabetes-derived pathologies, such as cardiovascular disease, retinopathy, or inflammatory bowel disease. In this scenario CXCL5 targeted therapy would prevent not only the development of type II diabetes in obese subjects, but also several other obesity-related co morbidities. Finally we propose to analyze the CXCL5 gene to find particular polymorphisms that could predict the development of type II diabetes in obese subjects

    Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods
